Heteromeric and Homomeric Geranyl Diphosphate Synthases from Catharanthus roseus and Their Role in Monoterpene Indole Alkaloid Biosynthesis

Catharanthus roseus is the sole source of two most important monoterpene indole alkaloid （MIA） anti- cancer agents： vinblastine and vincristine. MIAs possess a terpene and an indole moiety derived from terpenoid and shikimate pathways, respectively. Geranyl diphosphate （GPP）, the entry point to the formation of terpene moiety, is a product of the condensation of isopentenyl diphosphate （IPP） and dimethylallyl diphosphate （DMAPP） by GPP synthase （GPPS）. Here, we report three genes encoding proteins with sequence similarity to large subunit （***） and small subunit （***） of heteromeric GPPSs, and a homomeric GPPSs. *** is a bifunctional enzyme producing both GPP and geranyl geranyl diphosphate （GGPP）, *** is inactive, whereas CrGPPS is a homomeric enzyme forming GPP. Co-expression of both subunits in Escherichia coil resulted in heteromeric enzyme with enhanced activity producing only GPR While *** and CrGPPS showed higher expression in older and younger leaves, respectively, *** showed an increasing trend and decreased gradually. Methyl jasmonate （MelA） treatment of leaves sig- nificantly induced the expression of only ***. GFP localization indicated that *** is plastidial whereas CrGPPS is mitochondrial. Transient overexpression of *** in C. roseus leaves resulted in increased vindoline, immediate monomeric precursor of vinblastine and vincristine. Although C. roseus has both heteromeric and homomeric GPPS enzymes, our results implicate the involvement of only heteromeric GPPS with *** regulating the GPP flux for MIA biosynthesis.

Change in global climate is primarily due to rising concentrations of greenhouse gases in the atmosphere that is mostly caused by human *** important factors affecting the occurrence and spread of the plant diseases are temperature,moisture,light,and CO_(2) *** factors cause physiological changes in plants that result in increase in intensity of crop *** change causes a significant impact on germination,reproduction,sporulation and spore dispersal of *** change affects all life stages of the pathogen as well as its host to cause impact on host-pathogen interaction which facilitates the emergence of new races of the pathogen ultimately breakdowns the host *** also affects the microbial community in the soil which is beneficial to the plants in various *** minor diseases become major ones due to alteration in climatic parameters thus posing a threat to the food security.

Improvement of Bio-Efficacy to Reduce Bacterial Wilt Complex Disease in Tomatoes through Bacillus amyloliquefaciens and Pseudomonas fluorescens under Field Conditions

Bacterial wilt complex disease of tomato(Solanum lycopersicum L.)was incited jointly by bacterial wilt pathogen Ralstonia solanacearum and Meloidogyne incognita ***-efficacy of bacterial antagonists *** DSBA-11 and *** DTPF-3 was studied against the wilt disease complex in tomato at National Phytotran facility Indian Agricultural Research Institute(IARI),New Delhi,at 26±2°*** wilt disease incidence(26.00%)with the highest bio-control efficacy(64.15%),less juvenile population(19.33 J2/g of soil)of *** was recorded in the combined application of DTPF-3+DSBA-11 after 30 d of inoculation under glasshouse *** a field study,minimum bacterial wilt disease incidences 19.0%and 20.4%were recorded in the bleaching powder treatment followed by mixed application of DSBA-11+DTPF-3,19.6%and 21.2%wilt incidence in 2014 and 2015 ***,a reduction of root-knot gall index was recorded a maximum of 59.76%and 69.62%in DSBA-11+DTPF-3 treated plants followed by 54.88%and 60.13%over control in DTPF-3 treatment in 2014 and 2015 *** yield of tomato fruit was increased over control by 17.48%and 16.97%in 2014 and 2015 respectively under field conditions.A combination of *** DTPF-3+*** DSBA-11 suppressed bacterial wilt and root-knot diseases and also increased the yield of the tomato fruit significantly(p<0.05)under field conditions.

Conductivity Studies in Proton Irradiated AgI-Ag2O-V2O5-TeO2 Super-Ionic Glass System

The electrical properties of proton ion beam irradiated glass samples are carried out by impedance spectroscopy in the frequency range from 10 Hz to 32 MHz. The ion beam of energy 3 MeV and fluence of of 1014 particles cm-2 was chosen for irradiation. The conductivity of the super ionic glass samples increases after irradiation and other electrical parameters like dielectric constant, dielectric loss and modulus of the proton irradiated glass samples as a function of glass composition and temperature are observed to change. The dielectric constant and the dielectric losses are increased after irradiation and the modulus parameters confirm the non-Debye nature for irradiated samples also.

Digitally Programmable Voltage Mode Universal Filters—A Minimal Realization

Three voltage mode universal filter biquads using digitally programmable second generation current conveyor (CCII) are presented in this paper. Salient features of proposed filter configurations include realization of all the filter functions except allpass, independently programmable filter parameters, no component matching constraint and low sensitivity figure. Component count of proposed filter configurations is proved to be minimal for offering aforementioned set of features. Workability of proposed circuit is verified by including the SPICE simulations.

