The fall armyworm(FAW),Spodoptera frugiperda,is a major pest native to the Americas that has recently invaded the Old *** mutations in the target-site proteins acetylcholinesterase-1(ace-1),voltage-gated sodium channel(VGSC)and ryanodine receptor(RyR)have been identified in *** as major resistance mechanisms to organophosphate,pyrethroid and diamide insecticides *** in the adenosine triphosphate-binding cassette transporter C2 gene(ABCC2)have also been identified to confer resistance to Cry IF *** this study,we applied a whole-genome sequencing(WGS)approach to identify point mutations in the target-site genes in 150 FAW individuals collected from China,Malawi,Uganda and *** approach revealed three amino acid substitutions(A201S,G227A and F290V)of *** ace-1,which are known to be associated with organophosphate *** Brazilian population had all three ace-1 point mutations and the 227A allele(mean frequency=0.54)was the most *** from China,Malawi and Uganda harbored two of the three ace-1 point mutations(A201S and F290V)with the 290V allele(0.47-0.58)as the dominant *** mutations in VGSC(T929I,L932F and L1014F)and RyR(I4790M and G4946E)were not detected in any of the 150 individuals.A novel 12-bp insertion mutation in exon 15 of the ABCC2 gene was identified in some of the Brazilian individuals but absent in the invasive *** results not only demonstrate robustness of the WGS-based genomic approach for detection of resistance mutations,but also provide insights for improvement of resistance management tactics in ***.

A detailed analysis of a sea breeze front(SBF)that penetrated inland in the Beijing–Tianjin–Hebei urban agglomeration of China was *** focused on the boundary layer structure,turbulence intensity,and fluxes before and after the SBF passed through two meteorological towers in the urban areas of Tianjin and Beijing,*** changes in temperature,humidity,winds,CO_(2),and aerosol concentrations were observed as the SBF *** in these changes at the two towers mainly resulted from their distances from the ocean,boundary layer conditions,and background *** the SBF approached,a strong updraft appeared in the boundary layer,carrying near-surface aerosols aloft and forming the SBF *** was followed by a broad downdraft,which destroyed the near-surface inversion layer and temporarily increased the surface air temperature at *** feeder flow after the thermodynamic front was characterized by low-level jets horizontally,and downdrafts and occasional updrafts *** increased significantly during the SBF’s passage,causing an increase in the standard deviation of wind components in *** increase in turbulence was more pronounced in a stable boundary layer compared to that in a convective boundary *** passage of the SBF generated more mechanical turbulences,as indicated by increased friction velocity and turbulent kinetic energy(TKE).The shear term in the TKE budget equation increased more significantly than the buoyancy *** atmosphere shifted to a forced convective state after the SBF’s passage,with near isotropic turbulences and uniform mixing and diffusion of *** heat fluxes(latent heat and CO_(2)fluxes)showed positive(negative)peaks after the SBF’s passage,primarily caused by horizontal and vertical transport of heat(water vapor and CO_(2))during its *** study enhances understanding of boundary layer changes,turbulences,and fluxes during the passage of SBFs over u

【目的】探讨内源乙烯与1-MCP的相互作用对贮藏过程中番茄成熟和细胞壁代谢的影响。【方法】将破色期番茄("佛罗里达47")果实分为4组,其中2组经低压低氧(8kPa,1.7kPaO2)处理6h后,分别立即使用50μg/L液态1-MCP(HH+1-MCP)或去离子水(HH)浸泡1min;另外2组未经HH处理的番茄果实,也分别使用50μg/L1-MCP(1-MCP)或去离子水(对照)浸泡1min,测定处理后果实在贮藏(20℃)过程中成熟相关生理指标及细胞壁组分的变化。【结果】番茄果实经HH处理后,内源乙烯含量(IEC)急剧降低至对照水平的75.6%,且对1-MCP的反应敏感性有所提高。HH+1-MCP处理显著抑制了果实硬度的下降,其硬度值在贮藏第4～16天中,平均分别较对照和1-MCP处理高29.8%和15.6%;HH+1-MCP处理果实的呼吸和乙烯峰出现时间均较对照推迟4d,较1-MCP处理推迟2d,但对峰值影响未达到显著水平;与1-MCP处理相比,HH+1-MCP处理对多聚半乳糖醛酸酶(PG)、果胶甲脂酶(PME)、α-半乳糖苷酶(α-Gal)、β-半乳糖苷酶(β-Gal)等细胞壁降解酶活性的变化影响较大,对PG的影响尤为明显;HH+1-MCP处理还显著地抑制了番茄果实贮藏期水溶性和CDTA溶性糖醛酸(UA)含量的增加,其水溶性UA含量在贮藏的第6～12天平均值分别较对照和1-MCP处理低37.3%和21.9%,其CDTA溶性UA含量在贮藏的第6～12天平均值分别较1-MCP处理果实低31.4%和20.5%;凝胶渗透色谱分析结果显示,1-MCP和HH+1-MCP处理延缓了番茄果实多聚糖醛酸分子的降解过程,且后者效果更为明显。此外,本试验HH处理各项测定指标与对照无显著差异。【结论】内源乙烯含量是调节1-MCP反应能力的重要因素,解释了不同跃变型果实在成熟起始后对1-MCP反应敏感性差异的原因。

