Background:Liver cancer is a malignancy with high morbidity and mortality *** family E member 2(SERPINE2)has been reported to play a key role in the metastasis of many *** this study,we aimed to investigate the potential mechanism of SERPINE2 in liver cancer ***:The Cancer Genome Atlas database(TCGA),including DNA methy-lation and transcriptome sequencing data,was utilized to identify the crucial oncogene associated with DNA methylation and cancer progression in liver *** from the TCGA and RNA sequencing for 94 pairs of liver cancer tissues were used to explore the correlation between SERPINE2 expression and clin-ical parameters of *** methylation sequencing was used to detect the DNA methylation levels in liver cancer tissues and *** sequencing,cytokine assays,immunoprecipitation(IP)and mass spectrometry(MS)assays,protein stability assays,and ubiquitination assays were performed to explore the regulatory mechanism of SERPINE2 in liver cancer ***-derived xenografts and tumor organoid models were established to determine the role of SERPINE2 in the treatment of liver cancer using ***:Based on the public database screening,SERPINE2 was identified as a tumor promoter regulated by DNA ***2 expression was significantly higher in liver cancer tissues and was associated with the dismal prognosis in patients with liver ***2 promoted liver cancer metas-tasis by enhancing cell pseudopodia formation,cell adhesion,cancer-associated fibroblast activation,extracellular matrix remodeling,and ***/MS assays confirmed that SERPINE2 activated epidermal growth factor receptor(EGFR)and its downstream signaling pathways by interacting with ***,SERPINE2 inhibited EGFR ubiquitination and maintained its protein stability by competing with the E3 ubiquitin ligase,***,EGFR was activated in liver cancer cells after sorafenib treatment,and SER

Therapeutic utility of human umbilical cord-derived mesenchymal stem cells-based approaches in pulmonary diseases:Recent advancements and prospects

Pulmonary diseases across all ages threaten millions of people and have emerged as one of the major public health issues *** diverse disease con-ditions,the currently available approaches are focused on alleviating clinical symptoms and delaying disease progression but have not shown significant therapeutic effects in patients with lung *** umbilical cord-derived mesenchymal stem cells(UC-MSCs)isolated from the human UC have the capacity for self-renewal and multilineage ***,in recent years,these cells have been demonstrated to have unique advantages in the treatment of lung *** searched the Public Clinical Trial Database and found 55 clinical trials involving UC-MSC therapy for pulmonary diseases,including coronavirus disease 2019,acute respiratory distress syndrome,bron-chopulmonary dysplasia,chronic obstructive pulmonary disease,and pulmonary *** this review,we summarize the characteristics of these registered clinical trials and relevant published results and explore in depth the challenges and opportunitiesfaced in clinical ***,the underlying mole-cular mechanisms involved in UC-MSC-based therapy for pulmonary diseases are also analyzed in *** brief,this comprehensive review and detailed analysis of these clinical trials can be expected to provide a scientific reference for future large-scale clinical application.

External transient impact loads widely exist in the service environment of various engineering structures,and urgent engineering and military application requirements call for research on the theory and method of impact *** manuscript proposes a method to identify the impact *** employing the dispersive property of wave propagation through the waveguides,a dispersion-based average power spectral density(PSD)curve for quantitative representation of the dispersion is established,and its evolution law is investigated to apply the identification of impact *** critical parameters of the average PSD curve are obtained by analyzing the response of the structure from the perspective of *** effectiveness of this method is verified through the development of the triple-successive-impact split Hopkinson bar and the triple-successive-impact *** the method proposed in this manuscript,a kind of impact identification under multiple impact loads,which is not convenient to measure directly by the sensor,is realized,especially in the case of highly transient external impact.

An anion-rich electric double layer(EDL)region is favorable for fabricating an inorganic-rich solid-electrolyte interphase(SEI)towards stable lithium metal anode in ester ***,cetyltrimethylammonium bromide(CTAB),a cationic surfactant,is adopted to draw more anions into EDL by ionic interactions that shield the repelling force on anions during lithium *** situ electrochemical surface-enhanced Raman spectroscopy results combined with molecular dynamics simulations validate the enrichment of NO_(3)^(−)/FSI−anions in the EDL region due to the positively charged CTA^(+).In-depth analysis of SEI structure by X-ray photoelectron spectroscopy and time-of-flight secondary ion mass spectrometry results confirmed the formation of the inorganic-rich SEI,which helps improve the kinetics of Li^(+)transfer,lower the charge transfer activation energy,and homogenize Li *** a result,the Li||Li symmetric cell in the designed electrolyte displays a prolongated cycling time from 500 to 1300 h compared to that in the blank electrolyte at 0.5 mA cm^(-2) with a capacity of 1 mAh cm^(-2).Moreover,Li||LiFePO_(4) and Li||LiCoO_(2) with a high cathode mass loading of>10 mg cm^(-2) can be stably cycled over 180 cycles.

目的从食品销售环境样本中分离李斯特菌噬菌体,并对分离获得的噬菌体进行电镜形态观察、宿主谱及生物学特征分析。方法采用双层琼脂平板法和点滴法,以分离的英诺克李斯特菌Lin08作为宿主菌,成功分离并鉴定出一株烈性噬菌体LMLPA5,使用透射电镜观察噬菌体形态,并测定噬菌体的宿主谱、最佳感染复数(MOI)、一步生长曲线以及理化稳定性,同时探索噬菌体在4℃、-20℃及-80℃下的保存效果。结果分离获得的LMLPA5噬菌体属于肌尾噬菌体科,其形成的噬菌斑清晰透明,周围无晕环。该噬菌体为广谱的李斯特菌烈性噬菌体,能裂解多个李斯特菌种及单增李斯特菌多个血清型菌株。LMLPA5的最佳MOI为0.1,潜伏期为10 min,平均裂解量为95.2 PFU/cell。LMLPA5对高温较敏感,在70℃暴露1 h即完全失活,而在4℃~40℃作用32 h以上噬菌体仍能保持稳定。在pH为4~10 LMLPA5的活性不受影响,经紫外线照射60 min后噬菌体被完全灭活。LMLPA5对氯仿不敏感,为无囊膜型噬菌体。噬菌体在4℃及-80℃的保存效果较好,可稳定保存8个月以上。结论本研究获得的1株广谱李斯特菌肌尾噬菌体LMLPA5具有较强的裂解能力、抵抗力和稳定性,为进一步的应用提供了基础。
