Personalized nutrition to mitigate inflammation in genetically predisposed individuals: a secondary analysis of the Danish PREVENToMICs intervention

Deep phenotyping and genetic characterization of individuals are fundamental to assessing the metabolic status and determining nutrition-specific requirements. This study aimed to ascertain the utmost effectiveness of personalized interventions by aligning dietary adjustments with both the genotype and metabolotype of individuals. Therefore， we assessed here the usefulness of a polygenic score (PGs) characterizing a potential pro-inflammatory profile (PGsi) as a nutrigenetic tool to discern individuals from the Danish PREVENToMICs cohort that could better respond to precision nutrition (PN) plans， specifically targeted at counteracting the low-grade inflammatory profile typically found in obesity. The cohort followed a PN plan to counteract the pro-inflammatory profile (PNi group) or generic dietary recommendations (Control) for 10 weeks. PGsi was applied for genetic stratification (low/High). The effects of the intervention on anthropometrics and biomarkers related to inflammatory profile and carbohydrate metabolism were assessed. Around 30% of subjects had a high genetic predisposition to pro-inflammatory status (high-PGsi). These individuals demonstrated the most effective response to the dietary plan， experiencing improved body composition， with significant decreases in body weight (△: -4.84%; p=0.039) and body fat (△: -4.86%; p=0.007)， and beneficial changes in pro- and anti-inflammatory biomarkers， with significant increases in Il-10 (△: 71.3%; p=0.025) and decreases in TNF-α (△: -3.0%; p=0.048)， CRP (△: -31.1%)， ICAM1 (△: -5.8%)， and MCP1 (△: -4.2%) circulating levels， compared to low-PGsi individuals. Both phenotypic and genetic stratification contributed to a better understanding of metabolic heterogeneity in response to diet. This approach allows for refinement of the prediction of individual requirements and potentially for better management of obesity.

The study of the propagation of infectious diseases in urban centers finds a close connection with their population's social characteristics and *** work performs a spatial analysis of dengue cases in urban centers based on the basic reproduction numbers,R0,and incidence by planning areas(PAs),as well as their correlations with the Human Development Index(HDI)and the number of *** analyzed dengue epidemics in 2002 at two Brazilian urban centers,Belo Horizonte(BH)and Rio de Janeiro(RJ),using PAs as spatial *** results reveal heterogeneous spatial scenarios for both cities,with very weak correlations between R0 and both the number of trips and the HDI;in BH,the values of R0 show a less spatial heterogeneous pattern than in *** BH,there are moderate correlations between incidence and both the number of trips and the HDI;meanwhile,they weakly correlate for ***,the absence of strong correlations between the considered measures indicates that the transmission process should be treated considering the city as a whole.

C-reactive protein levels during a relapse of Crohn’s disease are associated with the clinical course of the disease

AIM： To explore if C-reactive protein （CRP） levels might serve as a prognostic factor with respect to the clinical course of Crohn＇s disease and might be useful for classification. METHoDs： In this retrospective cohort study we enrolled 94 patients from the inflammatory bowel disease （IBD） database of the University Medical Centre Utrecht. CRP levels during relapse were correlated with the number of relapses per year. severity of relapses was based on endoscopic reports and prednisone use. Furthermore, patients were categorized in a low or high CRP group based on their CRP response during relapse and demographic and clinical features were compared. REsUlTs： overall, a positive correlation between CRP levels, number of relapses, and severity of relapse was found （respectively rs = 0.31, P 〈 0.01 and rs = 0.50, P 〈 0.001）. Employing a cut-off level of 15 mg/l, the index CRP level was found to discriminate patients with respect to the number of relapses per year, as well as for severity of relapses （respectively 0.25 ± 0.16 vs 0.36 ± 0.24, P 〈 0.05 and 4.4 ± 1.2 vs 3.2 ± 1.1 on a 10-point visual analogue scale, P 〈 0.001 for the high CRP and low CRP groups respectively）. In addition, the high CRP group showed more cumulative days of prednisone use per year （107 ± 95 vs 58 ± 48, P 〈 0.05）, as well as a better response to infiiximab （93 % vs 33 %, P = 0.06）. CoNClUsIoN： A higher CRP level during relapse seems to be associated with a more severe clinical course of disease.

Ba-REE fluorcarbonate minerals from a carbonatite dyke at Bayan obo, Inner Mongolia, North China, are first reported in this contribution, in which chemical composition, rare earth element (REE) patterns, and intergrowth relationships for these minerals have been investigated. syntactic intergrowth or syntaxy between cebaite and cordylite, as well as cordylite and huanghoite were observed. This syntactic texture resulted from the variation of chemical composition of crystallizing agents for those minerals that crystallized directly from carbonatite magmas. It is worth noting that REE patterns of the Ba-REE fluorcarbonate minerals in the dyke are similar to those of the corresponding minerals from the ore hosted dolomite marble of the Bayan obo giant REE-Nb-Fe mineral deposit, which implies their relation in origin.

Peroxynitrite induced decrease in Na^+, K^+-ATPase activity is restored by taurine

AIM: Peroxynitrite (oNoo-) is a powerful oxidant shown to damage membranes. In the present study, the effect of taurine on changes of liver plasma membrane Na+, K+-ATPase induced by oNoo- was investigated. METHoDs: liver plasma membrane was exposed tooNoo-with or without taurine. Na+, K+-ATPase activity and lipid peroxidation as thiobarbituric acid reactive substances (TBARs) levels were ***: Different concentrations of oNoo- (100, 200,500, and 1 000 μmol/l) were found to decrease liver plasma membrane Na+, K+-ATPase activity significantly. The depletion of enzyme activity was not concentration dependent. Effects of different concentrations of taurine on liver plasma membrane Na+, K+-ATPase activity were also measured. Taurine did not cause any increase in enzyme activity. when plasma membranes were treated with 200 μmol/l oNoo- with different concentrations of taurine, a restoring effect of taurine on enzyme activity was observed. TBARs levels were also measured and taurine was found to decrease the elevated values. CoNClUsIoN: Taurine is observed to act as an antioxidant of oNoo-to decrease lipid peroxidation and thus affect liver plasma membrane Na+, K+-ATPase by restoring its activity.
