Engineering biomolecular systems: Controlling the self-assembly of gelatin to form ultra-small bioactive nanomaterials

The size of nanocarriers determines the biological property of the materials, especially as it relates to intratumoral distribution. Previous research has shown that sizes of 10-50 nm penetrate deep inside the tumor, resulting in better efficacy. On the other hand, studies have shown that gelatin exhibits excellent biological properties, including compatibility, degradability, and toxicity. Therefore, FDA approved gelatin as a safe material to use as an excipient in injectables. The bottleneck is the nonexistence of smaller-sized gelatin nanoparticles (GNPs) to realize the full potential of these biomaterials. Yet, GNPs with sizes of less than 50 nm have not been reported;the synthetic strategy reported in the literature uses “uncontrolled crosslinking coupled with nanoprecipitation”, resulting in larger particle size. We have developed a new method to self-assemble gelatin strands by using an anionic, phosphate-based crosslinker and controlled precipitation. The method we developed produced ultra-small gelatin nanoparticles (GX) of size 10 nm with a high degree of reproducibility, and it was characterized using dynamic light scattering (DLS), Energy-dispersive X-ray spectroscopy (EDS), High-resolution transmission, and scanning electron microscopy (HR-TEM/STEM). We also explored GX as a bioactive platform to encapsulate imaging and therapy agents within the cavity. Interestingly, we were able to encapsulate 2 nm size gold nanoparticles within the void of GX. The versatile nature of the GX particles was further demonstrated by surface functionalizing with larger size gelatin nanoparticles to form core-satellite nanocomposites. Additionally, we studied the tumor penetrability of dye-tagged 10, 50, and 200 nm gelatin nanoparticles. The study showed that smaller size gelatin nanoparticles penetrate deeper tumor regions than larger particles. In general, GX was efficient in penetrating the inner region of the spheroids. In this paper, we introduce new concepts of a-type F-contractive mappings which are essentially weaker than the class of F-contractive mappings given in [21, 22] and different from a-GF-contractions given in [8]. Then, sufficient conditions for the existence and uniqueness of fixed point are established for these new types of contractive mappings, in the setting of complete metric space. Consequently, the obtained results encompass various generalizations of the Banach contraction principle. Moreover, some examples and an application to nonlinear fractional differential equation are given to illustrate the usability of the new theory.

Computer vision is a consistent and advanced technique for image processing,with the propitious outcome,and enormous potential.A computer vision has been strongly adopted in the heterogeneous domain including *** the study of existing research on the role of computer vision in fruits and vegetables among various horticulture products of agriculture fields it is noticed that,the existing survey paper has not focused properly on mathematical framework,feature descriptor,defect detection on multiple datasets of fruits and vegetables *** has motivated us to undertake an extensive *** this paper,we examine the paper broadly related to fruits and vegetables among various horticulture products of agriculture fields,specific model,data pre-processing,data analysis method and overall value of performance accuracy by using a particular performance ***,we study the different type of disease present in various fruit and *** have also focused on the comparison of different machine learning approach with respect to different performance metrics on the same ***,we have found that among all existing machine learning techniques SVM give better classification accuracy.A generalized framework to grade the quality and defect detection of multiple fruits and vegetables is also proposed in this *** paper covers the survey of ninety-eight papers closely related to computer vision in the agricultural *** the survey,we have found that computer vision plays an important role and has a large potential to address the challenges related to the agricultural fields.

MELD or MELD-Na as a Predictive Model for Mortality Following Transjugular Intrahepatic Portosystemic Shunt Placement

Background and Aim:The model for end-stage liver disease(MELD)was originally developed to predict survival after transjugular intrahepatic portosystemic shunt(TIPS).The MELD-sodium(MELD-Na)score has replaced MELD for organ allocation for liver ***,there are limited studies to compare the MELD with MELD-Na to predict mortality after ***:We performed a retrospective chart review of patients who underwent TIPS placement between 2006 and 2016 at our *** primary outcome was mortality,and the secondary outcomes sought to assess which variables could provide prognostic information for mortality after TIPS *** performed receiver operating characteristic(ROC)curve analysis to assess the performance of MELD and ***:There were 186 eligible patients in the *** mean pre-TIPS MELD and MELD-Na were 13 and 15,***,mortality after TIPS was 15%at 30 days and 16.7%at 90 *** a comparison of the areas under the ROCs for MELD and MELD-Na,MELD was superior to MELD-Na for 30-day(0.762 vs.0.709)and 90-day(0.780 vs.0.730)mortality after *** optimal cutoff score for 30-day mortality was 15(0.676–0.848)for MELD and 17(0.610–0.808)for MELD-Na,whereas the optimal cutoff score for 90-day mortality was 16(95%CI:0.705–0.855)for MELD and 17(95%CI:0.643–0.817)for *** were 24 patients with high MELD-Na≥17,but with low MELD<15,and 90-day mortality in this group was 8.3%.Conclusions:Although MELD-Na is a superior prognostic tool to MELD for predicting overall mortality in cirrhotic patients,MELD tended to outperform MELD-Na to predict mortality after TIPS.

