Identification of novel genes associated with atherosclerosis in Bama miniature pig

Background:Atherosclerosis is a chronic cardiovascular disease of great concern.However,it is difficult to establish a direct connection between conventional small animal models and clinical practice.The pig's genome,physiology,and anatomy reflect human biology better than other laboratory animals,which is crucial for studying the pathogenesis of atherosclerosis.Methods:We used whole-genome sequencing data from nine Bama minipigs to perform a genome-wide linkage analysis,and further used bioinformatic tools to filter and identify underlying candidate genes.Candidate gene function prediction was performed using the online prediction tool STRING 12.0.Immunohistochemistry and immunofluorescence were used to detect the expression of proteins encoded by candidate genes.Results:We mapped differential single nucleotide polymorphisms(SNPs)to genes and obtained a total of 102 differential genes,then we used GO and KEGG pathway enrichment analysis to identify four candidate genes,including SLA-1,SLA-2,SLA-3,and TAP2.nsSNPs cause changes in the primary and tertiary structures of SLA-I and TAP2 proteins,the primary structures of these two proteins have undergone amino acid changes,and the tertiary structures also show slight changes.In addition,immunohistochemistry and immunofluorescence results showed that the expression changes of TAP2 protein in coronary arteries showed a trend of increasing from the middle layer to the inner layer.Conclusions:We have identified SLA-I and TAP2 as potential susceptibility genes of atherosclerosis,highlighting the importance of antigen processing and immune response in atherogenesis.

Photo-stimuli responsive materials show great potential in the fields of information encryption and storage due to their distinctive spatial or temporal color changes.However,the conventional single or multi-color static outputs by light stimulus difficulty meet practical requirements for high-security optical storage technologies.Here,a novel dynamic irradiation-responsive phosphor of Na_(2)Ba_(2)Si_(2)O_(7):Eu is demonstrated,exhibiting high storage stability and convenient readout behaviors.The inherent Eu^(2+)luminescence can be dynamically tuned,instantly read out,and conveniently erased by controlling irradiation duration of a portable diode laser(365 nm).The modulation mechanism is unraveled by optically induced oxidation reactions of Eu^(2+)→Eu^(3+)and defects as killer centers.The excellent luminescence modulation degree(ΔRt=89.5%)and the accompanying larger color contrast enable the creation of invisible optical codes with multi-level encryption in bright or dark field.These results indicate potential applications of Na_(2)Ba_(2)Si_(2)O_(7):-based materials in information encryption and invisible optical storage,and are expected to expand more investigations on optically induced PL modulation behaviors based on mixed valences and defects.

Monolayer boron-based materials are of current interests due to its polymorphism.Herein,motivated by the recent experimental synthesis of semiconducting hydrogenatedαʹ-borophene and the regulation of the physical properties in layered materials by surface functionalization,we study the thermal and electronic properties ofαʹ-borophene with three different types of gas functional groups(H,F,and Cl)based on first-principles and Boltzmann transport theory.It is found thatαʹ-borophene can be well stabilized by fluorination and chlorination and maintain the semiconductor nature.More interestingly,when hydrogen is replaced with fluorine or chlorine,the lattice thermal conductivity changes from 24.3 to 5.2 or 0.73 W/(m·K)along armchair direction at 300 K,exhibiting a huge reduction by two orders of magnitude.The main reason is the decrease of both phonon group velocities and acoustic phonon relaxation time resulting from the strong phonon mode softening due to the weaken B-B bond strength and heavier atomic mass of fluorine and chlorine.Consequently,the chlorinatedαʹ-borophene exhibits a high thermoelectric figure of merit~2 at 300 K along armchair direction.Our study illustrates the importance of the modulation of transport properties by gas functional groups,which may promote the thermoelectric application of boron-based materials.

Origami has become an optimal methodological choice for creating complex three-dimensional(3D)structures and soft robots.The simple and low-cost origami-inspired folding assembly provides a new method for developing 3D soft robots,which is ideal for future intelligent robotic systems.Here,we present a series of materials,structural designs,and fabrication methods for developing independent,electrically controlled origami 3D soft robots for walking and soft manipulators.The 3D soft robots are based on soft actuators,which are multilayer structures with a dielectric elastomer(DE)film as the deformation layer and a laser-cut PET film as the supporting flexible frame.The triangular and rectangular design of the soft actuators allows them to be easily assembled into crawling soft robots and pyramidal-and square-shaped 3D structures.The crawling robot exhibits very stable crawling behaviors and can carry loads while walking.Inspired by origami folding,the pyramidal and square-shaped 3D soft robots exhibit programmable out-of-plane deformations and easy switching between two-dimensional(2D)and 3D structures.The electrically controllable origami deformation allows the 3D soft robots to be used as soft manipulators for grasping and precisely locking 3D objects.This work proves that origami-inspired fold-based assembly of DE actuators is a good reference for the development of soft actuators and future intelligent multifunctional soft robots.

