Effects of ethanol on the tonicity of corporal tissue and the intracellular Ca^2＋ concentration of human corporal smooth muscle cells

Heavy alcohol consumption is associated with an increased risk of erectile dysfunction （ED）; however, the acute effects of ethanol （EtOH） on penile tissue are not fully understood. We sought to investigate the effects of EtOH on corporal tissue tonicity, as well as the intracellular Ca^2＋ concentration （[Ca^2＋]i） and potassium channel activity of corporal smooth muscle. Strips of corpus cavernosum （CC） from rabbits were mounted in organ baths for isometric tension studies. Electrical field stimulation （EFS） was applied to strips precontracted with 10 μmol L^-1 phenylephrine as a control. EtOH was then added to the organ bath and incubated before EFS. The [Ca^2＋]i levels were monitored by the ratio of fura-2 fluorescence intensities using the fura-2 loading method. Single-channel and whole-cell currents were recorded by the conventional patch-clamp technique in short-term cultured smooth muscle cells from human CC tissue. The corpus cavernosal relaxant response of EFS was decreased in proportion to the concentration of EtOH. EtOH induced a sustained increase in [Ca^2＋]i in a dose-dependent manner, Extracellular application of EtOH significantly increased whole-cell K^＋ currents in a concentration-dependent manner （P 〈 0.05）. EtOH also increased the open probability in cell-attached patches; however, in inside-out patches, the application of EtOH to the intracellular aspect of the patches induced slight inhibition of Ca^2＋-activated potassium channel （KCa） activity. EtOH caused a dose-dependent increase in cavemosal tension by alterations to [Ca^2＋]i. Although EtOH did not affect KCa channels directly, it increased the channel activity by increasing [Ca^2＋]i. The increased corpus cavemosal tone caused by EtOH might be one of the mechanisms of ED after heavy drinking.

In this study, we investigated the effects of a combination of Ginkgo biloba extracts （GBE） and phosphodiesterase type 5 （PDE-5） inhibitors on the muscular tone of the corpus cavernosum and potassium channel activity of corporal smooth muscle cells. Strips of corpus cavernosum from male New Zealand white rabbits were mounted in organ baths for isometric tension studies. After contraction with 1 × 10^-5 mol I^-1 norepinephrine, GBE （0.01-1 mg ml^-1） and mirodenafil （0.01-100 nmol I^-1） were added together into the organ bath. In electrophysiological studies, whole-cell currents were recorded by the conventional patch-clamp technique in cultured smooth muscle cells of the human corpus cavernosum. The corpus cavemosum was relaxed in response to GBE in a dose-dependent manner （from 0.64%±8.35% at 0.01 mg ml^-1 to 52.28%±11.42% at 1 mg ml^-1）. After pre-treatment with 0.03 mg ml^-1 of GBE, the relaxant effects of mirodenafil were increased at all concentrations, After tetraethylammonium （TEA）（1 mmol I^-1） administration, the increased effects were inhibited （P〈0.01）. Extracellular administration of GBE increased the whole-cell K^＋ outward currents in a dose-dependent fashion. The increase of the outward current was inhibited by I mmol 1-1 TEA. These results suggest that GBE could increase the relaxant potency of mirodenafil even at a minimally effective dose. The K＋ flow through potassium channels might be one of the mechanisms involved in this synergistic relaxation.

Surface-area-tuned, quantum-dot-sensitized heterostructured nanoarchitectures for highly efficient photoelectrodes

Harvesting solar energy to produce clean hydrogen from photoelectrolysis of water presents a valuable opportunity to find alternatives for fossil fuels. Three- dimensional nanoarchitecturing techniques can afford enhanced photoelectrochemical properties by improving geometrical and structural effects. Here, we report quantum-dot sensitized TiO2-Sb：SnO2 heterostructures as a model electrode to enable the optimization of the structural effects through the creation of a highly conductive pathway using a transparent conducting oxide （TCO）, coupled with a high surface area, by introducing branching and low interfacial resistance via an epitaxial relationship. An examination of various morphologies （dot, rod, and lamella shape） of TiO2 reveals that the rod-shaped TiO2-Sb：SnO2 is a more effective structure than the others. A photoelectrode fabricated using optimized CdS--TiO2-Sb：SnO2 produces a photocurrent density of 7.75 mA/cm2 at 0.4 V versus a reversible hydrogen electrode. These results demonstrate that constructing a branched heterostructure based on TCO can realize highperformance photoelectrochemical devices.

