Connective tissue growth factor is associated with the early renal hypertrophy in uninephrectomized diabetic rats

Background Renal hypertrophy has been regarded as the early feature of diabetic nephropathy （DN）, which may eventually lead to proteinuria and renal fibrosis. However, the exact mechanism of renal hypertrophy is still unclear. The aim of this study was to investigate the possible association of connective tissue growth factor （CTGF） with renal hypertrophy in uninephrectomized diabetic rats. Methods Seventy-two Sprague-Dawley （SD） rats were randomly divided into two groups： control group （group C, n=32） and diabetic nephropathy （group DN, n=40）. Each group was re-divided into 4 subgroups according to the experimental period. The rats were sacrificed at 1, 2, 4, and 8 weeks respectively after induction of diabetes. Diabetes was induced by intraperitoneal injection of streptozotocin （STZ） after rats had received uninephrectomy. Blood glucose （BG）, body weight （BW）, 24-h urinary albumin excretion （24hUalb）, kidney weight （KW）, KW/BW, glomerular tuft area （AG）, glomerular tuft volume （VG）, proximal tubular area （AT） at each time point, the width of glomerular basement membrane （GBM） and tubular basement membrane （TBM） at week 8 were measured when the rats were sacrificed. Renal expression of CTGF and p27kipl were detected by immnnohistochemical staining. The relationship between CTGF expression and increasing of VG and AT was analyzed. Results There was a significant increase of 24hUalb, KW, and KW/BW from week 1 onward in diabetic rats compared to those in group C （P〈0.05, respectively）, diabetic rats also had a significant increase of AG, VG, and AT from week 1 onward. It was also shown that diabetic rats had a thickening of GBM [（245.7±103.0） nm vs （121.8±19.1） nm, P〈0.01] and TBM [（767.7±331.1） nm vs （293.0±110.5） nm, P〈0.01] at week 8. There was a weak expression for CTGF and p27kipl in normal glomeruli and tubuli, while a significant increasing expression of CTGF and p27kipl was found in glomeruli and tubuli in diabetic kidney from week 1 onward （P〈0.05, respectively）, and the extent of CTGF expression was p

Studies have found that the absence of glial cell line-derived neurotrophic factor may be the primary risk factor for Parkinson’s disease. However, there have not been any studies conducted on the potential relationship between glial cell line-derived neurotrophic factor and cognitive performance in Parkinson’s disease. We first performed a retrospective case-control study at the Affiliated Hospital of Xuzhou Medical University between September 2018 and January 2020 and found that a decreased serum level of glial cell line-derived neurotrophic factor was a risk factor for cognitive disorders in patients with Parkinson’s disease. We then established a mouse model of Parkinson’s disease induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine and analyzed the potential relationships among glial cell line-derived neurotrophic factor in the prefrontal cortex, dopamine transmission, and cognitive function. Our results showed that decreased glial cell line-derived neurotrophic factor in the prefrontal cortex weakened dopamine release and transmission by upregulating the presynaptic membrane expression of the dopamine transporter, which led to the loss and primitivization of dendritic spines of pyramidal neurons and cognitive impairment. In addition, magnetic resonance imaging data showed that the long-term lack of glial cell line-derived neurotrophic factor reduced the connectivity between the prefrontal cortex and other brain regions, and exogenous glial cell line-derived neurotrophic factor significantly improved this connectivity. These findings suggested that decreased glial cell line-derived neurotrophic factor in the prefrontal cortex leads to neuroplastic degeneration at the level of synaptic connections and circuits, which results in cognitive impairment in patients with Parkinson’s disease.

Cracking Behavior in Additively manufactured Pure Tungsten

In this study, near fully dense(96.5%) pure tungsten bulks were additively manufactured and the cracking behavior was investigated. A crack network with a spacing of ～100 lm was observed in the fabricated bulks. It was observed that the laser scanning strategy, which could tailor the microstructure, affected the crack distribution pattern in fabricated tungsten.The calculated surface temperature difference(7300 K) was much higher than the cracking criterion(800 K) of tungsten,indicating that cracking is almost inevitable in laser additive manufacturing of tungsten. It could be concluded that crack network formed because the cracks emerged in every laser molten track and then interconnected in the layer-by-layer building process.

