Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Objective:Research has shown that celastrol can effectively treat a variety of diseases,yet when passing a certain dosage threshold,celastrol becomes toxic,causing complications such as liver and kidney damage and erythrocytopenia,among *** this dichotomy in mind,it is extremely important to find ways to preserve celastrol’s efficacy while reducing or preventing its ***:In this study,insulin-resistant Hep G2(IR-Hep G2)cells were prepared using palmitic acid and used for in vitro ***-Hep G2 cells were treated with celastrol alone or in combination with Nacetylcysteine(NAC)or ferrostatin-1(Fer-1)for 12,24 or 48 h,at a range of *** counting kit-8assay,Western blotting,quantitative reverse transcription-polymerase chain reaction,glucose consumption assessment,and flow cytometry were performed to measure celastrol’s cytotoxicity and whether the cell death was linked to ***:Celastrol treatment increased lipid oxidation and decreased expression of anti-ferroptosis proteins in IR-Hep G2 *** downregulated glutathione peroxidase 4(GPX4)m *** docking models predicted that solute carrier family 7 member 11(SLC7A11)and GPX4 were covalently bound by ***,we found for the first time that the application of ferroptosis inhibitors(especially NAC)was able to reduce celastrol’s toxicity while preserving its ability to improve insulin sensitivity in IR-Hep G2 ***:One potential mechanism of celastrol’s cytotoxicity is the induction of ferroptosis,which can be alleviated by treatment with ferroptosis *** findings provide a new strategy to block celastrol’s toxicity while preserving its therapeutic effects.

In silico screening of Chinese herbal medicines with the potential to directly inhibit 2019 novel coronavirus

Objective: In this study we execute a rational screen to identify Chinese medical herbs that are commonly used in treating viral respiratory infections and also contain compounds that might directly inhibit 2019 novel coronavirus(2019-nCoV), an ongoing novel coronavirus that causes ***: There were two main steps in the screening process. In the first step we conducted a literature search for natural compounds that had been biologically confirmed as against sever acute respiratory syndrome coronavirus or Middle East respiratory syndrome coronavirus. Resulting compounds were cross-checked for listing in the Traditional Chinese Medicine Systems Pharmacology *** meeting both requirements were subjected to absorption, distribution, metabolism and excretion(ADME) evaluation to verify that oral administration would be effective. Next, a docking analysis was used to test whether the compound had the potential for direct 2019-nCoV protein *** the second step we searched Chinese herbal databases to identify plants containing the selected compounds. Plants containing 2 or more of the compounds identified in our screen were then checked against the catalogue for classic herbal usage. Finally, network pharmacology analysis was used to predict the general in vivo effects of each selected ***: Of the natural compounds screened, 13 that exist in traditional Chinese medicines were also found to have potential anti-2019-nCoV activity. Further, 125 Chinese herbs were found to contain 2 or more of these 13 compounds. Of these 125 herbs, 26 are classically catalogued as treating viral respiratory infections. Network pharmacology analysis predicted that the general in vivo roles of these26 herbal plants were related to regulating viral infection, immune/inflammation reactions and hypoxia ***: Chinese herbal treatments classically used for treating viral respiratory infection might contain direct anti-2019-nCoV compounds.

Effect of low-speed waterjet pressure on the rock-breaking performance of unsubmerged cavitating abrasive waterjet

Unsubmerged cavitating abrasive waterjet(UCAWJ) has been shown to artificially create a submerged environment that produces shear cavitation, which effectively enhances rock-breaking performance. The shear cavitation generation and collapse intensity depend on the pressure difference between the intermediate high-speed abrasive waterjet and the coaxial low-speed waterjet. However, the effect of the pressure of the coaxial low-speed waterjet is pending. For this purpose, the effect of low-speed waterjet pressure on rock-breaking performance at different standoff distances was experimentally investigated,and the effects of erosion time and ruby nozzle diameter on erosion performance were discussed. Finally,the micromorphology of the sandstone was observed at different locations. The results show that increased erosion time and ruby nozzle diameter can significantly improve the rock-breaking performance. At different standoff distances, the mass loss increases first and then decreases with the increase of low-speed waterjet pressure, the maximum mass loss is 10.4 g at a low-speed waterjet pressure of0.09 MPa. The surface morphology of cavitation erosion was measured using a 3D profiler, the increase in both erosion depth and surface roughness indicated a significant increase in the intensity of the shear cavitation collapse. At a low-speed waterjet pressure of 0.18 MPa, the cavitation erosion surface depth can reach 600 μm with a roughness of 127 μm.