Reptilian vertebrates from Deccan volcanic associated sediments of Malwa Plateau in context to reptiles across Maastrichtian-Paleogene volcanic eruptions in Main Deccan Volcanic Province,India

The enormous eruptive volume of India's Deccan continental flood basalts has been linked to global mass extinction at Cretaceous-Paleogene boundary and destablilisation of both marine and terrestrial ecosystems before and after the Chicxulub bolide impact. Deccan Volcanic Province(DVP) currently covers around 500 000 km^2 of land between latitude 15°10′-24°30′N and longitude 70°10′-82°10′E over central India towards the west coast. The pre-erosional volume is estimated at around 1.6 million km^3 which is one of the largest volcanic eruptions on the Earth. The Deccan lava flows in geographically separated areas in DVP are considered to have separate sources and sites of eruptions and were outpoured in different magnetochrons viz. C30 n to C28 n of Maastrichtian to Paleocene. The Deccan volcanic sequences(DVS) in different provinces are associated with(i) sediments deposited before the arrival(Ground Zero) of the first lava flows-designated as infratrappean or Lameta sediments and(ii) sediments between the lava flows deposited during the period of quiescence at different stratigraphic levels designated as intertrappean(interflow) sediments. The Ground Zero Lameta sediments are the major dinosaur bearing sediments deposited in six inland basins viz. 1. Balasinor-Jhabua;2. Sagar;3. Jabalpur;4. Ambikapur-Amarkantak;5. Salburdi;6. Nand-Dongargaon. The sediments represent mainly the terrestrial ecosystems destabilised by the volcanism and they record history of biotic and environmental changes impacted by the volcanism across the Deccan volcanic eruptions. The titanosaurs, crocodylimorphs and bothremydid turtles survived across the first flows in the region in the C30 n. The skeletal remains of the surviving reptiles are preserved in the Deccan volcanic associated intertrappean lake sediments at different stratigraphic levels higher-up in the sequence. Soil and onsite nutrient conservation potential of aromatic grasses at field scale under a shifting cultivated,degraded catchment in Eastern Ghats,India

Land degradation due to soil erosion is a global problem, especially on cultivated hill slopes. Economically important aromatic grasses can protect degraded hill slopes more effectively than field crops, but little information is available on their performance. This study quantifies runoff, sediment yield,enrichment ratios of soil and nutrients, and sediment-associated organic carbon and nutrients losses under three aromatic grass species: citronella(Cymbopogon nardus), lemon(Cymbopogon flexuosus), and palmarosa(Cymbopogon martini), compared with a traditional field crop, finger millet(Eleusine coracana)grown at three land slopes(4%, 8%, and 12%). It was observed that the degree of slope and type of grass both significantly influenced runoff generation. Runoff and sediment yield(SY) were significantly higher at 12% slope than at 8% and 4% slopes. Relation between rainfall and runoff were significant for all the grass species(p < 0.05). Palmarosa, lemon, and citronella grass reduced the SY by 10, 54, and 60%,respectively, over finger millet. SY was also significantly related to rainfall for all the treatments(p <0.05). The threshold runoff values to produce SY were higher for aromatic grasses compared to finger millet. Enrichment of clay, silt, sand, soil organic carbon(SOC), available nitrogen(N), phosphorus(P) and potassium(K) in the sediment were not significantly different between slopes but differed significantly between aromatic grasses and finger millet. Sediment associated nutrient load varied inversely with SY mainly because of the nutrient dissolution effect of high runoff volume. Annual loss of SOC and nutrients varied from 84.7-156.8 kg ha-1y-1for SOC, 4.38-9.18 kg ha-1y-1for available N, 0.35-0.75 kg ha-1y-1for available P, and 2.22-5.22 kg ha-1y-1for available K, with the lowest values for citronella and highest for finger millet. In this paper,we present impedance spectroscopy of Sodium Bismuth Titanate-based materials belonging to(1-x)TNa_(1/2)Bi_(1/2)TiO_(3)-xBaTiO_(3)(x=0.04)(NBT-BT)***-BT ceramics are prepared by high temperature solid-state reaction method.X-ray diffraction technique showed single-phase polycrystalline sample with an ABO3 perovskite *** behavior and the impedance relaxation were investigated in a wide range of temperature(room temperature(RT)-500℃)and frequency(1 kHz-1 MHz).A broad dielectric constant peak was observed over a wide temperature range around the phase transition *** complex impedance plot exhibited one impedance semicircle identified over the frequency range of 1 kHz-1MHz,which is explained by the grain effect of the *** centers of the impedance semicircles lie below the real axis,which indicates that the impedance response is a Cole-Cole type relaxation.

A Botanist’s Cognitive View on Plant Growth: Cross-Talk between Developmental and Sensitivity Networks

An alteration in plant phenotypes assisted by their responses to the environmental stimuli (=tropism) has been fundamental to understand the “plant sensitivity ” that plays a crucial role in plants’ adaptive success. Plants succeed through the deployment of moderators controlling polar auxin-transport determining organ bending. Stimulus-specific effectors can be synthesized by the outer peripheral cells at the bending sites where they target highly conserved cellular processes and potentially persuade the plant sensitivity at large. Remarkably, the peripheral cells require different time-intervals to achieve the threshold expression-levels of stimulus-specific molecular responders. After stimulus perception, tropic curvatures (especially at growing root-apices) are duly coordinated via integrated chemical and electrical signalling which is the key to cellular communications. Thus, the acquired phenotypic alterations are the perplexed outcome of plant’s developmental pace, complemented by the sensitivity. A novel aspect of this study is to advance our understanding of plant developmental-programming and the extent of plant-sensitivity, determining the plant growth and their future applications.

Tobacco Exposed Buccal Mucosa Graft—A Potential Carcinogen for Scrotal Cancer

Squamous cell carcinoma of scrotum is not common. It was the first cancer directly associated with a specific occupation i.e. chimney sweeps. We report a case of squamous cell carcinoma of scrotum developed in a patient of stricture urethra with multiple perineal urinary fistulas treated with lay open urethra with buccal mucosal graft. Tobacco exposed buccal mucosa graft can act as a carcinogen for scrotal cancer in patients with multiple fistula and poor hygiene.
