Protein kinase A inhibition induces EPAC-dependent acrosomal exocytosis in human sperm

To in teract with the egg, the spermatozo on must un dergo several biochemical and motility modifications in the female reproductive tract, collectively called capacitation. Only capacitated sperm can undergo acrosomal exocytosis, near or on the egg, a process that allows the sperm to penetrate and fertilize the egg. In the present study, we investigated the invoIvement of cyclic adenosine monophosphate (cAMP)-dependent processes on acrosomal exocytosis. Inhibition of protein kinase A (PKA) at the end of capacitation induced acrosomal exocytosis. This process is cAMP-dependent;however, the addition of relatively high concentration of the membrane-permeable 8-bromo-cAMP (8Br-cAMP, 0.1 mmol l^-1) analog induced significant inhibition of the acrosomal exocytosis. The induction of acrosomal exocytosis by PKA inhibition was significantly inhibited by an exchange protein directly activated by cAMP (EPAC) ESI09 inhibitor. The EPAC selective substrate activated AE at relatively low concentrations (0.02-0.1 μmol l^1), whereas higher concerttrations (>5 pmol l^-1) were inhibitory to the AE induced by PKA inhibition. Inhibition of PKA revealed about 50% increase in intracellular cAMP levels, conditions under which EPAC can be activated to induce the AE. Induction of AE by activating the actin severing?protein, gelsolin, which causes F-actin dispersion, was inhibited by the EPAC inhibitor. The AE induced by PKA inhibition was mediated by phospholipase C activity but not by the Ca^2+-channel, CatSper. Thus, inhibition of PKA at the end of the capacitation process induced EPAC/phospholipase C-dependent acrosomal exocytosis. EPAC mediates F-actin depolymerization an d/or activation of effectors down stream to F-actin breakdown that lead to acrosomal exocytosis.

A texture-modified dessert with high nutritional value designed for people with dysphagia:effect of refrigeration and frozen storage

Dysphagia is commonly associated with malnutrition and an increased choking risk.To overcome these complications,food designed for people with dysphagia should have an appropriate texture and a high nutritional value.In this study,six formulations of a strawberry dessert enriched in protein(calcium caseinate)and fiber(wheat dextrin or inulin)were developed using different hydrocolloids(xanthan gum,carboxymethyl cellulose or modified starch)to provide desirable texture and stability.Nutritional value was calculated and total phenolic content and antioxidant activity of the samples were analyzed.Back-extrusion test,rheological measurements and sensory analysis were performed in refrigerated and frozen samples to characterize their textural and viscoelastic properties.The high content in protein(14.7 g/100 g)and fiber(7.9-8.7 g/100 g)made possible to use the claims“high protein”and“high fiber”.Phytochemicals supplied by strawberries contributed to the antioxidant properties of the dessert.Loss tangent ranged 0.28-0.35 for all the formulations,indicating a weak gel behavior,which could be considered safe to swallow.The formulations with dextrin in combination with carboxymethyl cellulose or xanthan gum seemed to be less susceptible to structural changes during frozen storage.This work provides insights for the development of a nutrient-dense dessert that meets the requirements of people with dysphagia.

Beneficial use of mussel shell as a bioadditive for TPU green composites by the valorization of an aqueous waste