目的探讨不同含量碳水化合物饮食对大鼠心脏和脑组织18F-FDG摄取的影响。方法以标准饮食饲养2周后,保持饮食总能量不变,将15只Wistar大鼠随机分为3组:低碳水化合物组(LC)、中碳水化合物组(IC)和高碳水化合物组(HC);每组大鼠每种饮食饲养4周后,对动物行18F-FDGPET全身显像,于第2天尾静脉取血测定酮体、葡萄糖、胰岛素和胰高血糖素的含量。图像经计算机处理,获取心脏和脑的最大标准摄取值(SUVmax)。结果心脏SUVmax分别为:LC组3.27±0.43,IC组10.92±0.89,HC组10.95±0.81;脑SUVmax:LC组4.78±0.19,IC组4.04±0.15,HC组3.77±0.24。LC组与IC和HC组之间的差异均有统计学意义(P<0.001),IC和HC组间差异无统计学意义。LC组酮体为(726.10±65.95)μmol/L,约为IC和HC组的6～7倍;血清酮体水平与心脏SUVmax呈负相关(r=-0.764,P<0.001),与脑SUVmax呈正相关(r=0.569,P<0.001);各组间血清胰岛素、血糖和胰高血糖素的差异均无统计学意义。结论长期极低碳水化合物饮食使大鼠心脏对FDG的摄取明显降低,而脑组织对葡萄糖的摄取略有增加。

The pyrimidine analog, 5-aza-2’-deoxycytidine (5-aza-dC) is a DNA methyltransferase inhibitor that triggers DNA demethylation leading to the reactivation of epigenetically silenced tumor suppressor genes. To understand the shift in gene expression which mediates the beneficial 5-aza-dC effects in leukemia, we have treated human myeloid derived leukemic cells with 5-aza-dC. Target genes were identified first in MV4-11 cells using a genome-wide gene expression profiling assay to detect differences in treated and untreated cells. From this analysis six genes were identified (HOXA4, HOXD4, HOXA8, HOXD12, CD9 and RGS2) as being significantly different expressed after treatment. To validate microarray data, we performed quantitative PCR on these genes from multiple leukemic cells. The results suggest that these genes are epigenetically regulated indicating that dysregulation of HOXA4, HOXD4, HOXA8, HOXD12, CD9 and RGS2 expression may play an important role in establishing the malignant phenotype in AML.

目的 :利用多通道记录技术在清醒大鼠前扣带回 (ACC)记录神经元群的痛反应模式 ,以证实ACC在痛觉情绪编码中的作用。方法 :在 5只成年雄性SD大鼠的双侧ACC脑区植入微电极阵列 ,在大鼠术后清醒状态下给予尾部及四肢足底伤害性辐射热刺激 ,神经元的放电信号经由电缆和连接器传送至多通道记录系统。结果 :伤害性辐射热刺激在大鼠ACC引起以兴奋为主的反应 ,该反应持续时间较长 ,可能与痛情绪有关 ;刺激开始附近ACC神经元有与痛刺激相关的预期性反应 ,可能与准备逃避的动机有关 ;同侧和对侧肢体刺激引起的ACC神经元的反应差别不显著 ,表明神经元感受野大 ,不适合精确定位。结论
