The heterogeneity of exhausted T cells(Tex)is a critical determinant of immune checkpoint blockade therapy ***,few studies have explored exhausted T cell subpopulations in human *** the present study,we examined samples from two cohorts of 175 patients with head and neck squamous cell cancer(HNSCC)by multiplex immunohistochemistry(mIHC)to investigate two subsets of Tex,CD8+PD1+TCF1+progenitor exhausted T cells(TCF1+Texprog)and CD8+PD1+TCF1−terminally exhausted T cells(TCF1−Texterm).Moreover,fresh tumor samples from 34 patients with HNSCC were examined by flow cytometry and immunohistochemistry to further investigate their properties and cytotoxic capabilities and their correlation with regulatory T cells(Tregs)in the tumor immune microenvironment(TIME).mIHC and flow cytometry analysis showed that TCF1−Texterm represented a greater proportion of CD8+PD1+Tex than TCF1+Texprog in most ***1+Texprog produced abundant TNFα,while TCF1−Texterm expressed higher levels of CD103,TIM-3,CTLA-4,and ***1−Texterm exhibited a polyfunctional TNFα+GZMB+IFNγ+phenotype;and were associated with better overall survival and recurrence-free *** results also indicated that larger proportions of TCF1−Texterm were accompanied by an increase in the proportion of ***,it was concluded that TCF1−Texterm was the major CD8+PD1+Tex subset in the HNSCC TIME and that these cells favor patient survival.A high proportion of TCF1−Texterm was associated with greater Treg abundance.

Prognostic value of tertiary lymphoid structure and tumour infiltrating lymphocytes in oral squamous cell carcinoma

Tertiary lymphoid structures(TLS)are ectopic lymphoid structures in cancers that are largely associated with favourable ***,the prognostic value of TLSs in oral squamous cell carcinoma(OSCC)is largely unknown,and the association between tumour infiltrating lymphocytes(TILs)and TLSs has been rarely explored in *** this study,associated markers of TLS,including peripheral node address(PNAd)in high endothelial venules,CD20 in B cells and CD3 in T cells,were examined in 168 OSCC patients,and survival analysis was performed between TLS-positive and TLS-negative *** detected the presence of TILs by staining CD8+cytotoxic T cells and CD57+NK cells as *** appeared as highly organized structures in 45(26.8%)*** patients had a better 5-year overall survival(OS)rate(88.9%vs.56.1%,P<0.001)and relapse-free survival(RFS)rate(88.9%vs.63.4%,P=0.002).Moreover,the presence of TLS was an independent prognostic factor for both the 5-year OS rate(hazard ratio[HR]=3.784;95%confidence interval[CI],1.498–9.562)and RFS rate(HR=3.296;95%CI,1.279–8.490)in multivariate ***,a higher density of CD8+T cells and CD57+NK cells was found in TLS-positive sections than in TLS-negative counterparts(P<0.001),and their combination provided a higher predictive accuracy(AUC=0.730;95%CI,0.654–0.805).In conclusion,our results suggest that TLS is an independent positive prognostic factor for OSCC *** findings provide a theoretical basis for the future diagnostic and therapeutic value of TLSs in OSCC treatment.

The appropriate catalysts can accelerate the reaction rate and effectively boost the efficient conversion of various molecules,which is of great importance in the study of chemistry,chemical industry,energy,materials and environmental ***,efficient,environmentally friendly,and easy to operate synthesis methods have been used to prepare various types of *** previous studies have reported the synthesis and characterization of the aforementioned catalysts,more still remain in trial and error methods,without in-depth consideration and improvement of traditional synthesis ***,we comprehensively summarize and compare the preparation methods of the trial-and-error synthesis strategy,structure–activity relationships and density functional theory(DFT)guided catalysts rational design for nanomaterials and atomically dispersed *** also discuss in detail the utilization of the nanomaterials and single atom catalysts for converting small molecules(H_(2)O,O_(2),CO_(2),N_(2),etc.)into value-added products driven by electrocatalysis,photocatalysis,and ***,the challenges and outlooks of mass preparation and production of efficient and green catalysts through conventional trial and error synthesis and DFT theory are featured in accordance with its current development.

