Participation of epididymal cysteine-rich secretory proteins in sperm-egg fusion and their potential use for male fertility regulation

Rat protein DE is an androgen-dependent cysteine-rich secretory protein （CRISP） synthesized by proximal epididymal regions. DE, also known as CRISP-1, is localized on the equatorial segment of acrosome-reacted spermatozoa and participates in gamete fusion through binding to egg complementary sites. Immunization of rats with DE inhibits fertility and sperm fusion ability, suggesting that DE represents a good epididymal contraceptive target. Recombinant DE fragments and synthetic peptides revealed that DE binds to the egg via a 12-amino acid region of an evolutionarily conserved motif, Signature 2 （S2）. The ability of other CRISP to bind to the rat egg was correlated with their S2 amino acid sequences. Although testicular protein Tpx- 1 （CRISP-2） was capable of binding to rodent eggs, human epididymal AEG-related protein （ARP） and helothermine （from lizard saliva） were not. The S2 region presented only two substitutions in Tpx-1 and four in ARP and helothermine, compared with the DE S2, suggesting that this amino acid sequence was relevant for egg interaction. Studies with Tpx- 1 and anti-Tpx- 1 revealed the participation of this protein in gamete fusion through binding to complementary sites in the egg. In competition studies, DE reduced binding of Tpx- 1 dose-dependently, indicating that both CRISP share the egg complementary sites. That anti-DE and anti-Tpx-1 inhibit sperm-egg fusion while recognizing only the corresponding proteins, suggests functional cooperation between these homologous CRISP to ensure fertilization success. These results increase our understanding of the molecular mechanisms of gamete fusion and contribute to the development of new and safer fertility regulating methods. （Asian j Androl 2007 july; 9： 528-532）

Light Meter for Measuring Photosynthetically Active Radiation

Measurement of photosynthetically active radiation (PAR) incident on photosynthetic organisms is a crucial measurement for understanding how organisms respond to various light conditions, and for calculating electron flow through the photosynthetic machinery. Measurements of PAR are typically performed in the region of the electromagnetic spectrum between 400 - 700 nm, which is the region of radiation that is responsible for promoting photosynthesis. Typically, to ensure that the sensor measures in this range, the implementation of long- and short-pass filters is required. Although this allows the exclusion of radiation outside of the PAR region, such filters can be expensive. Additionally, the implementation of autonomous PAR measurements requires costly commercial instruments. Here, a straight-forward, inexpensive apparatus has been designed and constructed using a sensor that can distinguish between red, green, blue and white light. The constructed apparatus was able to perform comparably to a commercial PAR sensor. Furthermore, the implementation of the device to measure PAR intensity over a three-day period shows how the apparatus can be implemented for use as a constant light monitor.

Gastrointestinal function testing model using a new laryngopharyngeal pH probe(Restech)in patients after Ivor-Lewis esophagectomy

Software improvement for evaluation of laryngopharyngeal pH testing(Restech)——a comparison between Data View 3 and 4

Carbohydrate Yield and Biomethane Potential from Enzymatically Hydrolysed <i>Saccharina latissima</i>and Its Industrial Potential

The demand for fuel for utilisation of machinery and transport has culminated in large amounts of fossil fuel usage in the last century. The environmental dangers attached with the usage of fossil fuels have created a large demand for alternative sources of fuels. There is an array of polysaccharides contained within macroalgae, such as mannitol, cellulose and laminarin. These polysaccharides have potential for production of alternative biofuels;however, they are not easily accessible for biological digestion. By pretreatment of macroalgae with enzymes, these polysaccharides may be easier to access by microbes, allowing effective utilization in anaerobic digestion. Saccharina latissima, available in abundance on the Norwegian coast line, is a brown macroalgae with a high level of carbohydrates. This study assesses the ability for utilisation of enzymatically pre-treated Saccharina latissima for production of biogas through anaerobic digestion. The harvested Saccharina latissima was analysed to contain 30.11 ± 2.30 g of reducing sugars per 100 g of dry sample upon enzymatic hydrolysis. This was able to yield 459 ± 30 mL per gVS of biogas through anaerobic digestion, with a methane content of 56%. This suggests a biomethane potential of 1760 m3 per ha of productive sea floor growing Saccharina latissima. An evaluation of this process has been performed to demonstrate the industrial potential of Saccharina latissima in biogas production.

