Background Our recent pilot study suggests intra-arterial tenecteplase(TNK)during the first pass of endovascular treatment(EVT)seems safe,may increase first-pass reperfusion and good outcome in acute ischaemic stroke(AIS)patients with large-vessel occlusion(LVO).Aims To determine the efficacy and safety of intra-arterial TNK administration during EVT in AIS-LVO patients presenting up to 24 hours from symptom onset.Sample size estimates A maximum of 380 patients are required to test the superiority hypothesis with 80%power according to a two-side 0.05 level of significance,stratified by age,gender,baseline systolic blood pressure,prestroke modified Rankin Scale(mRS),baseline National Institute of Health stroke scale,baseline ASPECTS,time from onset to groin puncture,intravenous thrombolysis before EVT,stroke territory and stroke aetiology.Design Intra-arterial TNK during thrombectomy for acute stroke(BRETIS-TNK II)study is a prospective,randomised,adaptive enrichment,open-label,blinded end point,multicentre study.Eligible AIS-LVO patients are randomly assigned into the experimental group and control group with a ratio of 1:1.The experimental group will be treated with intra-arterial infusion of TNK during EVT.The control group will be treated with standard EVT.Outcome The primary end point is a favourable outcome,defined as an mRS score of 0–2 at 90 days.The primary safety end point is symptomatic intracranial haemorrhage within 48 hours,which is defined as an increase in the National Institutes of Health Stroke Scale score of≥4 points as a result of the intracranial haemorrhage.Conclusions The results of BRETIS-TNK II will provide evidence for the efficacy and safety of intra-arterial TNK administration during EVT in AIS patients with LVO.

Optimization design of autofocusing metasurface for ultrasound wave application

In this paper,two optimized autofocusing metasurfaces(AFMs)with different desired focal distances are designed by using particle swarm optimization(PSO)algorithm.Based on the ffnite element simulation software COMSOL Multiphysics,the performance of ultrasound transducer(UT)with AFM at different design parameters in Airy distributions(r0,ω)and the bottom thickness(d)of AFM are simulated and analyzed.Based on the simulation data,the artiffcial neural network model is trained to describe the complex relationship between the design parameters of AFM and the performance parameters of UT.Then,the multiobjective optimization function for AFM is determined according to the desired performance parameters of UT,including focal position,lateral resolution,longitudinal resolution and absolute sound pressure.In order to obtain AFMs with the desired performance,PSO algorithm is adopted to optimize the design parameters of AFM according to the multiobjective optimization function,and two AFMs are optimized and fabricated.The experimental results well agree with the simulation and optimization results,and the optimized AFMs can achieve the desired performance.The fabricated AFM can be easily integrated with UT,which has great potential applications in wave ffeld modulation underwater,acoustic tweezers,biomedical imaging,industrial nondestructive testing and neural regulation.

The Submillimeter Wave Astronomy Satellite（SWAS） was the first space telescope capable of high spectral resolution observations of terahertz spectral lines. We have investigated the integration ability of its two receivers and spectrometer during five and a half years of on-orbit operation. The CI, O2, H2O, and13CO spectra taken toward all observed Galactic sources were analyzed. The present results are based on spectra with a total integration time of up to 2.72 × 10～4hr（■10～8s）. The noise in the spectra is generally consistent with that expected from the radiometer equation, without any sign of approaching a noise floor. This noise performance reflects the extremely stable performance of the passively cooled front end as well as other relevant components in the SWAS instrument throughout its mission lifetime.