口咽癌是头颈鳞癌中重要的癌种之一,与人类乳头瘤状病毒(human papillomavirus, HPV)感染有关,其发病率逐年上升。但在临床治疗过程发现,HPV 阳性口咽癌患者整体预后好于 HPV 阴性患者。目前,导致这一现象发生的分子机制尚不明确。本研究利用 TCGA 数据库,对 HPV 阳性和阴性患者的免疫细胞浸润和效应功能以及新抗原负荷进行了生物信息学分析。结果发现,HPV 阳性患者的总体生存率比 HPV 阴性患者显著提高。对肿瘤组织中的免疫细胞相对丰度进行分析显示,HPV 阳性患者的 CD8+ T 细胞较 HPV 阴性患者显著提高,其效应分子 IFN-γ和 Granzyme B 表达量显著升高。同时对新抗原数目进行分析发现,HPV 阳性患者肿瘤组织中新抗原较 HPV 阴性患者低。本研究结果为 HPV 相关口咽癌的治疗提供了一定的基础理论支撑。

Numerical Analysis of PRISM-PY Calculations for Hard-and Soft-Core Generic Polymer Models

Generic polymer models capturing the chain connectivity and excluded-volume interactions between polymer segments can be classified, according to whether or not the 3D integral of the latter diverges, into hard- and soft-core models. Taking homogeneous systems of compressible homopolymer melts (or equivalently homopolymer solutions in an implicit, good solvent) in the continuum as an example, we recently compared the correlation effects on the structural and thermodynamic properties of the hard- and soft-core models given by the polymer reference interaction site model (PRISM) theory with the Percus-Yevick (PY) closure (Polymers 2023, 15, 1180). Here we analyzed in detail the numerical errors and behavior of the interchain pair correlation functions (PCFs) given by the PRISM-PY calculations of these models using an efficient numerical approach that we proposed. Our numerical approach has the least number of independent variables to be iteratively solved, analytically treats the discontinuities caused by the non-bonded pair potential (such as that of the hard spheres) and takes only the inverse Fourier transform of the interchain indirect PCF between polymer segments (which is continuous and decays towards 0 with increasing wavenumber much faster than both the interchain direct and total PCFs), and is essential for us to accurately solve the PRISM-PY theory for chain length N as large as 106. To capture the correlation-hole effect, the real-space cut-off in the PRISM calculations should be proportional to the square root of N.

Linear-Quadratic Pareto Cooperative Game for Mean-Field Backward Stochastic System

This paper focuses on a Pareto cooperative differential game with a linear mean-field backward stochastic system and a quadratic form cost functional. Based on a weighted sum optimality method, the Pareto game is equivalently converted to an optimal control problem. In the first place,the feedback form of Pareto optimal strategy is derived by virtue of decoupling technology, which is represented by four Riccati equations, a mean-field forward stochastic differential equation(MF-FSDE),and a mean-field backward stochastic differential equation(MF-BSDE). In addition, the corresponding Pareto optimal solution is further obtained. Finally, the author solves a problem in mathematical finance to illustrate the application of the theoretical results.

A multi-source information fusion layer counting method for penetration fuze based on TCN-LSTM

When employing penetration ammunition to strike multi-story buildings,the detection methods using acceleration sensors suffer from signal aliasing,while magnetic detection methods are susceptible to interference from ferromagnetic materials,thereby posing challenges in accurately determining the number of *** address this issue,this research proposes a layer counting method for penetration fuze that incorporates multi-source information fusion,utilizing both the temporal convolutional network(TCN)and the long short-term memory(LSTM)recurrent *** leveraging the strengths of these two network structures,the method extracts temporal and high-dimensional features from the multi-source physical field during the penetration process,establishing a relationship between the multi-source physical field and the distance between the fuze and the target plate.A simulation model is developed to simulate the overload and magnetic field of a projectile penetrating multiple layers of target plates,capturing the multi-source physical field signals and their patterns during the penetration *** analysis reveals that the proposed multi-source fusion layer counting method reduces errors by 60% and 50% compared to single overload layer counting and single magnetic anomaly signal layer counting,*** model's predictive performance is evaluated under various operating conditions,including different ratios of added noise to random sample positions,penetration speeds,and spacing between target *** maximum errors in fuze penetration time predicted by the three modes are 0.08 ms,0.12 ms,and 0.16 ms,respectively,confirming the robustness of the proposed ***,the model's predictions indicate that the fitting degree for large interlayer spacings is superior to that for small interlayer spacings due to the influence of stress waves.