Thermal conductivity of two-dimensional(2D)materials has gained prominence due to the attractive applications in thermal management and thermoelectric devices.In this work,we present a new member of bilayer 2D boron allotropes,denoted as bilayerβ12 borophene,and study the thermal transport properties by solving phonon Boltzmann transport equation based on density functional theory.Based on quantitative chemical bonding analysis,we identify large degrees of covalent bonding of the interlayer interaction.In comparison to its monolayer counterpart,the bilayer exhibits much higher in-plane thermal conductivity despite the lower phonon group velocity and buckling structure,inferring a new physical mechanism.The thermal conductivity(κ)of bilayerβ_(12)borophene at 300 K is 140.5(86.3)W·m^(-1)·K^(-1)along armchair(zigzag)direction,andκ_(armchair)is about 52.7%higher than that of monolayerβ12 borophene.The abnormal enhancement is attributed to the suppressed phonon scattering possibility and elongation of phonon lifetime.More interesting,after forming bilayerβ12 borophene through interlayer covalent bonding,the dominated phonon branch to thermal conductivity changes to transverse acoustic phonons from out-of-plane flexural acoustic(ZA)phonons in the monolayer borophene.Our study elucidates the rich thermal transport characteristics in bilayer covalently bonded 2D materials,and injects fresh insights into the phonon engineering of 2D borophene relevant for emergent thermal management applications.

The piezoelectric performance serves as the basis for the applications of piezoelectric ceramics.The ability to rapidly and accurately predict the piezoelectric coefficient(d_(33))is of much practical importance for exploring high-performance piezoelectric ceramics.In this work,a data-driven approach combining feature engineering,statistical learning,machine learning(ML),experimental design,and synthesis is trialed to investigate its accuracy in predicting d_(33) of potassium-sodium-niobate(K,Na)NbO_(3),KNN)-based ceramics.The atomic radius(AR),valence electron distance(DV)(Schubert),Martynov-Batsanov electronegativity(EN-MB),and absolute electronegativity(EN)are summarized as the four most representative features in describing d_(33) out of all 27 possible features for the piezoelectric ceramics.These four features contribute greatly to regression learning for predicting d_(33) and classification learning for distinguishing polymorphic phase boundary(PPB).The ML method developed in this work exhibits a high accuracy in predicting d_(33) of the piezoelectric ceramics.An example of KNN combined with 6 mo1%liNbO_(3)demonstrates d_(33)3 of 184 pC/N,which is highly consistent with the predicted result.This work proposes a novel feature-oriented guideline for accelerating the design of piezoelectric ceramic systems with large d_(33),which is expected to be widely used in other functional materials.

Efficacy and safety of first-line treatment for metastatic triple-negative breast cancer:A network meta-analysis

Background:Metastatic triple-negative breast cancer(mTNBC)is an aggressive histological subtype with poor prognosis.Several first-line treatments are currently available for mTNBC.This study conducted a network meta-analysis to compare these first-line regimens and to determine the regimen with the best efficacy.Methods:A systematic search of PubMed,EMBASE,the Cochrane Central Register of Controlled Bases,and mi-nutes of major conferences was performed.Progression-free survival(PFS),overall survival(OS),and objective response rate(ORR)were analyzed via network meta-analysis using the R software(R Core Team,Vienna,Austria).The efficacy of the treatment regimens was compared using hazard ratios and 95%confidence intervals.Results:A total of 29 randomized controlled trials involving 4607 patients were analyzed.The ranking was based on the surface under the cumulative ranking curve.Network meta-analysis results showed that cisplatin combined with nab-paclitaxel or paclitaxel was superior to docetaxel plus capecitabine in terms of PFS and ORR.For programmed death-ligand 1(PD-L1)and breast cancer susceptibility gene(BRCA)mutation-positive tumors,atezolizumab/pembrolizumab combined with nab-paclitaxel and talazoparib was superior to docetaxel plus capecitabine.No significant difference was observed among the treatments in Os.Neutropenia,diarrhea,and fatigue were common serious adverse events.Conclusion:Cisplatin combined with nab-paclitaxel or paclitaxel is the preferred first-line treatment for mTNBC.For PD-L1 and BRCA mutation-positive tumors,atezolizumab/pembrolizumab combined with nab-paclitaxel and talazoparib is an effective treatment option,Neutropenia,diarrhea,and fatigue are frequently occurring serious adverseevents.
