Transcriptome‑wide mapping of milk somatic cells upon subclinical mastitis infection in dairy cattle

Background Subclinical intramammary infection(IMI)represents a significant problem in maintaining dairy cows’*** severity and extent depend on the interaction between the causative agent,environment,and *** investigate the molecular mechanisms behind the host immune response,we used RnA-Seq for the milk somatic cells(SC)transcriptome profiling in healthy cows(n=9),and cows naturally affected by subclinical IMI from Proto-theca spp.(n=11)and Streptococcus agalactiae(***;n=11).Data Integration Analysis for Biomarker discov-ery using Latent Components(DIABLO)was used to integrate transcriptomic data and host phenotypic traits related to milk composition,SC composition,and udder health to identify hub variables for subclinical IMI *** A total of 1,682 and 2,427 differentially expressed genes(DEGs)were identified when comparing Prototheca *** *** to healthy animals,***-specific pathway analyses evidenced that Proto-theca’s infection upregulated antigen processing and lymphocyte proliferation pathways while *** induced a reduction of energy-related pathways like the tricarboxylic acid cycle,and carbohydrate and lipid *** integrative analysis of commonly shared DEGs between the two pathogens(n=681)referred to the core-mastitis response genes,and phenotypic data evidenced a strong covariation between those genes and the flow cytometry immune cells(r2=0.72),followed by the udder health(r2=0.64)and milk quality parameters(r2=0.64).Variables with r≥0.90 were used to build a network in which the top 20 hub variables were identified with the Cytoscape cyto-hubba *** genes in common between DIABLO and cytohubba(n=10)were submitted to a ROC analysis which showed they had excellent predictive performances in terms of discriminating healthy and mastitis-affected animals(sensitivity>0.89,specificity>0.81,accuracy>0.87,and precision>0.69).Among these genes,CIITA could play a key role in re

Genome-wide association study identifies QTL for eight fruit traits in cultivated tomato (Solanum lycopersicum L.)

Genome-wide association study(GWAS)is effective in identifying favorable alleles for traits of interest with high mapping resolution in crop *** this study,we conducted GWAS to explore quantitative trait loci(QTL)for eight fruit traits using 162 tomato accessions with diverse genetic *** eight traits included fruit weight,fruit width,fruit height,fruit shape index,pericarp thickness,locule number,fruit firmness,and *** variations of these traits in the tomato collection were evaluated with three replicates in field trials over three *** filtered 34,550 confident SnPs from the 51K Axiom®tomato array based onng data and>5%of minor allele frequency for association *** 162 tomato accessions were divided into seven clusters and their membership coefficients were used to account for population structure along with a kinship *** identify marker-trait associations(MTAs),four phenotypic data sets representing each of three years and combined were independently analyzed in the multilocus mixed model(MLMM).A total of 30 significant MTAs was detected over data sets for eight fruit traits at Pnumber of MTA per trait ranged from one(brix)to seven(fruit weight and fruit width).Two SnP markers on chromosomes 1 and 2 were significantly associated with multiple traits,suggesting pleiotropic effects of ***,16 of 30 MTAs suggest potential novel QTL for eight fruit *** results facilitate genetic dissection of tomato fruit traits and provide a useful resource to develop molecular tools for improving fruit traits via marker-assisted selection and genomic selection in tomato breeding programs.

Roger Ames＇s Confucian Role Ethics.＂ A Vocabulary, is an ambitious and significant exposition of Confucianism. Despite my disagreements with some of Ames＇s interpretation of Confucius, I have both enjoyed this book and learned a lot from it. Due to the limited scope of this review, I restrict it to an evaluation of Ames＇s exposition of the highest Confucian virtue, ren 4Y-. By Ames＇s own account, this book does not offer an alternative ethical theory, but a Confucian account of human flourishing, which begins with the roles and relations in the family. These roles and relations provide the basis of morality for Ames, as they not only constitute us but also enable us to become virtuous, respond appropriately in any situation, and enhance our relationships. In Ames＇s own words, we are ＂interrelated persons living a multiplicity of roles that constitute who we are, and ... allow us to pursue a unique distinctiveness and virtuosity in our conduct＂（122） and ＂Confucian role ethics takes the substance of morality to be nothing more or less than positive growth in the constitutive relations of any particular situation＂（258）.