Generally the gold investment material consists of cristobalite,quartz and *** physical property of gold investment materials depends on its thermal expansion coefficients,compressive strength,and particles size *** the thermal expansion coefficient of cristobalite and quartz are 2.6×10^-6/℃and 2.32×10^-6/℃respectively,the composition ratio of each components influence the thermal and physical properties of gold investment *** the clinical applications,it is necessary to improve the properties of gold investment materials such as homogeneous size distribution and thermal expansion *** the present study,effect of inorganic fillers such as cristobalite and quartz on gold alloy investment was investigated to improve the properties of *** compressive strength and thermal expansion coefficients of the specimens were *** results showed that cristobalite and quartz were homogeneously distributed by milling. The optimum compressive strength was obtained at the ratio of 42：22 cristobalite and quartz,respectively.

Enhancing the total organic carbon measurement efficiency for water samples containing suspended solids using alkaline and ultrasonic pretreatment methods

In this study,we investigated the effect of sample pretreatments(ultrasonication and alkaline extraction)on total organic carbon(TOC)measurements for water samples containing suspended solids(SS)of four different origins(algae,soil,sewage sludge,and leaf litter)to more clearly assess the impact of particulate organic carbon(POC)in *** effects each of ultrasonication(power,pulse,etc.)and alkaline extraction condition(concentration,time,etc.)on the TOC recovery and precision were investigated,and the results were compared with those of a new sample pre treatment method combining both *** treatment(0.01 mol/L NaOH)showed higher precision than ultrasonication(100/5 on/off pulse),and notably,the differences among the measured TOC values in samples of different origins were also further reduced in the alkaline *** suggests that the ultrasonic pretreatment results can be mainly attributed to the increase in POC recovery through particle size reduction,whereas the alkaline treatment results are achieved through the enhancement of POC *** is also particularly noteworthy that a higher TOC recovery of 87.6%±7.4%with a higher precision of 8.4%could be obtained using the combined method,compared to each treatment(ultrasonic:TOC recovery 34.7%,relative standard deviation 63.1%;alkaline:49.6%and 23.0%,respectively).Thus,simultaneous pretreatment with ultrasonication and alkaline extraction is expected to increase the oxidation rate of organic matter and the homogeneity of the samples,minimizing the loss of POC measurement values,and thereby improving the reliability of the TOC measurements of water samples containing SS.

Primordial germ cell-mediated transgenesis and genome editing in birds

Transgenesis and genome editing in birds are based on a unique germline transmission system using primordial germ cells(PGCs), which is quite different from the mammalian transgenic and genome editing system. PGCs are progenitor cells of gametes that can deliver genetic information to the next generation. Since avian PGCs were first discovered in nineteenth century, there have been numerous efforts to reveal their origin, specification, and unique migration pattern, and to improve germline transmission efficiency. Recent advances in the isolation and in vitro culture of avian PGCs with genetic manipulation and genome editing tools enable the development of valuable avian models that were unavailable before. However, many challenges remain in the production of transgenic and genome-edited birds,including the precise control of germline transmission, introduction of exogenous genes, and genome editing in ***, establishing reliable germline-competent PGCs and applying precise genome editing systems are critical current issues in the production of avian models. Here, we introduce a historical overview of avian PGCs and their application, including improved techniques and methodologies in the production of transgenic and genome-edited birds, and we discuss the future potential applications of transgenic and genome-edited birds to provide opportunities and benefits for humans.

