Objective:To investigate the morphological structure of ovarian follicular cells and biochemical parameters of both ovaries and fat bodies(sites of vitellogenesis)from Rhodnius(R.)prolixus infected with Trypanosoma(T.)rangeli.Methods:Adult virgin females of R.prolixus were fed upon a membrane apparatus containing heat-inactivated citrated rabbit blood and a suspension of T.rangeli epimastigotes(Macias strain).Females from the control group and all the males received parasite-free blood.Transmission electron microscopy was used to reveal the morphological aspects of ovarian follicle cells in both control and parasite-infected groups.Protein profile,proteolytic activities and Western blotting analyses were performed in either ovary or fat body samples of control and parasite-infected groups.Results:According to the ultrastructural data,T.rangeli infection elicited a degeneration process in the ovarian follicular cells of R.prolixus.Proteolytic assays indicated a reduction in the activity of aspartic peptidases in the ovary and fat body from parasite-infected group,while a significant increase in the cysteine peptidase activity was measured in both insect organs.Additionally,immunoblotting revealed that vitellogenin was overexpressed in the ovary of parasite-infected insects.Conclusions:T.rangeli infection seems to elicit an early programmed cell death in the ovarian follicle cells as well as induces the modulation on the activities of different peptidase classes in either ovaries or fat bodies and the overexpression of the vitellogenin in the ovary of R.prolixus.

Use of CRISPR-Cas9(Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)-CRISPR-associated 9)-mediated genome editing has proliferated for use in numerous plant species to modify gene function and expression,usually in the context of either transient or stably inherited genetic alternations.While extremely useful in many applications,modification of some loci yields outcomes detrimental to further experimental evaluation or viability of the target organism.Expression of Cas9 under a promoter conferring gene knockouts in a tissue-specific subset of genomes has been demonstrated in insect and animal models,and recently in Arabidopsis.We developed an in planta GFP(green fluorescent protein)assay system to demonstrate fruit-specific gene editing in tomato using a phosphoenolpyruvate carboxylase 2 gene promoter.We then targeted a SET-domain containing polycomb protein,SlEZ2,previously shown to yield pleiotropic phenotypes when targeted via ^(35)S-driven RNA interference and we were able to characterize fruit phenotypes absent additional developmental perturbations.Tissue-specific gene editing will have applications in assessing function of essential genes otherwise difficult to study via germline modifications and will provide routes to edited genomes in tissues that could not otherwise be recovered when their germline modification perturbs their normal development.

Pretreatment of Soil Samples Rich in Short-Range-Order Minerals Before Particle-Size Analysis by the Pipette Method

The possibilities of combining the dissolution of short-range-order minerals (SROMs) like allophane and imogolite, by ammonium oxalate and a particle size distribution analysis performed by the pipette method were investigated by tests on a soil sample from Reunion, a volcanic island located in the Indian Ocean, having a large SROMs content. The need to work with moist soil samples was again emphasized because the microaggregates formed during air-drying are resistant to the reagent. The SROM content increased, but irregularly, with the number of dissolutions by ammonium oxalate: 334 and 470 mg g-1 of SROMs were dissolved after one and three dissolutions respectively. Six successive dissolutions with ammonium oxalate on the same soil sample showed that 89% of the sum of oxides extracted by the 6 dissolutions were extracted by the first dissolution (mean 304 mg g-1 ). A compromise needs to be found between the total removal of SROMs by large quantities of ammonium oxalate and the preservation of clay minerals, which were unexpectedly dissolved by this reagent. These tests enabled a description of the clay assemblage of the soil (gibbsite, smectite, and traces of kaolinite) in an area where such information was lacking due to the difficulties encountered in recuperation of the clay fraction.

Tracking a Tip Vortex with Adaptive Vorticity Confinement and Hybrid RANS-LES

The prediction of coherent vortices with standard RANS solvers suffers especially from discretisation and modelling errors which both introduce numerical diffusion. The adaptive Vorticity Confinement (VC) method targets to counteract one part of the discretisation error: the one due to the discretisation of the convection term. This method is applied in conjunction with a hybrid RANS-LES turbulence model to overcome the overprediction of turbulence intensity inside vortex cores which is a typical deficiency of common RANS solvers. The third main source for numerical diffusion originates from the spatial discretisation of the solution domain in the vicinity of the vortex core. The corresponding error is analysed within a grid convergence study. A modification of the adaptive VC method used in conjunction with a high-order discretisation of the convection term is presented and proves to be superior. The simulations of a wing tip vortex flow are validated in terms of vortex velocity profiles using the results of a wind tunnel experiment performed by Devenport and colleagues (1996). Besides, the results are compared with another numerical study by Wells (2009) who uses a Reynolds Stress turbulence model. It turns out that the application of the modified adaptive VC method on the one hand reinforces the tip vortex, and on the other hand accelerates the axial flow which leads to a slight degradation compared to the experimental results. The result of Wells is more accurate close to the wing, but the result obtained here is superior further downstream as no excessive diffusion of the tip vortex occurs.

通过人工合成的人类免疫缺陷症病毒（Ｈｕｍａｎｉｍｍｕｎｏｄｅｆｉｃｉｅｎｃｙｖｉｒｕｓ，ＨＩＶ）糖蛋白肽ＧＰ１２０对人和无脊椎动物（Ｍｙｔｉｌｕｓｅｄｕｌｉｓ）免疫细胞的抑制作用的研究。人单核细胞和Ｍｙｔｉｌｕｓｅｄｕｌｉｓ免疫细胞分别与ＧＰ１２０保温后，均抑制细胞的吞噬细菌（Ｐｓｕｄｏｍｏｎａｓｓｔｒｅｔｚｉ）作用。应用计算机显微图像术（Ｃｏｍｐｕｔｅｒ－ａｓｓｉｓｔｅｄｍｉｃｒｏｓｃｏｐｙ）直接计算细菌被吞噬的情况，并测定细胞形状改变的参数。结果发现，细菌存在时，ＧＰ１２０使免疫细胞活性降低，细胞变圆（形状因子＝０．８６±０．０６），　而对照组免疫细胞则因细菌的刺激作用变长或阿米巴样（形状因子＜０．５），噬菌能力明显增加，提示ＧＰ１２０是ＡＩＤＳ病人细胞免疫机制受损害的原因之一。

Much of the genetic predisposition to polyposis,and particularly to serrated polyposis(SP),remains unknown.Only germline pathogenic variants in RNF43,a tumor suppressor that exerts negative feedback in the Wnt/β-catenin signaling pathway,have been causally linked to some SP cases(<2%),a disease associated with increased risk of colorectal cancer(CRC).^(1) Most known hereditary CRC and polyposis genes affect DNA repair,BMP/TGF-β,or Wnt signaling,being the latter associated with adenomatous and serrated polyposis phenotypes.2 Based on this observation,we evaluated the presence and role of germline variants in those pathways in unsolved polyposis patients.