Scientific studies have focused on environmentally friendly solutions as effective as the reuse of crop products owing to plastic-waste problems in recent years.This issue is the main driving force for upcoming academic research attempts in waste valorization-related studies.Herein,we integrated an aqua-waste,mussel shell(MS),as a bioadditive form into green thermoplastic polyurethane(TPU)green composites.Tuning of the MS surface was performed to achieve strong adhesion between composite phases.The surface functionalities of MS powders were evaluated via infrared spectroscopy and scanning electron microscopy(SEM)images.Composite samples were prepared by melt-compounding followed by injection molding techniques.It was confirmed by morphological analysis that relatively better adhesion between the phases was achieved for composites involving surface-modified MS compared to unmodified MS.Tensile strength and Young’s modulus of surface-modified MS-filled composites were found to be higher than those of unmodified MS,whereas the elongation at break shifted to lower values with MS inclusions.The shore hardness of TPU was remarkably improved after being incorporated with silane-treated MS(AS-MS).Stearic acid-treated MS(ST-MS)additions resulted in an enhancement in the thermal stability of the composites.Thermo-mechanical analysis showed that the storage moduli of composites were higher than those of unfilled TPU.ST-MS additions led to an increase in the characteristic glass transition temperature of TPU.Melt flow index(MFI)of neat TPU was highly improved after MS loading regardless of modification type.According to the wear test,surface modification of MS displayed a positive effect on the wear resistance of TPU.As the water absorption data of the composites were evaluated,the TPU/AS-MS composite yielded the lowest water absorption.The silane layer on MS inclusion promoted water repellency of composites due to the hydrophobicity of silane.The results of the biodegradation investigation demonstrated that adding unmodifie

Diagnostic challenges and individualized treatment of cervical adenocarcinoma metastases to the breast:A case report

BACKGROUND Cervical cancer is a rare primary tumor resulting in metastases to the breast with few cases reported in literature.Breast metastases are associated with poor prognosis.The following case highlights the diagnostic challenges associated with metastatic cervical cancer to the breast along with individualized treatment.CASE SUMMARY A 44-year-old G7P5025 with no significant past medical or surgical history presented with heavy vaginal to an outside emergency department where an exam and a pelvic magnetic resonance imaging showed a 4.5 cm heterogenous lobulated cervical mass involving upper two thirds of vagina,parametria and lymph node metastases.Cervical biopsies confirmed high grade adenocarcinoma with mucinous features.A positron emission tomography/computed tomography(PET/CT)did not show evidence of metastatic disease.She received concurrent cisplatin with external beam radiation therapy.Follow up PET/CT scan three months later showed no suspicious fluorodeoxyglucose uptake in the cervix and no evidence of metastatic disease.Patient was lost to follow up for six months.She was re-imaged on re-presentation and found to have widely metastatic disease including breast disease.Breast biopsy confirmed programmed death-ligand 1 positive metastatic cervical cancer.The patient received six cycles of carboplatin and paclitaxel with pembrolizumab.Restaging imaging demonstrated response.Patient continued on pembrolizumab with disease control.CONCLUSION Metastatic cervical cancer to the breast is uncommon with nonspecific clinical findings that can make diagnosis challenging.Clinical history and immunohistochemical evaluation of breast lesion,and comparison to primary tumor can support diagnosis of metastatic cervical cancer to the breast.Overall,the prognosis is poor,but immunotherapy can be considered in select patients and may result in good disease response.

In this paper we study a bilinear optimal control problem for a diffusive Lotka-Volterra competition model with chemo-repulsion in a bounded domain of ℝ^(ℕ),N=2,3.This model describes the competition of two species in which one of them avoid encounters with rivals through a chemo-repulsion mechanism.We prove the existence and uniqueness of weak-strong solutions,and then we analyze the existence of a global optimal solution for a related bilinear optimal control problem,where the control is acting on the chemical signal.Posteriorly,we derive first-order optimality conditions for local optimal solutions using the Lagrange multipliers theory.Finally,we propose a discrete approximation scheme of the optimality system based on the gradient method,which is validated with some computational experiments.

Japanese candlestick charts for diabetes

Continuous glucose monitoring(CGM)is a popular technology among the diabetic population,especially in patients with type 1 diabetes and those with type 2 diabetes treated with insulin.The American Diabetes Association recommends standardization of CGM reports with visual cues,such as the ambulatory glucose profile.Nevertheless,interpreting this report requires training and time for CGM to be cost-efficient.In this work it has been proposed to incorporate Japanese candlestick charts in glucose monitoring.These graphs are used in price analysis in financial markets and are easier to view.Each candle provides extra information to make prudent decisions since it reports the opening,maximum,minimum and closing glucose levels of the chosen time frame,usually the daily one.The Japanese candlestick chart is an interesting tool to be considered in glucose control.This graphic representation allows identification of glucose trends easily through the colors of the candles and maximum and minimum glucose values.

