Washed microbiota transplantation for Crohn’s disease:A metagenomic,metatranscriptomic,and metabolomic-based study

BACKGROUND Fecal microbiota transplantation(FMT)is a promising therapeutic approach for treating Crohn’s disease(CD).The new method of FMT,based on the automatic washing process,was named as washed microbiota transplantation(WMT).Most existing studies have focused on observing the clinical phenomena.However,the mechanism of action of FMT for the effective management of CD-particularly in-depth multi-omics analysis involving the metagenome,metatranscriptome,and metabolome-has not yet been reported.AIM To assess the efficacy of WMT for CD and explore alterations in the microbiome and metabolome in response to WMT.METHODS We conducted a prospective,open-label,single-center clinical study.Eleven CD patients underwent WMT.Their clinical responses(defined as a decrease in their CD Activity Index score of>100 points)and their microbiome(metagenome,metatranscriptome)and metabolome profiles were evaluated three months after the procedure.RESULTS Seven of the 11 patients(63.6%)showed an optimal clinical response three months post-WMT.Gut microbiome diversity significantly increased after WMT,consistent with improved clinical symptoms.Comparison of the metagenome and metatranscriptome analyses revealed consistent alterations in certain strains,such as Faecalibac-terium prausnitzii,Roseburia intestinalis,and Escherichia coli.In addition,metabolomics analyses demonstrated that CD patients had elevated levels of various amino acids before treatment compared to the donors.However,levels of vital amino acids that may be associated with disease progression(e.g.,L-glutamic acid,gamma-glutamyl-leucine,and prolyl-glutamine)were reduced after WMT.CONCLUSION WMT demonstrated therapeutic efficacy in CD treatment,likely due to the effective reconstruction of the patient’s microbiome.Multi-omics techniques can effectively help decipher the potential mechanisms of WMT in treating CD.

Although e-learning is increasingly accepted by China people as an important learning style, the e-learning resource configuration and distribution in China villages and small towns is not as good as China cities, therefore, children from villages and small towns don’t have enough chances to use e-learning resource. To solve this problem, a project named "villages’ e-learning resource configuration ＆ distribution management" was started by China government, which is focusing on promoting the application quality of e-learning resource in villages and small towns. As the researchers of this project, authors of this paper designed and implemented an e-learning resource distribution and management system based on e-learning standards, and applied the system into practical environment.

BACKGROUND Adolescent major depressive disorder(MDD)is a significant mental health concern that often leads to recurrent depression in adulthood.Resting-state functional magnetic resonance imaging(rs-fMRI)offers unique insights into the neural mechanisms underlying this condition.However,despite previous research,the specific vulnerable brain regions affected in adolescent MDD patients have not been fully elucidated.AIM To identify consistent vulnerable brain regions in adolescent MDD patients using rs-fMRI and activation likelihood estimation(ALE)meta-analysis.METHODS We performed a comprehensive literature search through July 12,2023,for studies investigating brain functional changes in adolescent MDD patients.We utilized regional homogeneity(ReHo),amplitude of low-frequency fluctuations(ALFF)and fractional ALFF(fALFF)analyses.We compared the regions of aberrant spontaneous neural activity in adolescents with MDD vs healthy controls(HCs)using ALE.RESULTS Ten studies(369 adolescent MDD patients and 313 HCs)were included.Combining the ReHo and ALFF/fALFF data,the results revealed that the activity in the right cuneus and left precuneus was lower in the adolescent MDD patients than in the HCs(voxel size:648 mm3,Pding of the neuropathology of affected adolescents.

本文通过在碳纳米管（ MWNTs ）表面原位自发还原形成普鲁士蓝纳米颗粒（ PB ），制备 PB 包覆的MWNTs纳米复合物（ PB@MWNTs），并将其修饰于电极表面，直接催化过氧化氢，成功制得了一种无酶过氧化氢传感器。由于PB和MWNTs对过氧化氢都有较好的催化能力，使得制备的传感器具有好的性能。该传感器在H2O2浓度为1．8x10-7mol.L-1-4．2x215;10-3mol.L-1范围内，呈良好的线性，检测下限达到8．0x10-8mol.L-1（S/N=3），灵敏度为71．8mol.L/mol/cm2。此外，传感器还表现出较好的稳定性、重现性和选择性。

