RAPD-PCR Based Marker Approach for the Genetic Differentiation of Two Species of Cockroach （Order-Dictyoptera）

Random amplified polymorphic DNA （RAPD） analysis was conducted for the differentiation of two most commonly occurring insect species Periplaneta americana and Blatella germanicana. This technique is proved to be a quick and effective to establish genetic markers to differentiate morphologically similar populations. During the study cockroach species Periplanata americana and Blatella germanicana were considered. Ten random primers were used for polymerase chain reaction （PCR）. Many of such bands obtained, which differentiate between the two species. On the basis of interpretability, simplicity and reproducibility, six primers P1 （GATGACCGCC）, P3 （GGCACGTAAC）, P6 （GGTGCGCCTT）, P7 （GTCAGAGTCG）, P8 （GTCGCCGTCT） and PI0 （GTGCCCGATG） were considered positive for genetic differentiation and analysis. A series of bands ranging from -300 bp to -1,000 bp obtained indicates that these two species are related, however they exhibit some variations. It has also been observed that the same primers also amplified some DNA fragments of the same size in both the species, which indicates the presence of conserved regions, sharing ancestral relationship. Some of the fragments were unique in both the species which may be used for diagnostic purposes. The study concludes that the RAPD-PCR technique is useful for the study of molecular taxonomy in insects.

A Mathemataical model for a modified micro- cylinder electrode in which polyphenol oxidase ( PPO) occurs for all values of the concentration of catechol and o-quinone is analysed. This model is based on system of reaction-diffusion Equations containing a non-linear term related to Michaelis Menten kinetics of the enzymatic reaction. Here a new analytical technique Homotopy Perturbation Method is used to solve the system of non-linear differential Equations that describe the diffusion coupled with a Michaelis-Menten kinetics law. Here we report an analytical expressions pretaining to the concentration of catechol and o-quinone and corresponding current in terms of dimensionless reaction-diffusion parameters in closed form. An excellent agreement with available limiting case is noticed.

In this study, hybrid composites based on carbon fabric and epoxy (C/Ep) were fabricated by hand lay-up method followed by compression moulding. The C/Ep with optimum carbon fiber (60 wt%) was chosen as a reference material, and to it, the metallic nanoparticles like aluminum (Al) and zinc (Zn) of different wt% (0.5 and 1.0) were included as secondary fillers. To understand the synergism effect of these hybrid reinforcements, mechanical properties and tribological behavior of composites were studied. From the test results, it was proved that hybridization improved the mechanical and tribological properties. The C/Ep consisting of 0.5 wt% Zn and Al showed higher tensile properties in comparison with all other fabricated composites. Increase in flexural strength and flexural modulus also observed as the filler content increased in C/Ep composite. Higher impact strength is noted at 1 wt% Zn filled C/Ep composite. Wear test data revealed that 0.5 wt% Zn in C/Ep has got superior wear resistance. Wear mechanisms were discussed using scanning electron micrographs of selected worn surfaces of the composites.

A simple, precise, accurate stability-indicating gradient reversed-phase high-performance liquid chromatographic （RP-HPLC） method was developed for the quantitative determination of zotepine （ZTP） in bulk and pharmaceutical dosage forms in the presence of its degradation products （DPs）. The method was developed using Phenomenex C18 column （250 mm ~ 4.6 mm i.d., 5 mm） with a mobile phase containing a gradient mixture of solvents, A （0.05%trifluoroacetic acid （TFA）, pH ？ 3.0） and B （acetonitrile）. The eluted compounds were monitored at 254 nm;the run time was within 20.0 min, in which ZTP and its DPs were well separated, with a resolution of 41.5. The stress testing of ZTP was carried out under acidic, alkaline, neutral hydrolysis, oxidative, photolytic and thermal stress conditions. ZTP was found to degrade significantly in acidic, photolytic, thermal and oxidative stress conditions and remain stable in basic and neutral conditions. The developed method was validated with respect to specificity, linearity, limit of detection, limit of quantification, accuracy, precision and robustness as per ICH guidelines. This method was also suitable for the assay determination of ZTP in pharmaceutical dosage forms. The DPs were characterized by LC-MS/MS and their fragmentation pathways were proposed.

