Molecular recognition of live methicillin-resistant staphylococcus aureus cells using DNA aptamers

AIM: To generate DNA-aptamers binding to Methicillinresistant Staphylococcus aureus(MRSA).METHODS: The Cell-Systematic Evolution of Ligands by Exponential Enrichment(SELEX) technology was used to run the selection against MRSA bacteria and develop target-specific aptamers. MRSA bacteria were targeted while Enterococcus faecalis bacteria were used for counter selection during that process. Binding assays to determine the right aptamer candidates as well as binding assays on clinical samples were performed through flow cytometry and analyzed using the FlowJ o software. The characterization of the aptamers was done by determination of their Kd values and determined by analysis of flow data at different aptamer concentration using Sigma Plot. Finally, the recognitionof the complex Gold-nanoparticle-aptamer to the bacteria cells was observed using transmission electron microscopy(TEM).RESULTS: During the cell-SELEX selection process, 17 rounds were necessary to generate enrichment of the pool. While the selection was run using fixed cells, it was shown that the binding of the pools with live cells was giving similar results. After sequencing and analysis of the two last pools, four sequences were identified to be aptamer candidates. The characterization of those aptamers showed that based on their Kd values, DTMRSA4 presented the best binding with a Kd value of 94.61 ± 18.82 nmol/L. A total of ten clinical samples of MRSA, S. aureus and Enterococcus faecalis were obtained to test those aptamers and determine their binding on a panel of samples. DTMRSA1 and DTMRSA3 showed the best results regarding their specificity to MRSA, DTMRSA1 being the most specific of all. Finally, those aptamers were coupled with gold-nanoparticle and their binding to MRSA cells was visualized through TEM showing that adduction of nanoparticles on the aptamers did not change their binding ***: A total of four aptamers that bind to MRSA were obtained with Kd values ranking from 94

Measuring the temperature of the spin via weak measurement

In this study,we give a new proposal to measure the temperature of the spin in a sample in a magnetic resonance force microscopy system by using postselected weak measurement,and investigating the Fisher information to estimate the precision of the *** show that in a high temperature regime the temperature of the spin can be measured via a weak measurement technique with proper postselection and our scheme is able to increase the precision of temperature estimation.

Effects of postselected von Neumann measurement on the properties of single-mode radiation fields

Postselected von Neumann measurement characterized by postselection and weak value has been found to possess potential applications in quantum metrology and solved plenty of fundamental problems in quantum theory. As an application of this new measurement technique in quantum optics and quantum information processing, its effects on the features of single-mode radiation fields such as coherent state, squeezed vacuum state and Schrödinger cat sate are investigated by considering full-order effects of unitary evolution. The results show that the conditional probabilities of finding photons, second-order correlation functions, Qm-factors and squeezing effects of those states after the postselected measurement is significantly changed are comparable with the corresponding initial pointer states.

The promise of curing many untreatable forms of human male infertility is now one small step closer based on a recent paper in Cell. In this work, scientists from Kyoto University in Japan created mature, fertile sperm from embryonic and induced pluripotent stem cells by grabbing the primitive germ cells called primordial germ cells just as they were being made from their pluripotent precursors in vitro and transplanting them back into genetically sterile mice.

Numerical Studies of Vanka-Type Smoothers in Computational Solid Mechanics

In this paper multigrid smoothers of Vanka-type are studied in the context of Computational Solid Mechanics(CSM).These smoothers were originally developed to solve saddle-point systems arising in the field of Computational Fluid Dynamics(CFD),particularly for incompressible flow *** treating(nearly)incompressible solids,similar equation systems arise so that it is reasonable to adopt the‘Vanka idea’for *** there exist numerous studies about Vanka smoothers in the CFD literature,only few publications describe applications to solid mechanical *** this paper we want to contribute to close this *** depict and compare four different Vanka-like smoothers,two of them are oriented towards the stabilised equal-order Q_(1)/Q_(1)finite element *** means of different test configurations we assess how far the smoothers are able to handle the numerical difficulties that arise for nearly incompressible material and anisotropic *** the one hand,we show that the efficiency of all Vanka-smoothers heavily depends on the proper parameter *** the other hand,we demonstrate that only some of them are able to robustly deal with more critical ***thermore,we illustrate how the enclosure of the multigrid scheme by an outer Krylov space method influences the overall solver performance,and we extend all our examinations to the nonlinear finite deformation case.

Proposed EU legislation to force changes in sewage sludge disposal:A case study

The consequences of changes planned in the European Union legislation relevant to the disposal of sewage sludges are discussed. A specific municipal waste water treatment plant is analyzed in terms of drying and subsequent combustion or pyrolysis of the produced stabilized sludge, and the respective net energy balances are carried out. A simplified economic analysis of the two disposal options is presented, which suggest that combustion of the sludge would be economically infeasible while pyrolysis of the sludge in a modular, self-sufficient container unit can bring a small financial benefit due to the selling of the produced phosphorus-rich biochar.