BACKGROUND Radiotherapy stands as a promising therapeutic modality for colorectal cancer(CRC);yet,the formidable challenge posed by radio-resistance significantly undermines its efficacy in achieving CRC remission.AIM To elucidate the role played by microRNA-298(miR-298)in CRC radio-resistance.METHODS To establish a radio-resistant CRC cell line,HT-29 cells underwent exposure to 5 gray ionizing radiation that was followed by a 7-d recovery period.The quantification of miR-298 levels within CRC cells was conducted through quantitative RT-PCR,and protein expression determination was realized through Western blotting.Cell viability was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and proliferation by clonogenic assay.Radio-induced apoptosis was discerned through flow cytometry analysis.RESULTS We observed a marked upregulation of miR-298 in radio-resistant CRC cells.MiR-298 emerged as a key determinant of cell survival following radiation exposure,as its overexpression led to a notable reduction in radiation-induced apoptosis.Intriguingly,miR-298 expression exhibited a strong correlation with CRC cell viability.Further investigation unveiled human dual-specificity tyrosine(Y)-regulated kinase 1A(DYRK1A)as miR-298’s direct target.CONCLUSION Taken together,our findings underline the role played by miR-298 in bolstering radio-resistance in CRC cells by means of DYRK1A downregulation,thereby positioning miR-298 as a promising candidate for mitigating radioresistance in CRC.

目的:检测心肌细胞缺血再灌注(IR)过程中热休克蛋白70(HSP70)的表达,探讨多巴胺对HSP70表达的影响。方法:不同时间阻断W istar大鼠冠状动脉的前降支,用含多巴胺或生理盐水的灌注液再灌注5 h后,取出心脏前降支支配区的全层心肌组织;用W esten B lot方法检测HSP70的表达。结果:心肌细胞的HSP70表达随缺血时间的延长而增加,缺血30 m in时表达最高,其后逐渐减少;灌注液含多巴胺的实验组较相同缺血时间的灌注生理盐水的对照组HSP70表达均有明显增加(P<0.05)。结论:心肌HSP70表达与心肌缺血相关,且呈时间依赖性,最大缺血耐受时间为30 m in左右。多巴胺在心肌缺血再灌注过程中促进HSP70的表达。

Premature ovarian insufficiency(POI)is a heterogeneous female disorder characterized by the loss of ovarian function before the age of 40.It represents a significant detriment to female fertility.However,the known POI-causative genes currently account for only a fraction of cases.To elucidate the genetic factors underlying POI,we conducted whole-exome sequencing on a family with three fertile POI patients and identified a deleterious missense variant in RNF111.In a subsequent replication study involving 1,030 POI patients,this variant was not only confirmed but also accompanied by the discovery of three additional predicted deleterious RNF111 variants.These variants collectively account for eight cases,representing 0.78%of the study cohort.A further study involving 500 patients with diminished ovarian reserve also identified two additional RNF111 variants.Notably,RNF111 encodes an E3 ubiquitin ligase with a regulatory role in the TGF-β/BMP signaling pathway.Our analysis revealed that RNF111/RNF111 is predominantly expressed in the oocytes of mice,monkeys,and humans.To further investigate the functional implications of RNF111 variants,we generated two mouse models:one with a heterozygous missense mutation(Rnf111+/M)and another with a heterozygous null mutation(Rnf111^(+/-)).Both mouse models exhibited impaired female fertility,characterized by reduced litter sizes and small ovarian reserve.Additionally,RNA-seq and quantitative proteomics analysis unveiled that Rnf111 haploinsufficiency led to dysregulation in female gonad development and negative regulation of the BMP signaling pathway within mouse ovaries.In conclusion,our findings strongly suggest that monoallelic deleterious variants in RNF111 can impair female fertility and induce POI in both humans and mice.

基于联想记忆神经网络模型的BP算法

针对在加筋土挡墙设计中,一般不考虑地基变形影响,而软土地基过大变形会造成加筋土挡墙失效的问题,结合某加筋土挡墙实例,利用数值模拟方法建立了软土地基上加筋土挡墙数值模型,分析了软土地基不均匀变形对加筋土挡墙的影响。结果表明:由于加筋土挡墙基底压力的偏心作用,会引起软土地基的差异沉降,对墙体侧向变形及向临空面倾覆有不利影响;加筋土挡墙面板变形主要是由地基不均匀沉降引起的;加筋土挡墙地基的沉降曲线近似线性,面板下地基沉降量最大。参数分析还揭示,地基土的杨氏模量和面板仰角、回填土特性等对加筋土挡墙变形特征均存在较大影响。