Celastrol targets IRAKs to block Toll-like receptor 4-mediated nuclear factor-κB activation

OBJECTIVE： Celastrol has been established as a nuclear factor-κB（NF-κB） activation inhibitor; however, the exact mechanism behind this action is still unknown. Using text-mining technology, the authors predicted that int erleukin-1 receptor-associated kinases（IRA Ks） are potential celastrol targets, and hypothesized that targeting IRAKs might be one way that celastrol inhibits NF-κB. This is because IRAKs are key molecules for some crucial pathways to activate NF-κB（e.g., the inter leukin-1 receptor（IL-1R）/Toll- like receptor（TLR） superfamily）.METHODS： The human hepatocellular cell line（Hep G2） treated with palmitic acid（PA） was used as a model for stimulating TLR4/NF-κB activation, in order to observe the potential effects of celastrol in IRAK regulation and NF-κB inhibition. The transfection of small interfering RNA was used for down-regulating TLR4, IRAK1 and IRAK4, and the Western blot method was used to detect changes in the protein ***： The results showed that celastrol could effectively inhibit PA-caused TLR4-dependent NF-κB activation in the Hep G 2 cells; PA also activated IRAKs, which were inhibited by celastrol. Knocking down IRAKs abolished PA-caused NF-κB ***： The results for the first time show that targeting IRAKs is one way in which celastrol inhibits NF-κB activation.

ZrO_(2)-strengthened porous mullite insulation materials were prepared by foaming technology utilizing ZrSiO_(4) and Al_(2)O_(3) as primary materials and Y_(2)O_(3) as an *** effects of Y_(2)O_(3) contents on the phase composition,microstructure,mechanical properties,and heat conductivity of the porous mullite insulation materials were investigated.A suitable Y_(2)O_(3) content could promote phase transition of monoclinic ZrO_(2)(m-ZrO_(2))to tetragonal ZrO_(2)(t-ZrO_(2)),reduce pore size,and improve the strengths of as-prepared *** cold crushing strength and bending strength of as-prepared specimens with a 119µm spherical pore size using 6 wt.%Y_(2)O_(3) were 35.2 and 13.0 MPa,respectively,with a heat conductivity being only 0.248 W/(m K).

The incineration technology of kitchen waste is one of the effective technologies to achieve the resource utilization of municipal solid *** is an important stage of *** is a rich initial product in the pyrolysis process of kitchen waste,and the presence of H_(2)O has a significant impact on the decomposition of indole to form NO_(x) ***,this study uses density functional theory method to study the effect of H_(2)O on the thermal decomposition of indole to produce NH_(3),HNCO,and *** H_(2)O participates in the reaction,it can provide oxidative groups to generate a new product HNCO,which is different from the previous findings by indole pyrolysis without the presence of H_(2)***,this study theoretically proves that NH_(3) is easier to form than *** is consistent with the phenomenon that NH_(3) release is higher than HCN release in pyrolysis *** addition,compared with the individual pyrolysis of indole,the participation of H_(2)O reduces the energy barriers for the formation of NH_(3) and HCN during indole pyrolysis,thereby promoting the formation of NH_(3) and HCN.

Transparent micropatterned conductive films based on highlyordered nanowire network

Effect of annealing temperature on microstructure and mechanical properties of Mg-Zn-Zr-Nd alloy with large final rolling deformation

In this study,the Mg-3Zn-0.5Zr-χNd(χ=0,0.6)alloys were subjected to final rolling treatment with large deformation of 50%.The impact of annealing temperatures on the microstructure and mechanical properties was *** rolled Mg-3Zn-0.5Zr-0.6Nd alloy exhibited an ultimate tensile strength of 386 MPa,a yield strength of 361 MPa,and an elongation of 7.1%.Annealing at different temperatures resulted in reduced strength and obviously increased elongation for both *** mechanical properties for the Mg-3Zn-0.5Zr-0.6Nd alloy were achieved after annealing at 200℃,with an ultimate tensile strength of 287 MPa,a yield strength of 235 MPa,and an elongation of 26.1%.The numerous deformed microstructures,twins,and precipitated phases in the rolled alloy could impede the deformation at room temperature and increase the work hardening *** annealing,a decrease in the work hardening effect and an increase in the dynamic recovery effect were obtained due to the formation of fine equiaxed grains,and the increased volume fraction of precipitated phases,which significantly improved the elongation of the ***,the addition of Nd element could enhance the annealing recrystallization rate,reduce the Schmid factor difference between basal and prismatic slip systems,facilitate multi-system slip initiation and improve the alloy plasticity.

Bi-Frobenius Algebra Structures on Quantum Complete Intersections

This paper is to look for bi-Frobenius algebra structures on quantum complete intersections over field *** find a class of comultiplications,such that if√−1∈k,then a quantum complete intersection becomes a bi-Frobenius algebra with comultiplication of this form if and only if all the parameters qij=±***,it is proved that if√−1∈k then a quantum exterior algebra in two variables admits a bi-Frobenius algebra structure if and only if the parameter q=±√*** if−1/∈k,then the exterior algebra with two variables admits no bi-Frobenius algebra *** prove that the quantum complete intersections admit a bialgebra structure if and only if it admits a Hopf algebra structure,if and only if it is commutative,the characteristic of k is a prime p,and every ai a power of *** also provides a large class of examples of bi-Frobenius algebras which are not bialgebras(and hence not Hopf algebras).In commutative case,other two comultiplications on complete intersection rings are given,such that they admit non-isomorphic bi-Frobenius algebra structures.