Tumor-suppressor microRNA-139-5p restrains bladder cancer cell line ECV-304 properties via targeting Connexin 43

Background:In our previous paper,we demonstrated that Connexin 43(CX43)was highly expressed in bladder cancer(BC)tissues.But the molecular mechanism about microRNAs(miRNAs)regulation upstream of CX43 in BC has not been well elucidated and remains to be further studied.MicroRNA-139-5p(miR-139-5p)is a tumor suppressor in progression of multifarious cancers including BC.Nevertheless,the underlying mechanisms of CX43/miR-139-5p in tumorigenesis of BC are still not well illustrated.The specific objective of our study was to inquiry the effect of CX43/miR-139-5p on BC progression and its underlying mechanism.Methods:The bioinformatics analysis softwares were applied to predict the miRNAs in the upstream of CX43.First,the expression levels of miR-139-5p in BC tissues(tumor)and paracancer tissues(normal)were investigated using the data from The Cancer Genome Atlas database.Quantitative reverse transcription-polymerase chain reaction(qRT-PCR)was used to detect the mRNA expression level of miR-139-5p in three human BC cell lines 5637,T24,ECV-304 and a human bladder epithelial immortalized cell line SV-HUC-1(normal control).Then si-CX43,si-control,miR-139-5p mimic,and its negative control(NC)were transfected into BC cell line ECV-304.The relationship of miR-139-5p and CX43 was analyzed by dual-luciferase reporter assay.The qRT-PCR and Western blotting were used to test the mRNA and protein expression level of CX43.The proliferation of ECV-304 and T24 cells were examined by cell counting kit-8.The migration and invasion of ECV-304 cells were tested by transwell assay.To determine whether miR-139-5p would affect cell proliferation,migration and invasion by targeting CX43,we executed the rescue assay.The comparison between two groups was analyzed by Student’s t test,and comparisons among multiple samples were performed by oneway analysis of variance and a Bonferroni post hoc test.Results:The expression of miR-139-5p was remarkably down-regulated in BC tissues(tumor vs.normal,2.286±0.017 vs.3.211±0.034,t=11.540,P<0.0001)and cel

Effects of geometrical characteristics on defect distributions in alloy components produced by selective laser melting

Selective laser melting(SLM)has been applied to manufacture various alloy components with excellent properties,but its further application is restricted by the intrinsic defects.In this work,the internal defect distributions in samples of three alloys(316L stainless steel,AlSi10Mg and Inconel 718)were investigated respectively,considering the effects of geometrical characteristics of the samples.The defects in the 316L stainless steel sample tend to be formed densely in the central part with large wall thickness,indicating a strong sensitivity to heat accumulation.Contrarily,the Inconel 718 sample shows a higher relative density with homogeneous defect distribution,indicating better formability for the SLM process.For the AlSi10Mg sample,the defect density keeps increasing as the deposition goes on.Typically,the defect density located at sample edges shows an abnormally high level comparing with the inner part,especially in the top sections of AlSi10Mg and Inconel 718 samples.The results are helpful for the geometrical design,the adjustment of building orientation and the further optimization of process parameters in the SLM process.

Dynamic stress concentrations in thick plates with two holes based on refined theory

Based on complex variables and conformal mapping, the elastic wave scat- tering and dynamic stress concentrations in the plates with two holes are studied by the refined dynamic equation of plate bending. The problem to be solved is changed to a set of infinite algebraic equations by an orthogonM function expansion method. As examples, under free boundary conditions, the numerical results of the dynamic moment concen- tration factors in the plates with two circular holes are computed. The results indicate that the parameters such as the incident wave number, the thickness of plates, and the spacing between holes have great effects on the dynamic stress distributions. The results are accurate because the refined equation is derived without any engineering hypothese.