Optical Extinctions of Inter-Arm Molecular Clouds in M31: A Pilot Study for the Upcoming CSST Observations

Recent submillimeter dust thermal emission observations have unveiled a significant number of inter-arm massive molecular clouds in M31. However, the effectiveness of this technique is limited to its sensitivity, making it challenging to study more distant galaxies. This study introduces an alternative approach, utilizing optical extinctions derived from space-based telescopes, with a focus on the forthcoming China Space Station Telescope（CSST）. We first demonstrate the capability of this method by constructing dust extinction maps for 17 inter-arm massive molecular clouds in M31 using the Panchromatic Hubble Andromeda Treasury data. Our analysis reveals that inter-arm massive molecular clouds with an optical extinction（AV） greater than 1.6 mag exhibit a notable AVexcess, facilitating their identification. The majority of these inter-arm massive molecular clouds show an AVaround 1 mag, aligning with measurements from our JCMT data. Further validation using a mock CSST RGB star catalog confirms the method's effectiveness. We show that the derived AVvalues using CSST z and y photometries align more closely with the input values. Molecular clouds with AV> 1.6 mag can also be identified using the CSST mock data. We thus claim that future CSST observation clouds provide an effective way for the detection of inter-arm massive molecular clouds with significant optical extinction in nearby galaxies.

SFGA-CPA: A Novel Screening Correlation Power Analysis Framework Based on Genetic Algorithm

Correlation power analysis(CPA)combined with genetic algorithms(GA)now achieves greater attack efficiency and can recover all subkeys simultaneously.However,two issues in GA-based CPA still need to be addressed:key degeneration and slow evolution within populations.These challenges significantly hinder key recovery efforts.This paper proposes a screening correlation power analysis framework combined with a genetic algorithm,named SFGA-CPA,to address these issues.SFGA-CPA introduces three operations designed to exploit CPA characteris-tics:propagative operation,constrained crossover,and constrained mutation.Firstly,the propagative operation accelerates population evolution by maximizing the number of correct bytes in each individual.Secondly,the constrained crossover and mutation operations effectively address key degeneration by preventing the compromise of correct bytes.Finally,an intelligent search method is proposed to identify optimal parameters,further improving attack efficiency.Experiments were conducted on both simulated environments and real power traces collected from the SAKURA-G platform.In the case of simulation,SFGA-CPA reduces the number of traces by 27.3%and 60%compared to CPA based on multiple screening methods(MS-CPA)and CPA based on simple GA method(SGA-CPA)when the success rate reaches 90%.Moreover,real experimental results on the SAKURA-G platform demonstrate that our approach outperforms other methods.

BACKGROUND B56εis a regulatory subunit of the serine/threonine protein phosphatase 2A,which is abnormally expressed in tumors and regulates various tumor cell functions.At present,the application of B56εin pan-cancer lacks a comprehensive analysis,and its role and mechanism in hepatocellular carcinoma(HCC)are still unclear.The Cancer Genome Atlas,Genotype-Tissue Expression,Gene Expression Profiling Interactive Analysis,and Tumor Immune Estimation Resource databases were used to analyze B56εexpression,prognostic mutations,somatic copy number alterations,and tumor immune characteristics in 33 tumors.The relationships between B56εexpression levels and drug sensitivity,immuno-therapy,immune checkpoints,and human leukocyte antigen(HLA)-related genes were further analyzed.Gene Set Enrichment Analysis(GSEA)was performed to reveal the role of B56εin HCC.The Cell Counting Kit-8,plate cloning,wound healing,and transwell assays were conducted to assess the effects of B56εinterference on the malignant behavior of HCC cells.RESULTS In most tumors,B56εexpression was upregulated,and high B56εexpression was a risk factor for adrenocortical cancer,HCC,pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma(all Pdiated signaling in HCC.Knockdown of B56εexpression in HCC cells inhibited the proliferation,migration,and invasion capacity of tumor cells.Core Tip:The expression of protein phosphatase 2A(PP2A)subunit B56εis up-regulated in most tumors,and its high expression is a risk factor for adrenocortical c

