The advantages of comb.ning qualitative and quantitative analy.is on a single analy.ical technique have further extended the applications of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.(MALDI-TOF MS) to the quantitation of various b.omolecules. To achieve ab.olute quantitation, it is necessary.to perform a calib.ation with standard dilutions. For the purpose of measuring DNA samples, a pure DNA oligonucleotide at different concentrations was chosen as a standard to perform the calib.ation of MALDI-TOF MS. In order to overcome the variation of signal intensity.from repeated measurements of each DNA standard dilution, fixed amount of an internal standard was added into each DNA standard dilution. Instead of maintaining at a constant level, the signals of fixed amount of internal standard were decreased 73% from its initial level while the signals of DNA standard continued to increase within a linear dy.amic range for quantitation from 0.20 μM to 12.5 μM of DNA. Attempts to identify.the cause of signal reduction were sy.tematically.carried out. This is the first report on the extent of signal reduction in quantitative MALDI-TOF MS. These results represent a limitation on using MALDI-TOF MS to monitor the changes in concentration of two different compounds within a chemical or b.ological sy.tem.

Tumour induced osteomalacia(TIO) is a rare and often unrecognized cause of hy.ophosphatemia. We report on a case of TIO due to a hemangiopericy.oma originating from the left nasal sinus. The patient was a 55-y.ar-old male with a 3-y.ar history.of left hip pain and an undisplaced left hip fracture. b.ochemical testing demonstrated low levels of serum phosphate and serum 1,25-dihy.roxy.itamin D, and an elevated level of fib.ob.ast growth factor 23. Octreotide scanning demonstrated uptake in the left nasal sinus area and a computed tomography.scan revealed a left nasal sinus mass. The patient underwent surgical resection of the mass and histology.was consistent with a sinonasal hemangiopericy.oma. His serum phosphate levels normalized almost immediately.after surgery.and he had complete resolution of hip pain. Our case highlights the importance of considering TIO when assessing patients with low serum phosphate.

A tab.et matrix sy.tem was developed for ib.profen and the influence of the poly.er b.end and concentration on the release rate of the drug was *** containing different concentrations of calcium silicate and PVP(poly.iny. py.rolidone)were prepared using direct compression and the weight uniformity.crushing strength,friab.lity.drug content uniformity.dissolution profile,and in vitro release kinetics were *** tab.ets were found to b. within the official acceptab.e limits of phy.ical and chemical parameters except for the thickness test that was b.low the conformation of extended-release *** crushing strength of the tab.ets was in the range of 2.5 to 5.6 kg/f,the weight variations of the tab.ets of all the formulation was less than±5%.The friab.lity.of all the formulations was in the range of 0.6%to 1.83%.Tab.et thickness and diameter was in range of 3.18 mm to 4.48 mm and 12.53 mm to 12.64 mm *** drug contents of all the formulations were found to b. in range of 83.50%to 98%.The release kinetic of F3 containing 20 mg of calcium silicate,40 mg of PVP as matrix formers showed the b.st linearity.r^2=0.6975)with%drug release of 96 showing that comb.nation of the two poly.ers(20 mg calcium silicate and 40 mg PVP)for use as a matrix former is b.st for extended-release formulation of ib.profen.
