Green tea extract and Piper retrofractum attenuate deoxycholic acid-induced damage and enhance the tight junction barrier: An analysis in a Caco-2 cell culture model and a DSS coinduced mouse model

This study investigated the effect of green tea extract(GTE)and green tea-Piper retrofractum complex(GPX),on tight junctions(TJs)in a cellular model of deoxycholic acid(DCA)-induced intestinal barrier disruption and a mouse DSS+DCA-induced colitis *** tea and *** were extracted and coadministered to examine the bioavailability and effects of combined catechins and piperine on colonic TJ *** ability of GPX to alleviate TJ damage was determined by comparing the transepithelial electrical resistance(TEER),FITC-dextran flux,and mRNA expression of TJ-related components and target expression pathway in the GTE and GPX *** with the DCA-treated group,the GTE-and GPX-treated groups showed significantly enhanced TEER,FITC-dextran flux,and TJ-related mRNA expression(pdownregulated the mRNA encoding bile acid receptor and decreased ERK1/2 *** with the GTE-treated group,the GPX-treated group showed higher mRNA expression of zonula occludens-1,occludin,claudin-3,and claudin-4;greater restoration of TJs,as determined by immunofluorescence;and better modulation of ERK1/2 *** the in vivo model,GPX-pretreated mice showed an improved intestinal damage index and colon length compared with mice in the other colitis-induced *** extracts recovered the distal colon mRNA expression of zonula occludens-1 and *** results suggest that GTE improves intestinal health by protecting TJs from secondary bile acid damage and that *** may potentiate the bioactivity of green tea catechins.

BACKGROUND Compared to earlier,there has been an increase in the tattoo procedures for cosmetic purposes;and there has also been an increase in the tattoo procedures performed by non-medical *** South Korea,only tattoos performed by a doctor are considered legal;however,there is still some debate over whether tattoo procedures performed by non-healthcare providers should be considered *** SUMMARY A 28-year-old woman visited our hospital with pain in both nipples and heat sensation over the last 4 *** had a history of a nipple tattoo performed by an unlicensed *** bleeding was noted in both areolar areas,and the exudate mixed with pus and orange color ink was *** medication and tulle with foam dressing were performed under the impression of cellulitis and allergic *** 4 wk,nipples remained dark brown in color,resulting in a color mismatch between the nipple and orange-colored *** size of the areola was also found to be distinctly asymmetrical after *** complication may have been caused by the use of illegal ink or unsanitary procedures,or a problem may have occurred in the post-tattoo management *** doctors use approved ink,aseptic procedure and appropriate postoperative care,and appropriate management can be performed in case of complications.

Numerical Visualization of Air Intake Induced by Free Surface Vortex

Free surface vortex control is vital in a pump sump system because the air absorbed by free surface vortex induc- es noise, vibration, and cavitation corrosion on the pumping system. In this study, the change of free surface vor- tex and air absorption in a pump intake has been investigated by the Volume of Fraction （VOF） method with steady multiphase flow model in order to represent the behavior of the free surface vortex exactly. The homoge- neous free surface model is used to apply interactions of air and water. The results show that air intake by the free surface vortex motion can be visualized using the iso-surface of air volume fraction. The vortices make an air column from the free surface to the pump intake. Also, it was found that the flee surface vortex can be controlled by installing curtain walls.

Image-guided conservative management of right colonic diverticulitis

AIM： To study the clinical outcomes of medical therapy in patients with right colonic diverticulitis. METHODS： The records of 189 patients with right colonic diverticulitis which was finally diagnosed by computed tomography, ultrasonography, or operative findings were retrospectively reviewed. RESULTS： Of the 189 patients hospitalized for right colonic diverticulitis, the stages of diverticulitis by a modified Hinchey classification were 26 patients （13.8%） in stage 0, 139 patients （73.5%） in stage I a, 23 patients （12.2%） in stage I b, and 1 patient （0.5%） in stage Ⅲ. Medical therapy was undertaken in 185 of 189 patients （97.9%）. One hundred and eighty three of 185 patients were successfully treated with bowel rest and antibiotics. Two patients in stage I b required a resection or surgical drainage because of an inadequate response to conservative treatment. Recurrent diverticulitis developed in 15 of 183 patients （8.2%） who responded to medical therapy. All 15 patients who suffered a second attack had uncomplicated diverticulitis, and were successfully treated with medical ***： Our results indicate that right colonic diverticulitis is essentially benign and image-guided conservative treatment is primarily required.

