Mouse models of a.tibiotic-induced a.la.ion of the intestina. microbiome ha.e been used to study the microbiome in hea.th a.d disea.e. The feca. microbiomes of mice trea.ed with broa.-spectrum a.tibiotics while being fed different la.ora.ory chows were a.a.yzed by Gra. sta.n, qua.tita.ive flow cytometry, ba.teria. cell culture, next genera.ion sequencing of the V3 regions of the 16S ribosoma. RNa.(rRNa. gene, microscopy, a.d sequence a.a.ysis of the tuf gene. Noncultiva.a.le gra.-positive cocci a.d cultiva.a.le yea.t were the microorga.isms most rea.ily detected in feces of a.tibiotic-trea.ed mice fed a.defined diet tha. utilizes ca.ein a. a.protein source, ma.todextrin 10 a.d sucrose a. sources of ca.bohydra.es, a.d la.d a. the ma.or source of fa.. High-throughput sequencing of the va.ia.le regions of the 16S rRNa.gene a.d tuf gene sequencing identified the ma.or ba.teria. phylotype a. La.tococcus. The mouse chow wa. hea.ily la.en with noncultiva.a.le La.tococcus, which domina.ed the intestina. flora.a.ter consumption. The microbiome of a.tibiotic-trea.ed mice fed a.gra.n-ba.ed diet (ma.nly whea., corn a.d a.fa.fa. consisted predomina.tly of a.member of the Enteroba.teria.ea. identified a. Escherichia.coli, a.d yea.t wa. not detected by culture or Gra. sta.n. a.pea.a.ce of intestina. yea.t by culture a.d Gra. sta.n wa. dependent on the specific chow, a.though yea.t wa. not detected by culture or Gra. sta.n in the chow. We conclude tha. ba.teria.found in food sources ca. influence qua.ita.ive a.d qua.tita.ive a.sessments of the feca. microbiome, a. lea.t in the context of a.tibiotic thera.y, a.d potentia.ly confound molecula. studies tha. a.sess the effects of diet on the intestina. ecology. Not surprisingly, different food sources ca. influence the microbiome, pa.ticula.ly in the context of a.tibiotic-media.ed a.la.ion of the intestina. microbiome. Whether a.d how the food-derived dea. ba.teria.a.ter intestina. physiology needs to be determined.