Impact of nutritional support on nutritional status and nitrogen balance in surgical patients of a developing country

To assess the nutritional status, its change after surgery and impact of nutritional support on urinary nitrogen losses (UNL) and nitrogen balance (NB) based on changing serum albumin levels in a developing country, 89 patient (58 M) aged 39 yrs ±15 were studied. Nutritional assessment was done at admission and 2 weeks postoperatively. UNL and NB was assessed on postoperative day one (D1), two (D2), four (D4), six (D6), eight (D8) and eleven (D11) respectively. Malnutrition varied from 27 (30.3%) using body mass index to 78 (87%) by triceps skin fold thickness. Patients with falling or static serum albumin levels (Group I) showed a significant decrease (‘p’ = 0.001) in their nutritional parameters compared to those with increasing albumin (Group II). Postoperatively till D2 both groups had similar negative NB but Group II improved earlier to become positively balanced by D11 (‘p’ < 0.001). Regression analysis of nitrogen intake (Ni) on UNL on each day revealed negative association till D2 to become positively associated from D4 onwards being significant on D4 (‘p’ = 0.001) & D6 (‘p’ = 0.05*). There is a significant nutritional depletion after surgery especially in patients with falling/static serum albumin levels and NB improves earlier in patients with rising albumin levels. Increasing Ni helps in decreasing UNL.

Single VDVTA-Based Voltage-Mode Biquad Filter

In this paper, an application of voltage differencing voltage transconductance amplifier (VDVTA) in the realization of voltage-mode (VM) multi-input single output (MISO) type biquad is presented. The proposed topology uses one VDVTA as an active element, two capacitors and a grounded resistor. The configuration realizes low pass (LP), high pass (HP), band pass (BP) and notch (BR) filters without the requirement of any matching condition. The natural frequency (w0) and bandwidth (BW) are independently controllable. The proposed circuit offers low active and passive sensitivities of w0. The operation of the proposed circuit has been verified through SPICE simulation with TSMC CMOS 0.18 μm process parameters.

Disease prediction in plants has acquired much attention in recent *** factors such as:temperature,relative humidity,rainfall,sunshine play an important role in a plan’s growth only if they are present in adequate amounts as required by the *** the other hand,if the factors are inadequate,they may also support the growth of a disease in the *** current study focuses on the Rust disease in Aonla fruits and leaves by utilizing a real time dataset of weather *** different models are tested for spray prediction on conducive *** resampling techniques,random over sampling(ROS)and synthetic minority oversampling technique(SMOTE)have been used to balance the dataset and five different classifiers:support vector machine(SVM),logistic regression(LR),k-nearest neighbor(kNN),decision tree(DT)and random forest(RF)have been used to classify a particular day based on weather conditions as conducive or *** classifiers are then evaluated based on four performance metrics:accuracy,precision,recall and *** results indicate that for imbalanced dataset,kNN is appropriate with high precision and recall *** both balanced and imbalanced dataset models,the proposed model SMOTE-RF performs best among all models with 94.6%accuracy and can be used in a real time application for spray ***,timely fungicide spray prediction without over spraying will help in better productivity and will prevent the yield loss due to rust disease in Aonla crop.

A Novel Packet Switch Node Architecture for Contention Resolution in Synchronous Optical Packet Switched Networks

Packet contention is a key issue in optical packet switch (OPS) networks and finds a viable solution by including optical buffering techniques incorporating fiber delay lines (FDLs) in the switch architecture. The present paper proposes a novel switch architecture for packet contention resolution in synchronous OPS network employing the packet circulation in FDLs in a synchronized manner. A mathematical model for the proposed switch architecture is developed employing packet queuing control to estimate the blocking probability for the incoming traffic. The switch performance is analyzed with a suitable contention resolution al-gorithm through the computer simulation. The simulation results substantiate the proposed model for the switch architecture.

The Effect of Intraperitoneal Ropivacaine for Post-Operative Pain Management in Patients Undergoing Laparoscopic Cholecystectomy: A Prospective Double-Blind Randomized Control Study

Objective: To compare the effect of intraperitoneal ropivacaine with placebo for post-operative pain management in patients undergoing laparoscopic cholecystectomy. Material and Methods: All patients were pre-medicated with glycopyrrolate 0.2 mg, ondansetron 4 mg and ranitidine 150 mg intravenously half an hour prior to induction of anesthesia. All patients were given standard general anaesthesia with propofol (2-2.5 mg/kg), fentanyl 2 μg/kg, and succinylcholine (2 mg/kg) to facilitate tracheal intubation. Anesthesia was maintained with 60% N2O in oxygen with 0.5% to 1% Halothane. Group A: Patients received 20 ml of 0.9% normal saline as placebo (n = 25). Group B: Patients received 20 ml of 0.5% ropivacaine (n = 25). Results: The age and sex distribution of both the groups was similar. The heart rate, systolic & diastolic blood pressure, mean blood pressure and mean trend of SpO2 in both groups remained similar over the periods. The mean VAS varied considerably within (between time) and between the groups (treatment) and was especially comparatively higher in Group A at initial hours 15 min to 30 min and at end hours 12-24 hrs as compared with Group B. On an average, the frequent dosing of rescue analgesia and mean No. of rescue analgesia doses were higher in Group A followed by Group B. In both groups, the treatment related adverse events were mostly emetic symptoms and shoulder pain with the highest being in Group A. Conclusion: We conclude that intraperitoneal instillation of local anaesthetic is an easy, cheap, and non-invasive method which provides good analgesia in the immediate postoperative period after laparoscopic surgery.