Determination of a potential set of factors which influence the situation of methane emission into roadways being driven

The currently applied methodology of absolute gassiness forecasting in roadways being driven in Polish hard coal mines is based on the results of investigations frombefore more than twenty years. At present the roadways are being driven in majority ofcases in quite different conditions than those which had been the subject of the said investigations. The application of 'old' methods of forecasting of methane emission during driving of roadways often brings about great discrepancies between the results of the forecasts and actual situation. It is firstly necessary to determine a potential set of factorswhich exert an influence on situation regarding methane emission into roadways beingdriven in order to prepare a verified mathematical model of absolute gassiness forecastingOn the basis of available literature and the investigations conducted by the Central MiningInstitute an initial list of factors has been prepared which can have an impact upon methane emission into roadways being driven. The assumptions of heuristic method have beentaken advantage of for selection of parameters and indices describing the phenomenon ofmethane emission. It has been dictated by the fact that the available literature is lacking apretty unambiguous viewpoint on the subject under consideration and at the same time itis necessary to perform an a priori assessment of the information contained in a givenfeature (factor). A prepared initial set has been scrutinized to be verified on the basis of aconducted expert survey among seventy specialists in this field. After collecting of the results of the survey their statistical elaboration was prepared. The co-efficient of agreementof multiple ordering, the so called concordance co-efficient has been taken advantage of inorder to check the competency of experts. Hydraulic energy indices reveal spatial dependence in a subtropical soil under maize crop in Southern Brazil

Soil physical quality(SPQ)assessment is an important part in the evaluation of soil use,management,and *** can be assessed using several physical properties,hydraulic indices,and *** from tropical and temperate regions represent different physical behaviors,and the quantification of their physical properties is important to support soil evaluation and *** objective of this study was to evaluate the SPQ in a subtropical field under maize crop cultivation according to its physical properties,hydraulic indices,and functions in an attempt to infer the spatial variability and to determine the behavior of soil physical structure across a spatial *** used soil key physical variables,such as texture,bulk density,total porosity,saturated hydraulic conductivity,and organic carbon content,were measured in a regular grid with a soil sampling density of 30 points per hectare,covering an area of0.5 *** hydraulic conductivity varied strongly between subsamples and in the field,suggesting the heterogeneity of the soil structure regarding water *** physical variables were combined with other indicators,which were based on the soil water retention curve and the pore size distribution(PSD)*** analysis was performed to verify the relationship between the measured and calculated variables,and some strong linear correlations were revealed,such as between aeration energy index and microporosity(r=0.608)and water retention energy index with microporosity(r=0.532)and with bulk density(r=0.541).For most sampled locations,the shape and location parameters of PSD showed results outside of the optimum ranges,whereas the hydraulic energy indices and cumulative hydraulic energy functions presented values that were similar to those found for some tropical soils described in the *** spatial variability of these indices was described using semivariograms and kriged maps,indicating the variability of the SPQ i

Prolonged hypervolemic hemodilution decreases functional capillary density of ileal mucosa in pigs revealed by sidestream dark-field imaging

Objective： Hemodilution changes the physical properties of blood by reducing its hematocrit and blood viscosity. We tested whether prolonged hypervolemic hemodilution （HHD） impairs functional capillary density （FCD） of ileal mucosa in healthy mechanically-ventilated pigs and if there is any correlation between changes in FCD of ileal and sublingual mucosas during HHD. Methods： Sixteen domestic female pigs were anesthetized, mechanically-ventilated, and randomly assigned to the HHD （20 ml/（kg.h） Hartmann＇s solution for 3 h） or fluid restrictive （5 ml/（kg-h） Hartmann＇s solution for 3 h） group. Microcirculations of sublingual and ileal mucosas via ileostomy were visualized using sidestream dark-field （SDF） imaging at baseline conditions （t=0 h） and at selected time intervals of fluid therapy （t=-l, 2, and 3 h）. Results： A significant decrease of ileal FCD （285 （278-292） cm/cm^2） in the HHD group was observed after the third hour of HHD when compared to the baseline （360 （350-370） cm/cm2）（P〈0.01）. This trend was not observed in the restrictive group, where the ileal mucosa FCD was significantly higher after the third hour of fluid therapy as compared to the HHD group （P〈0.01）. No correlation between microhemodynamic parameters obtained from sublingual and ileal mucosas was found throughout the study. Conclusions： Prolonged HHD established by crystalloid solution significantly decreased ileal villus FCD when compared to restrictive fluid regimen. An inappropriate degree of HHD can be harmful during uncomplicated abdominal surgery.