An experimental study on tsunami inundation over complex coastal topography

In recent studies, the effects of complex costal topography on tsunami run-up has sparked heated discussion. This study mainly aims at investigating the effects of complex costal topography on the tsunami inundation distance and the effectiveness of sand dunes in dissipating tsunami wave energy. The experiments were carried out in a wave flume to investigate the potential reduction effects of wave run-up by non erodible sand dune like features. The results show that increasing dunes spacing could not significantly affect inundation distance. However, if the height of sand dunes is of the same order of magnitude as the incoming tsunami wave and the gaps between the dunes are large enough, successful tsunami mitigation could also be possible.

Diagnostic criteria for autoimmune hepatitis in children: A challenge for pediatric hepatologists

Autoimmune hepatitis (AIH) is a progressive inflammatory liver disorder that is rare in children and adolescents. AIH has a broad clinical spectrum and a quick response to treatment with corticosteroids and immunosuppressive medication. The available diagnosis criteria have limitations and should be evaluated in pediatric populations. Recently, some studies reported that the 2008 simplified diagnostic criteria for AIH could be used in children with high sensibility and specificity. In addition, the authors reported that globulin and immunoglobulin G levels can be used interchangeably for diagnostic purposes. They also demonstrated that the 2008 simplified criteria fail in identifying patients with fulminant hepatic failure. Here, we discuss the limitations of the use of these criteria in pediatric patients and the requirement of more studies to improve the diagnosis of AIH in children.

Compensating the impurities on the Cu surface by MOFs for enhanced hydrocarbon production in the electrochemical reduction of carbon dioxide

Copper(Cu)provides a cost-effective means of producing value-added fuels through the electrochemical reduction of carbon dioxide(CO_(2)RR).However,we observed the production of hydrocarbons via CO_(2)RR on commercial Cu films is less efficient because of the surface impurities,i.e.,*** monoxide(CO),a reaction intermediate of CO_(2)RR to hydrocarbons,binds strongly to the Fe sites and interrupts the production of hydrocarbons,resulting in an active hydrogen evolution reaction(HER).Herein,we report a method of blocking the effect of Fe impurities on the Cu surface through the preferential growth of nano-sized metal-organic frameworks(MOFs)on Fe *** zirconium(Zr)-based MOFs(UiO-66)forms a compensating layer on Cu film via the terephthalic acid(TPA)-Fe coordination bond,the Ui O-66 coated Cu film(UiO-66@Cu)presents significantly improved hydrocarbon Faradaic efficiency(FE)of 37.59%compared to 14.68%FE on commercial Cu film(99.9%purity)by suppressing *** to X-ray photoelectron spectroscopy(XPS)analysis,the UiO-66 ligand binds to entire metallic Fe site on the Cu surface,while metallic Cu is ***,UiO-66@Cu provides active sites of Cu for CO_(2)RR and leads to highly efficient and selective production of hydrocarbons.

Infectious diarrhea is a major cause of infant mortality in most developing countries. In this research, we evaluated the potential of immobilized Lactobacillus plantarum nCIMB 8826 on weanimix infant cereal and its effectiveness in reducing the severity of Citrobacter rodentium-induced diarrhea in weanling mice. Thirty-six C57BL/6 weanling mice were placed into four groups (n = 9 each;negative, positive, prevention and cure). Mice received either L. plantarum (109 CFU/g) immobilized on weanimix infant cereal 3 days before C. rodentium (109 CFU/ ml) infection (Prevention) or 3 days after C. rodentium infection (Cure). A positive control group was infected with C. rodentium only, while a nega-tive control group received neither L. plantarum nor C. rodentium. Positive control mice showed colonic mucosal and submucosal inflammation, erosion, and mucosal epithelia hyperplasia with the C. rodentium infection. Mice in the prevention and cure groups had less severe histologic alterations in the colon. Some beneficial effect of L. plantarum was observed in cecal short-chain fatty acid concentrations, which stimulates water and electrolytes absorption to reduce diarrhea. Our findings demonstrated that L. plantarum nCIMB 8826 could be immobilized on weanimix infant cereal to help reduce diarrhea during weaning.