Objective:To characterize otologic clinical trials and examine otologic clinical trial trends from 2008 to 2018 using the *** ***:Data was collected from *** and included all clinical trials that focused on otology from 2008 to *** measures include status of trials,funding sources,details regarding otologic conditions studied,and trends in clinical ***:There were 992 otology clinical trials from 2008 to 2018.457(46.1%)were completed and 94(9.5%)were *** remained the highest(76.5%)contributor to otology clinical *** otologic conditions studied,from most common to least common,include hearing loss(40.6%),vestibulopathy(18.8%),tinnitus(18.8%),and otitis media(15.1%).The number of otology clinical trials increased by an average of 12.0 trials per year from 2008 to 2018(p<0.001).The number of otology clinical trials focusing on hearing loss and vestibulopathy significantly increased over the studied period(p<0.001),while those focusing on tinnitus and otitis media did not(p=0.09 and p=0.20,respectively).The majority of clinical trials on each of these four conditions focused on treatment ***:Our study describes trends in otology clinical trials registered on *** from 2008 through *** total number of clinical trials over this time period increased significantly,driven by trials investigating hearing loss and ***,most clinical trials were industrysponsored and focused on treatment *** study provides an outline of otology clinical trials registered in a US web-based database,which may be of use for the development of future clinical trials.

To evaluate the risk of immediate and delayed bleeding following sphincterotomy procedure. METHODSThis retrospective cohort study was conducted with all patients who underwent endoscopic sphincterotomy during January 2006 to September 2015 at a tertiary academic center. Patients were grouped according to pre procedural usage of serotonin reuptake inhibitors (SRIs). Both groups were matched for demographic and clinical characteristics. Patients with thrombocytopenia, increased international normalized ratio, or a history of bleeding or coagulation disorders, concurrent use of other antiplatelet/anticoagulants were excluded from the study. RESULTSA total of 447 patients were included, of which 219 (45.9%) used SRIs and 228 (54.1%) cases did not. There was no significant difference in acute or delayed bleeding during endoscopic sphincterotomy between the two groups. (8.2% vs 12.3%, P = 0.16). CONCLUSIONThe use of SRIs was not associated with an increased risk of post-sphincterotomy bleeding. To our best knowledge, this is the first study to explore this association.

The public health significance of Yersinia spp. gives a new dimension to the prevailing food chain, wherein the foods do get exposed to heat and cold treatments. In this study, the effect of heat treatment on the native isolates of Yersinia enterocolitica CFR 2301 and Y. intermedia CFR 2303 revealed the D-values ranging from the lowest of 0.08 min at 65℃ in skim milk/beef gravy to the highest of 18.52 min at 50℃ in beef gravy. The heat sensitivity of both these cultures was in the order of Milli-Q water > 0.85% saline > skim milk > beef gravy. The z-values of the test cultures ranged from 7.55℃ for Y. intermedia to 12.08℃ for Y. enterocolitica. The heat sensitivity in Y. enterocolitica appeared to be related with growth incubation temperatures and also fatty acid profile of cell membrane. The effect of low temperature treatments (–20 ℃, 0℃ and 4℃ for 20 d) in water, saline and skim milk revealed the ability of Y. enterocolitica to survive more efficiently at –20℃, while Y. intermedia was more tolerant at 0℃. In packaged drinking water, Y. enterocolitica could survive and grow at 4℃ and 16℃, while at 30℃, inactivation was rapid. The findings did indicate that heat and cold treatments would not always ensure safety from Y. enterocolitica and Y. intermedia in the food chain.

Alumina coating was formed on AA7075 aluminum alloy by micro arc oxidation（MAO） method and its corrosion and stress corrosion cracking（SCC） behaviors were examined in 3.5%（mass fraction） NaCl *** impedance spectroscopy（EIS） was used to evaluate the degradation of the coating as a function of immersion time and was modeled with appropriate equivalent *** load stress corrosion cracking（SCC） results followed by post-test metallographic observations demonstrated the usefulness of MAO coating to avoid the premature failure of the alloy due to severe localized corrosion initiated by Cu-and Fe-rich intermetallic phases.