Moutan Cortex Radicis inhibits the nigrostriatal damage in a 6-OHDA-induced Parkinson's disease model

The traditionally used oriental herbal medicine Moutan Cortex Radicis [MCR; Paeonia Suffruticosa Andrews(Paeoniaceae)] exerts anti-inflammatory, anti-spasmodic, and analgesic effects. In the present study, we investigated the therapeutic effects of differently fractioned MCR extracts in a 6-hydroxydopamine(OHDA)-induced Parkinson's disease model and neuro-blastoma B65 cells. Ethanol-extracted MCR was fractionated by n-hexane, butanol, and distilled water. Adult Sprague-Dawley rats were treated first with 20 μg of 6-OHDA, followed by three MCR extract fractions(100 or 200 mg·kg^(-1)) for 14 consecutive days. In the behavioral rotation experiment, the MCR extract-treated groups showed significantly decreased number of net turns compared with the 6-OHDA control group. The three fractions also significantly inhibited the reduction in tyrosine hydroxylase-positive cells in the substantia nigra pars compacta following 6-OHDA neurotoxicity. Western blotting analysis revealed significantly reduced tyrosine hydroxylase expression in the substantia nigra pars compacta in the 6-OHDA-treated group, which was significantly inhibited by the n-hexane or distilled water fractions of MCR. B65 cells were exposed to the extract fractions for 24 h prior to addition of 6-OHDA for 30 min; treatment with n-hexane or distilled water fractions of MCR reduced apoptotic cell death induced by 6-OHDA neurotoxicity and inhibited nitric oxide production and neuronal nitric oxide synthase expression. These results showed that n-hexane-and distilled water-fractioned MCR extracts inhibited 6-OHDA-induced neurotoxicity by suppressing nitric oxide production and neuronal nitric oxide synthase activity, suggesting that MCR extracts could serve as a novel candidate treatment for the patients with Parkinson's disease.

Single port laparoscopic right hemicolectomy with D3 dissection for advanced colon cancer

MicroRNA miR-252-5p regulates the Notch signaling pathway by targeting Rab6 in Drosophila wing development

The Notch signaling pathway plays a central role in the development of various ***,dysregulation of microRNAs(miRNAs),which are crucial regulators of gene expression,can disrupt signaling pathways at all stages of *** Notch signaling is involved in wing development in Drosophila,the mechanism underlying miRNA-based regulation of the Notch signaling pathway is ***,we report that loss of Drosophila miR-252 increases the size of adult wings,whereas the overexpression of miR-252 in specific compartments of larval wing discs leads to patterning defects in the adult *** miR-252 overexpression-induced wing phenotypes were caused by aberrant Notch signaling with intracellular accumulation of the full-length Notch receptor during development,which could be due to defects in intracellular Notch trafficking associated with its recycling to the plasma membrane and autophagy-mediated ***,we identified Rab6 as a direct target of miR-252-5p;Rab6 encodes a small Ras-like GTPase that regulates endosomal trafficking *** with this finding,RNAi-mediated downregulation of Rab6 led to similar defects in both wing patterning and Notch ***,co-overexpression of Rab6 completely rescued the wing phenotype associated with miR-252 overexpression,further supporting that Rab6 is a biologically relevant target of miR-252-5p in the context of wing ***,our data indicate that the miR-252-5p-Rab6 regulatory axis is involved in Drosophila wing development by controlling the Notch signaling pathway.

C-terminal domain of hepatitis C virus core protein is essential for secretion

AIM: We have previously demonstrated that hepatitis C virus (HCV) core protein is efficiently released into the culture medium in insect cells. The objective of this study is to characterize the HCV core secretion in insect cells. METHODS: We constructed recombinant baculoviruses expressing various-length of mutant core proteins, expressed these proteins in insect cells, and examined core protein secretion in insect cells. RESULTS: Only wild type core was efficiently released into the culture medium, although the protein expression level of wild type core was lower than those of other mutant core proteins. We found that the shorter form of the core construct expressed the higher level of protein. However, if more than 18 amino acids of the core were truncated at the C-terminus, core proteins were no longer secreted into the culture medium. Membrane flotation data show that the secreted core proteins are associated with the cellular membrane protein, indicating that HCV core is secreted as a membrane complex. CONCLUSION: The C-terminal 18 amino acids of HCV core were crucial for core secretion into the culture media. Since HCV replication occurs on lipid raft membrane structure, these results suggest that HCV may utilize a unique core release mechanism to escape immune surveillance, thereby potentially representing the feature of HCV morphogenesis.

BACKGROUND This report describes and discusses recurrent intramuscular lipoma(IML)of the extensor pollicis brevis(EPB).An IML usually occurs in a large muscle of the limb or *** of IML is *** IMLs,especially those with unclear boundaries,necessitate complete *** cases of IML in the hand have been ***,recurrent IML appearing along the muscle and tendon of EPB on wrist and forearm has not been reported *** SUMMARY In this report,the authors describe clinical and histopathological features of recurrent IML at EPB.A 42-year-old Asian woman presented with a slowgrowing lump in her right forearm and wrist area six months *** patient had a history of surgery for a lipoma of the right forearm one year ago with a scar of 6cm on the right *** resonance imaging confirmed that the lipomatous mass,which had attenuation similar to subcutaneous fat,had invaded the muscle layer of *** and biopsy were performed under general *** histological examination,it was identified as an IML showing mature adipocytes and skeletal muscle ***,surgery was terminated without further *** recurrence occurred during a follow-up of five years after *** Recurrent IML in the wrist must be examined to differentiate it from *** to surrounding tissues should be minimized during excision.