Facial emotion recognition(FER)has become a focal point of research due to its widespread applications,ranging from human-computer interaction to affective computing.While traditional FER techniques have relied on handcrafted features and classification models trained on image or video datasets,recent strides in artificial intelligence and deep learning(DL)have ushered in more sophisticated approaches.The research aims to develop a FER system using a Faster Region Convolutional Neural Network(FRCNN)and design a specialized FRCNN architecture tailored for facial emotion recognition,leveraging its ability to capture spatial hierarchies within localized regions of facial features.The proposed work enhances the accuracy and efficiency of facial emotion recognition.The proposed work comprises twomajor key components:Inception V3-based feature extraction and FRCNN-based emotion categorization.Extensive experimentation on Kaggle datasets validates the effectiveness of the proposed strategy,showcasing the FRCNN approach’s resilience and accuracy in identifying and categorizing facial expressions.The model’s overall performance metrics are compelling,with an accuracy of 98.4%,precision of 97.2%,and recall of 96.31%.This work introduces a perceptive deep learning-based FER method,contributing to the evolving landscape of emotion recognition technologies.The high accuracy and resilience demonstrated by the FRCNN approach underscore its potential for real-world applications.This research advances the field of FER and presents a compelling case for the practicality and efficacy of deep learning models in automating the understanding of facial emotions.

NIBAN1 overexpression has been reported in a widerange of thyroid carcinoma subtypes,and in other cancers,while it is not expressed in thyroid benign lesions and normal thy-roid.1-3 However,the mechanism associated with its expression and prognostic value remains unclear.In this study,six independent cohorts were enrolled,in which NIBAN1 expression was evaluated.A total of 583patients with thyroid tumors,326 patients with skin cancers,293 patients with colorectal carcinoma,and 189 patients with lung carcinoma were analyzed.

A prototype for on-site generation of chlorinated disinfectant for use in rural aqueducts

Sodium hypochlorite has significant potential as a sanitation solution in hard-to-reach areas.Few studies have investigated the optimal electrolysis parameters for its production with volumes greater than 1o L.This study evaluated sodium hypochlorite production through electrolysis in a 22-L prototype and identified the optimal operating parameters.Tests were performed using graphite electrodes with areas of 68.4 cm²at the laboratory scale and 1865.0 cm?at the prototype scale.A design for experiments with different operating times,chloride concentrations,and electric current intensities was developed.The optimal operating time,sodium chloride concentration,and current intensity at the laboratory scale were 120 min,150 g of chloride per liter,and 3 A,respectively,leading to the production of 5.02 g/L of the disinfectant with an energy efficiency of 12.21 mg of Cl2 per kilojoule.At the prototype scale,the maximum sodium hypochlorite concentration of 3.99 g of chloride per liter was achieved with an operating time of 120 min,a sodium chloride concentration of 100 g of chloride per liter,and a current intensity of 70 A,reaching an energy efficiency of 42.56 mg of Cl2 per kilojoule.In addition,this study evaluated the influences of the chloride concentration,current intensity,and operating time on the production of sodium hypochlorite at the two scales,and formulated the equations showing the trends of sodium hypochlorite production and energy efficiency in the electrochemical systems.The 22-L prototype model for production of this oxidizing substance is promising for disinfection of large volumes of water in areas that are difficult to access.

Isolated Third Nerve Cranial Palsy: Complication of Brain Aneurysm

This case report highlights a critical and emergent condition, isolated third nerve cranial palsy due to a brain aneurysm. It emphasizes the importance of differential diagnosis and attentiveness to the physical exam in an emergency setting. The detailed progression from initial symptoms and misinterpretation to final diagnosis offers valuable insight into the dangers of overlooking critical diagnoses. A bilobed intracranial aneurysm arising from the internal carotid artery (ICA) caused symptoms in this patient that could have been attributed to a less malignant etiology, such as a complex migraine. Overlooking the urgency of the situation and missing the diagnosis could have had a grave and irreversible outcome.
