This report describes triethylammonium acetate （TEAA） ionic liquid catalyzed one pot synthesis of 6-amino-4-aryl-5-cyano-3- methyl-1-phenyl-1,4-dihydropyrano [2,3-c]pyrazoles by the reaction of aromatic aldehyde, malononitrile and 3-methyl-1-phenyl-2- pyrazolin-5-one at room temperature. TEAA plays dual role as reaction media and catalyst. It can also be easily recovered and reused in several runs. TEAA provides greener reaction protocol to present methodology which obviates the need of organic solvents, expensive and toxic catalyst.

Somatic embryogenesis (SE) is a prominent mode of regeneration in plants. The acquisition of SE is predominantly invoked by the oxidative stress which plays an important role in signal transduction and cellular redox. Since balanced generation of oxidants is important to cellular differentiation, modulation in cell redox could be responsive to genotypic refinement for SE. To study the dynamics of cellular redox during SE, we conducted comparative expression analyses of cotton (Gossypium hirsutum), using two independently purified near-isogenic lines for the trait of SE. We interrogated expression changes in cell-signaling factor Somatic Embryogenesis Receptor Kinase (SERK) and activity of antioxidant Glutathione in different developmental stages including cotyledonary leaf, calli from different stages of regeneration of fully-regenerating (FR) and non-regenerating (NR) lines of Coker310 cultivar. At evolutionary scale, the cotton SERKs showed high sequence similarity in receptor kinase domain with diverse systems. Exclusively, SERK1 responsible for potential signaling processes during SE revealed significant expression up-regulation in the embryogenic calli of FR line. Similarly, activity of antioxidant glutathione was substantially up-regulated in embryogenic calli of FR line in comparison to its counterpart form. In contrast, calli from early-stages of regeneration of both FR and NR lines had no significant influences on the regulation of SERK and glutathione levels prior to the acquisition of embryogenesis. These results highlight that in vitro purification of FR line in cotton for enhanced regeneration potential (through SE) resulted in signaling and metabolic transformations of the manner in which cellular redox levels have become modulated.

The regiospecific oxidation of alkyl group of both sterically hindered and unhindered aromatic amine to corresponding carbonyl compound was done in aq. medium by using DDQ. The optimized reaction protocol was found to be most simple, high yielding and novel method for oxidation of alkyl group of aromatic amine in to its carbonyl compound.

Cretaceous-Paleogene Transition of Reptilian Tetrapods across Deccan Volcanism in India

Eruptions of lava flows of Deccan large igneous province straddled the Cretaceous-Palaeogene boundary (K-Pg). Sediments associated at different stratigraphic levels within the lava piles of Deccan volcanic sequences (DVS) are mainly terrestrial. We studied the sediments of Eastern and Central Deccan Volcanic Province, and the Malwa Plateau for tracking changes in reptilian tetrapods across the volcanism. The reptiles are mainly represented by abelisaurid-titanosaurid dinosaurs, Alethinophidia madtsoiia snakes, Notosuchian crocodylomorph Simosuchus, bothremydid Kurmademydinae turtles and Anguimorph and Scincomorphs lizards. The evidences suggest that the non-avian dinosaurs were adversely affected by the arrival of the first volcanic flows locally in the province either within C30N or C29R Maastrichtian. The abelisaurid theropods became extinct whereas a single or two species of titanosauriforme dinosaurs survived but eventually became extinct at least 350 ky before the K-Pg boundary with increasing volcanism. The madtsooid snakes and crocodylimorphs were also adversely affected with decline in their diversity and abundance, whereas the Bothremydid turtles survived the initial onslaught of Deccan volcanism and continued across the K-Pg boundary.