OsPDR20 is an ABCG metal transporter regulating cadmium accumulation in rice

Cadmium(Cd)is a non-essential toxic heavy metal,seriously posing high environmental risks to human health.digging genetic resources relevant to functional genes is important for understanding the metal absorption and accumulation in crops and bioremediation of Cd-polluted environments.This study investigated a functionally uncharacterized ATP binding cassette transporter G family(ABCG)gene encoding a Pleiotropic Drug Resistance 20(PDR20)type metal transporter which is localized to the plasma membrane of rice.OsPDR20 was transcriptionally expressed in almost all tissues and organs in lifespan and was strongly induced in roots and shoots of young rice under Cd stress.Ectopic expression of OsPDR20 in a yeast mutant ycf1 sensitive to Cd conferred cellular tolerance with less Cd accumulation.Knockdown of OsPDR20 by RNA interference(RNAi)moderately attenuated root/shoot elongation and biomass,with reduced chlorophylls in rice grown under hydroponic medium with 2 and 10μmol/L Cd,but led to more Cd accumulation.A field trial of rice grown in a realistic Cd-contaminated soil(0.40 mg/kg)showed that RNAi plants growth and development were also compromised compared to wild-type(WT),with smaller panicles and lower spikelet fertility but little effect on yield of grains.However,OsPDR20 suppression resulted in unexpectedly higher levels of Cd accumulation in rice straw including lower leaves and culm and grain.These results suggest that OsPDR20 is actively involved in Cd accumulation and homeostasis in rice crops.The increased Cd accumulation in the RNAi plants has the potential application in phytoremediation of Cd-polluted wetland soils.

High spatiotemporal resolution radiances from the advanced imagers onboard the new generation of geostationary weather satellites provide a unique opportunity to evaluate the abilities of various reanalysis datasets to depict multilayer tropospheric water vapor(WV),thereby enhancing our understanding of the deficiencies of WV in reanalysis datasets.Based on daily measurements from the Advanced Himawari Imager(AHI)onboard the Himawari-8 satellite in 2016,the bias features of multilayer WV from six reanalysis datasets over East Asia are thoroughly evaluated.The assessments show that wet biases exist in the upper troposphere in all six reanalysis datasets;in particular,these biases are much larger in summer.Overall,we find better depictions of WV in the middle troposphere than in the upper troposphere.The accuracy of WV in the ERA5 dataset is the highest,in terms of the bias magnitude,dispersion,and pattern similarity.The characteristics of the WV bias over the Tibetan Plateau are significantly different from those over other parts of East Asia.In addition,the reanalysis datasets all capture the shift of the subtropical high very well,with ERA5 performing better overall.

We present the interstellar scintillation analysis of fast radio burst(FRB)20220912A during its extremely active episode in 2022using data from the Five-hundred-meter Aperture Spherical Radio Telescope(FAST).We detect a scintillation arc in the FRB’s secondary spectrum,which describes the power in terms of the scattered FRB signals’time delay and Doppler shift.The arc indicates that the scintillation is caused by a highly localized region.Our analysis favors a Milky Way origin of the ionized interstellar medium(IISM)for the localized scattering medium but cannot rule out a host galaxy origin.We present our method for detecting the scintillation arc,which can be applied generally to sources with irregularly spaced bursts or pulses.These methods could help shed light on the complex interstellar environment surrounding the FRBs and in our Galaxy.

Long-term complete response to anti-programmed-death-1 monotherapy in a patient with relapsed and refractory ovarian adenocarcinoma: A case report

BACKGROUND Ovarian cancer is the most common malignant tumor of the female reproductive system,and the survival rate of patients with relapsed and refractory ovarian cancer is very low.CASE SUMMARY Here,we report a case of high-grade serous papillary adenocarcinoma of the ovary that was successfully treated with immunotherapy.Radical surgery and adjuvant chemotherapy for the 56-year-old patient were successful;however,her tumor relapsed.Subsequent second-line chemotherapy,targeted agents,and other treatments were ineffective,as the tumor continued to recur and metastasize.Anti-programmed cell death-1(PD-1)monotherapy(tislelizumab)completely alleviated the tumor,and the multiple metastatic tumors disappeared.To date,the patient has used anti-PD-1 for 32 months,experiencing no disease progression and maintaining good health without additional treatment.CONCLUSION This case suggests that anti-PD-1 immunotherapy may have long-term positive effects on outcomes in some refractory recurrent solid tumors.Further research is needed to identify patients most likely to respond to anti-PD-1 therapy.