Hepatocellular carcinoma presenting as organized liver abscess:A case report

BACKGROUND Hepatocellular carcinoma(HCC)is difficult to treat and has a high mortality rate,emphasizing the importance of early diagnosis and *** characteristic radiologic findings and underlying liver disease are present,a diagnosis can be made without a ***,when HCC is accompanied by a liver abscess,diagnosis might be delayed by atypical radiologic *** case report aims to assist in the diagnosis of HCC,which can manifest in various *** SUMMARY A 75-year-old male presented to the Emergency Department with worsening fever and mental *** was diagnosed with liver cirrhosis six months *** computed tomography(CT)raised our suspicion of an organized liver abscess.A follow-up CT scan after four weeks of antibiotic treatment showed a decrease in the liver lesion ***,high fever recurred,and C-reactive protein increased to 14 mg/*** of the liver lesion was performed,but no bacteria were *** culture revealed the presence of *** patient received an additional four weeks of antibiotics and antifungal agents before being *** 10 mo later,a CT scan showed an increase in the lesion size,and biopsy was *** biopsy revealed an organized abscess with focal carcinomatous changes,for which surgery was *** histopathological examination revealed HCC,clear-cell *** nontumor liver tissue showed cirrhosis and an organized *** Even if a liver abscess is suspected in a patient with cirrhosis,the possibility of HCC should be considered.

Effect of Radiant Spectral Characteristics on Performance of a Near-Infra-Red Heating Module

We simulated the heat transfer phenomena of the heating module that is primarily based on the radiant energy in the near-infra-red(NIR) *** the module,the power emitted by the lamp filament is distributed to the lamp glass,reflector,and the target medium,which are cooled by an air *** radiant heat transfer is simulated by using the ray-tracing scheme,in which the spectral characteristics of the emission and the materials are *** heat transport from the lamp glass to the cooling air is analyzed by using the finite volume *** the lamp-filament temperature rises in the range of 3000-3400K,the NIR radiant power on the target medium ***,the lamp-glass temperature also rises,and the proportion of the NIR power to the entire radiation has a peak in the temperature *** spectral distributions of the absorbed energies in all the components in the module are highly non-uniform,and a monochromatic model of the radiant heat transfer may result in a significant discrepancy.

Anti-cancer effect of ethylacetate fraction from Orostachys japonicus on HT-29 human colon cancer cells by induction of apoptosis through caspase-dependent signaling pathway

Objective: To investigate the anti-colon cancer effects of ethylacetate fraction from Orostachys japonicus(0. japonicus) on HT-29 cancer cells. Methods: The viability of HT-29 cells was assayed by the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2 H-tetrazolium(MTS) method. Apoptosis induction and cell cycle inhibition were confirmed by fluorescein isothiocyanate and propidium iodide staining using flow *** changes in the nucleus were observed, using a fluorescence microscope with4',6-diamidino-2-phenylindole(DAPI) nuclear staining. The expression levels of the upstream and downstream proteins involved in the anti-cancer mechanism were confirmed by Western blotting. Results: After treating HT-29 cells with different concentrations of ethylacetate fraction from O. japonicus, the viability of cells decreased in a concentration-dependent manner,while apoptosis induction and apoptotic body formation increased. Cell cycle analysis showed that the arrest occurred at the sub-G_1 and S phase. Among the upstream and downstream proteins involved in anti-cancer activity, the level of B cell lymphoma-2 decreased, and the bcl-2-associated x protein increased. The level of pro-caspase-3, pro-caspase-8, and pro-caspase-9 decreased, while the level of cleaved-caspase-3, cleaved-caspase-8, and cleaved-caspase-9 increased. Moreover, the phosphorylation, that is, activation of extracellular signal regulated kinase 1/2, Jun-N-terminal kinase, and p38 increased. Conclusions: Combining the above results, it is thought that the survival of HT-29 cells is suppressed by ethylacetate fraction from0. japonicus through mitochondrial regulation-induced caspase cascade activation, induction of apoptosis and cell cycle arrest.