This review focuses on research findings in the area of diagnosis and pathogenesis of hepatitis C virus(HCV)infection over the last few *** information based on published literature provides an update on these two aspects of *** infection,previously called blood transmitted non-A,non-B infection,is prevalent globally and poses a serious public health problem *** diagnosis of HCV infection has evolved from serodetection of non-specific and low avidity anti-HCV antibodies to detection of viral nucleic acid in serum using the polymerase chain reaction(PCR)*** PCR assays detect viral nucleic acid with high accuracy and the exact copy number of viral ***,multiplex assays using real-time PCR are available for identification of HCV-genotypes and their *** contrast to previous methods,the newly developed assays are not only fast and eco-nomic,but also resolve the problem of the window period as well as differentiate present from past *** is a non-cytopathic virus,thus,its pathogenesis is regulated by host immunity and metabolic changes including oxidative stress,insulin resistance and hepatic *** innate and adaptive immunity play an important role in HCV *** lymphocytes demonstrate crucial activity during viral eradication or viral persistence and are influenced by viral proteins,HCV-quasispecies and several metabolic factors regulating liver *** pathogenesis is a very complex phenomenon and requires further study to determine the other factors involved.

The crystal and molecular structure of 2-Amino-3-ethyl carboxamido-4-methyl-5-carboxy ethyl thiophene (C11H16N2O3S) has been investigated from single crystal X-ray diffraction data. The primary focus is to investigate the molecular geometry of this compound in the solid state along with the associated inter and intra-molecular hydrogen bonding and related weak interactions present in this molecule. This compound crystallizes in the monoclinic space group P21/c with cell parameters, a = 8.1344(3) , b = 13.7392(4) , c = 11.4704(4) , β = 100.769(2), V = 1259.36 (7) 3, D = 1.352 g·cm–3, Z = 4. The molecular geometry is stabilized by intra-molecular N-H…O=C and C-H…O interactions along with intramolecular C-H…N and C-H…O interactions which contribute towards the stability of the crystal packing.

Expression of TTV-ORF2 Protein for Detection of Anti-TTV IgG Antibodies in Human Sera

The present study describes the cloning and expression of ORF-2 region of TTV genome and the use of expressed peptide in developing immunoassay for detection of anti-TTV antibodies in serum. Presence of TTV-DNA in serum was detected by PCR amplifying N-22 region of ORF-1 of TTV genome. This was followed by genotyping of TTV by RFLP using N-22 amplicon. Using genotype-1 positive serum as the source of TTV, the ORF-2 region was amplified by PCR and subsequently cloned and expressed in pET-19b vector. The expressed protein, identified as 20 kDa protein on SDS-PAGE gel, was purified by affinity chromatography and then used as antigen to develop western blot assay for detection of anti-TTV antibodies in serum. Analysis of sera for anti-TTV antibodies and their comparison with presence of TTV-DNA, produced encouraging results. There was a good relation between presence of anti-TTV and TTV-DNA in these sera samples. Anti-TTV antibodies could be detected in all TTV-DNA positive sera irrespective of the presence of TTV-genotype. This investigation demonstrates that ORF-2 peptide may be used in developing immunoassay for identification of TTV infection.

Multiplex qPCR for serodetection and serotyping of hepatitis viruses: A brief review

The present review describes the current status of multiplex quantitative real time polymerase chain reaction(q PCR) assays developed and used globally for detection and subtyping of hepatitis viruses in body fluids. Several studies have reported the use of multiplex q PCR for the detection of hepatitis viruses, including hepatitis A virus(HAV), hepatitis B virus(HBV), hepatitis C virus(HCV), hepatitis D virus(HDV), and hepatitis E virus(HEV). In addition, multiplex q PCR has also been developed for genotyping HBV, HCV, and HEV subtypes. Although a single step multiplex q PCR assay for all six hepatitis viruses, i.e., A to G viruses, is not yet reported, it may be available in the near future as the technologies continue to advance. All studies use a conserved region of the viral genome as the basis of amplification and hydrolysis probes as the preferred chemistries for improved detection. Based on a standard plot prepared using varying concentrations of template and the observed threshold cycle value, it is possible to determine the linear dynamic range and to calculate an exact copy number of virus in the specimen. Advantages of multiplex q PCR assay over singleplex or other molecular techniques in samples from patients with co-infection include fast results, low cost, and a single step investigation process.