The physics of compressible turbulence in high energy density(HED) plasmas is an unchartered experimental *** of compressible and radiative flows relevant for astrophysics rely mainly on subscale parameters. Therefore,we plan to perform turbulent hydrodynamics experiments in HED plasmas(TurboHEDP) in order to improve our understanding of such important phenomena for interest in both communities: laser plasma physics and astrophysics. We will focus on the physics of supernovae remnants which are complex structures subject to fluid instabilities such as the Rayleigh–Taylor and Kelvin–Helmholtz instabilities. The advent of megajoule laser facilities, like the National Ignition Facility and the Laser Megajoule, creates novel opportunities in laboratory astrophysics, as it provides unique platforms to study turbulent mixing flows in HED plasmas. Indeed, the physics requires accelerating targets over larger distances and longer time periods than previously achieved. In a preparatory phase, scaling from experiments at lower laser energies is used to guarantee the performance of future Mj experiments. This subscale experiments allow us to develop experimental skills and numerical tools in this new field of research, and are stepping stones to achieve our objectives on larger laser facilities. We review first in this paper recent advances in high energy density experiments devoted to laboratory astrophysics. Then we describe the necessary steps forward to commission an experimental platform devoted to turbulent hydrodynamics on a megajoule laser facility. Recent novel experimental results acquired on LULI2000, as well as supporting radiative hydrodynamics simulations, are presented. Together with the development of LiF detectors as transformative X-ray diagnostics, these preliminary results are promising on the way to achieve micrometric spatial resolution in turbulent HED physics experiments in the near future.

Hazard Assessment of Alternatives to Dicofol

Dicofol was listed by International POPs Elimination Network (IPEN) as requiring immediate and urgent considera-tion and thus was considered as a new candidate by Persistent Organic Pollutant Review Committee (POPRC) as a possible persistent organic pollutant (POP). Dicofol is structurally similar to DDT. It is persistent in food and water, highly toxic to aquatic life and causes egg-shell thinning in some bird species. High concern, due to the lack of dicofol measurements in the Arctic, proving long range transport and bioaccumulation in wild life species, supports further impact assessment of this product. Under Stockholm Convention, substances identified as POPs are regulated with the objective to protect the environment and the human health. According to this objective, the search of environmental and healthy alternatives is helpful. This paper discusses the use of three groups of chemicals currently applied as alternatives to dicofol. An exhaustive review of the synthesis of dicofol, starting from DDT, and compared to possible substitutes is presented: 1) active principle with fluoralkenyl are proposed as an environmental and healthy alternative to dicofol, 2) inhibitor agents of mitochondrial electron transport as chlorfenapyr, hydramethylnon and pyridaben and 3) pesticides commonly applied in agricultural practices as oxythioquinox, fenbutatin-oxide and formetanate hydrochloride.

Downregulation of Hes1 expression in experimental biliary atresia and its effects on bile duct structure

AIM To analyze the expression and function of the notchsignaling target gene Hes1 in a rhesus rotavirusinduced mouse biliary atresia model. METHODS The morphologies of biliary epithelial cells in biliary atresia patients and in a mouse model were examined by immunohistochemical staining. Then, the differential expression of Notch signaling pathway-related molecules was investigated. Further, the effects of the si RNAmediated inhibition of Hes1 expression were examined using a biliary epithelial cell 3 D culture *** Both immature(Ep CAM+) and mature(CK19+) biliary epithelial cells were detected in the livers of biliary atresia patients without a ductile structure and in the mouse model with a distorted bile duct structure. The hepatic expression of transcripts for most Notch signaling molecules were significantly reduced on day 7 but recovered to normal levels by day 14, except for the target molecule Hes1, which still exhibited lower m RNA and protein levels. Expression of the Hes1 transcriptional co-regulator, RBP-jκ was also reduced. A 3 D gel culture system promoted the maturation of immature biliary epithelial cells, with increased expression of CK19+ cells and the formation of a duct-like structure. The administration of Hes1 si RNA blocked this process. As a result, the cells remained in an immature state, and no duct-like structure was *** Our data indicated that Hes1 might contribute to the maturation and the cellular structure organization of biliary epithelial cells, which provides new insight into understanding the pathology of biliary atresia.