Glial cell line-derived neurotrophic factor(GDNF)plays an important role in the protection of dopaminergic neurons,but there are few reports of the relationship between GDNF and its precursors(α-pro-GDNF andβ-pro-GDNF)and cognitive impairment in Parkinson’s disease.This study aimed to investigate the relationship between the serum levels of GDNF and its precursors and cognitive impairment in Parkinson’s disease,and to assess their potential as a diagnostic marker.Fifty-three primary outpatients and hospitalized patients with Parkinson’s disease(23 men and 30 women)with an average age of 66.58 years were enrolled from the Affiliated Hospital of Xuzhou Medical University of China in this case-control study.The patients were divided into the Parkinson’s disease with cognitive impairment group(n=27)and the Parkinson’s disease with normal cognitive function group(n=26)based on their Mini-Mental State Examination,Montreal Cognitive Assessment,and Clinical Dementia Rating scores.In addition,26 age-and sex-matched healthy subjects were included as the healthy control group.Results demonstrated that serum GDNF levels were significantly higher in the Parkinson’s disease with normal cognitive function group than in the other two groups.There were no significant differences in GDNF precursor levels among the three groups.Correlation analysis revealed that serum GDNF levels,GDNF/α-pro-GDNF ratios,and GDNF/β-pro-GDNF ratios were moderately or highly correlated with the Mini-Mental State Examination,Montreal Cognitive Assessment,and Clinical Dementia Rating scores.To explore the risk factors for cognitive impairment in patients with Parkinson’s disease,logistic regression analysis and stepwise linear regression analysis were performed.Both GDNF levels and Hoehn-Yahr stage were risk factors for cognitive impairment in Parkinson’s disease,and were the common influencing factors for cognitive scale scores.Neitherα-pro-GDNF norβ-pro-GDNF was risk factors for cognitive impairment in Parkinson’s disease.A receiver operating character

学界关于《礼记·玉藻》“君赐车马”句的读法、释义主要有两种观点。一种以郑玄为代表,认为“拜”“赐”二字当分读,后一句言卿大夫受赐于天子,归必致于其君,待君有命才能使用;另一种以陆佃为代表,认为“拜”“赐”当合读,后一句承接前一句,言没有国君之命则不敢乘服。经考辨,这两句是统言“三赐”之礼,陆氏的读法更符合经文语境,但他对第二句的释义不甚清晰,应释为:若没有得到国君的正式授命,则不可以立即乘服。关于“据掌致诸地”,多数学者认为是解释“稽首”之仪,但实属赘述;段玉裁等人认为是臣受君面赐所行之礼。经考察,臣受君面赐确实应先稽首然后受所赐之物,则“据掌致诸地”应释为:臣稽首而后双手捧所赐之物于地上。

目的了解和掌握山东省市、县级疾病预防控制中心(疾控中心)媒介生物防制能力现状,为积极应对媒介生物传染病防控新形势,提升山东省媒介生物防制能力提供依据。方法通过现场访谈和问卷调查的形式,收集山东省市、县两级疾控中心媒介生物防制专业技术人员基本信息及实验室配置等资料,采用WPS 2010和SPSS 19.0软件进行统计分析。计量资料的组间比较采用t检验或Wilcoxon秩和检验,计数资料组间比较采用χ^(2)检验。结果山东省市、县级疾控中心从事媒介生物监测与控制专职人员均数分别为4.10和1.94人。职称以初、中级为主,占比分别为34.43%和36.79%;预防医学(公共卫生)专业占比46.70%(99/212);工作年限以1~5年为主,占比58.49%。工作内容以开展媒介生物生态学监测为主,绝大部分媒介生物实验室仅能开展媒介生物分类鉴定;县级疾控中心培训占比59.38%,年均学术论文和科研成果量分别为5.2篇和0.8项。结论为应对媒介生物防制面临新挑战,各地需要加大经费投入,加强人才和技术储备,强化科研培训。专业技术人员要立足现有各监测项目,加强数据分析利用;做好媒介生物防制科普知识宣传,不断提升社会服务水平。
