The auxin signaling molecule controls a variety of growth and developmental processes in land plants. Auxin regulates gene expression through a nuclear auxin signaling pathway (NAP) consisting of the ubiquitin ligase auxin receptor TIR1/AFB, its Aux/IAA degradation substrate, and DNA-binding ARF transcription factors. Although extensive qualitative understanding of the pathway and its interactions has been obtained, mostly by studying the flowering plant Arabidopsis thaliana, it remains unknown how these translate to quantitative system behavior in vivo, a problem that is confounded by the large NAP gene families in most species. Here, we used the minimal NAP of the liverwort Marchantia polymorpha to quantitatively map NAP protein accumulation and dynamics in vivo through the use of knockin fluorescent fusion proteins. Beyond revealing the dynamic native accumulation profile of the entire NAP protein network, we discovered that the two central ARFs, MpARF1 and MpARF2, are proteasomally degraded. This auxin-independent degradation tunes ARF protein stoichiometry to favor gene activation, thereby reprogramming auxin response during the developmental progression. Thus, quantitative analysis of the entire NAP has enabled us to identify ARF degradation and the stoichiometries of activator and repressor ARFs as a potential mechanism for controlling gemma germination.

Fundamentals about onset and progressive disease character of type 2 diabetes mellitus

As the starting point of a new life cycle,activation of the first division of the zygote is a critical event in both plants and animals. Due to poor accessibility of plant zygotes,it has remained poorly understood how this process is regulated. Through genetic analyses in Arabidopsis we identified a zygote-arrest 1(zyg1) mutant that exhibited zygote-lethality and over-accumulation of cyclin-GUS phenotypes. Map-based cloning showed that ZYG1 encodes the anaphase promoting complex/cyclosome(APC/C) subunit 11(APC11). Live-cell imaging showed that APC11 is expressed in both egg and sperm cells,and that expression continued in zygotes and during early embryogenesis. Using an APC11-GFP fusion construct,we showed that APC11-GFP expression persisted throughout the mitotic cell cycle,and localized to cell plates during cytokinesis. Expression of non-degradable cyclin under the control of a zygote-specific DD45 promoter led to a zyg1-like zygote-lethal phenotype. Biochemical studies showed that APC11 is able to ubiquitinate cyclin,thus promoting cyclin degradation. These results together suggest that APC11-mediated cyclin degradation in Arabidopsis is critical for the first division of the zygote.

Interactions between inhomogeneous magnetic fields and a superconductor lie at the heart of several high‐temperature superconducting(HTS)*** example is the situation where the magnetic field from a small permanent magnet(PM)is applied to a coated‐conductor HTS tape,such that flux penetrates the *** PM field is highly spatially inhomogeneous,such that there is significant variation in the applied field magnitude and direction across the *** leads to a varying local critical current density,J_(c)(B,θ),across the tape,and consequently different‘shielding’(magnetic flux penetration)behaviour than is the case for a uniform applied ***,we report results from measurements and numerical simulations of the penetrating magnetic field within an HTS coated‐conductor tape which occur as a PM dipole approaches from a *** simulations were performed using a finite‐element model based on the H‐formulation,and which incorporated experimentally‐measured anisotropic J_(c)(B,θ)Þproperties from a coated‐conductor *** showed good agreement with experimental *** effects of varying key modelling assumptions and parameters were then studied,including changing the widths of the PM and HTS tape and the magnitude of J_(c)(B,θ).The inhomogeneity of the PM field leads to a characteristic gull‐wing distribution of magnetic flux across the superconductor at elevated applied *** approach of the PM to the tape suppresses Jc in the centre of the tape,and this results in the observation of a characteristic maxima for the total shielding currents circulating in the tape.A figure of merit is introduced which uses this effect to provide a threshold definition for‘full flux penetration’of an HTS tape by an inhomogeneous dipole field.
