Hypoxia promotes proliferation and differentiation of neural stem cells from embryonic day 12 rat brain tissue, but the concentration and time of hypoxic preconditioning are controversial. To address this, we cultured neural stem cells isolated from embryonic day 14 rat cerebral cortex in 5% and 10% oxygen in vitro. MTT assay, neurosphere number, and immunofluorescent staining found that 5% or 10% oxygen preconditioning for 72 hours improved neural stem cell viability and proliferation. With prolonged hypoxic duration （120 hours）, the proportion of apoptotic cells increased. Thus, 5% oxygen preconditioning for 72 hours promotes neural stem cell prolif- eration and neuronal differentiation. Our findings indicate that the optimal concentration and duration of hypoxic preconditioning for promoting proliferation and differentiation of neural stem cells from the cerebral cortex are 5% oxygen for 72 hours.

Crystalline carbon nanowire arrays were fabricated taking advantage of near-field electrospinning and stress decyanation.A novel fabrication method for carbon nanowires with radii ranging from~2.15μm down to~25 nm was developed based on implementing nitrogen pretreatment on the silica surface and then aligning polymer nanofibers during near-field electrospinning at an ultralow voltage.Stress decyanation was implemented by subsequently pyrolyzing a polymer nanofiber array on the silica surface at 1000°C for 1 h in an N_(2)atmosphere,thus obtaining a crystalline carbon nanowire array with a nanostructured surface.Various crystalline nanostructures were fabricated on the nanowire surface,and their electrochemical performance was evaluated by cyclic voltammetry(CV)and electrochemical impedance spectroscopy(EIS).Crystalline carbon wires with diameters ranging from micrometers to submicrometers displayed carbon nanoelectrode-like behavior with their CV curve having a sigmoidal shape.A highly crystalline carbon nanowire array showed distinct behavior,having a monotonically increasing straight line as its CV curve and a semicircular EIS spectrum;these results demonstrated its ultrastable current,as determined by electron transfer.Furthermore,nanocrystalline-structured carbon wires with diameters of~305 nm displayed at least a fourfold higher peak current density during CV(4000 ma/m2)than highly crystalline carbon nanowires with diameters of~100 nm and porous microwires with diameters of~4.3μm.

目的观察耳内镜下鼓膜置管联合左氧氟沙星治疗分泌性中耳炎的疗效。方法选取我院2012年3月;014年6月收治的分泌性中耳炎患者66例作为研究对象,并对其临床资料进行详细分析,随机分成观察组和对照组,各33例。观察组采用耳内镜鼓膜置管联合左氧氟沙星滴耳液治疗;对照组仅采用耳内镜鼓膜置管治疗。对比分析两组治疗总有效率以及听觉功能恢复情况。结果观察组治疗总有效率为90.91%,对照组为72.73%,差异有统计学意义（P<0.05）;治疗后观察组患者听阈比较、耳峰压值及静态声顺值比较,观察组明显优于对照组,差异有统计学意义（P<0.05）。结论耳内镜鼓膜置管联合左氧氟沙星滴耳液治疗分泌性中耳炎较单独使用耳内镜下鼓膜置管治疗的疗效更确切,临床使用价值大。

Comparative gene identification-58(CGI-58),also known asα/βhydrolase domain containing 5,is the co-activator of adipose triglyceride lipase that hydrolyzes triglycerides stored in the cytosolic lipid droplets.Mutations in CGI-58 gene cause Chanarin-Dorfman syndrome(CDS),an autosomal recessive neutral lipid storage disease with ichthyosis.The liver pathology of CDS manifests as steatosis and steatohepatitis,which currently has no effective treatments.Perilipin-3(Plin3)is a member of the Perilipin-ADRP-TIP47 protein family that is essential for lipid droplet biogenesis.The objective of this study was to test a hypothesis that deletion of a major lipid droplet protein alleviates fatty liver pathogenesis caused by CGI-58 deficiency in hepatocytes.Adult CGI-58-floxed mice were injected with adeno-associated vectors simultaneously expressing the Cre recombinase and microRNA against Plin3 under the control of a hepatocyte-specific promoter,followed by high-fat diet feeding for 6 weeks.Liver and blood samples were then collected from these animals for histological and biochemical analysis.Plin3 knockdown in hepatocytes prevented steatosis,steatohepatitis,and necroptosis caused by hepatocyte CGI-58 deficiency.Our work is the first to show that inhibiting Plin3 in hepatocytes is sufficient to mitigate hepatocyte CGI-58 deficiency-induced hepatic steatosis and steatohepatitis in mice.

To produce a three-dimensional micro/nanocarbon structure,a manufacturing design technique for sub-10 nm carbon fiber arrays on three-dimensional carbon micropillars has been developed;the method involves initiating electrostatic jetting,forming submicron-to-nanoscale PAN-based fibers,and maximizing the shrinkage from polyacrylonitrile(PAN)-based fibers to carbon fibers.Nanoforming and nanodepositing methods for polyacrylonitrilebased jet fibers as precursors of carbon fibers are proposed for the processing design of electrostatic jet initiation and for the forming design of submicron-to-nanoscale PAN-based fibers by establishing and analyzing mathematical models that include the diameter and tensile stress values of jet fibers and the electric field intensity values on the surfaces of carbon micropillars.In accordance with these methods,an array of jet fibers with diameters of~80 nm is experimentally formed based on the thinning of the electrospinning fluid on top of a dispensing needle,the poking of drum into an electrospinning droplet,and the controlling of the needle–drum distance.When converting thin PANbased jet fibers to carbon fibers,a pyrolysis method consisting of the suspension of jet nanofibers between carbon micropillars,the bond between the fibers and the surface of the carbon micropillar,and the control of micropillar spacing,stabilization temperature,and carbonation rate is presented to maximize the shrinkage from PAN-based fibers to carbon fibers and to form sub-10 nm carbon fiber arrays between three-dimensional carbon micropillars.The manufacturing design of a three-dimensional micro/nanocarbon structure can produce thin PAN-based jet nanofibers and nanofiber arrays aligned on micropillar surfaces,obtain shrinkage levels reaching 96%and incorporate sub-10 nm carbon fibers into three-dimensional carbon micropillars;these actions provide new research opportunities for correlated three-dimensional micro/nanocarbon structures that have not previously been technically possible.

The development of large-scale time-domain surveys provides an opportunity to study the physical properties as well as the evolutionary scenario of B-type subdwarfs(sdBs)and M-type dwarfs(dMs).Here,we obtained 33 sdB+dM eclipsing binaries based on the Zwicky Transient Facility(ZTF)light curves and Gaia Early Data Release3(EDR3)parallaxes.By using the PHOEBE code for light curve analysis,we obtain probability distributions for parameters of 29 sdB+dMs.R1,R2 and i are well determined,and the average uncertainty of mass ratio q is 0.08.Our parameters are in good agreement with previous works if a typical mass of sdB is assumed.Based on parameters of 29 sdB+dMs,we find that both the mass ratio q and the companion’s radius R2 decrease with the shortening of the orbital period.For the three sdB+dMs with orbital periods less than 0.075 days,their companions are all brown dwarfs.The masses and radii of the companions satisfy the mass–radius relation for low-mass stars and brown dwarfs.Companions with radii between 0.12 R_(⊙)and 0.15 R_(⊙)seem to be missing in the observations.As more short-period sdB+dM eclipsing binaries are discovered and classified in the future with ZTF and Gaia,we will have more information to constrain the evolutionary ending of sdB+dMs.

BACKGROUND Horseshoe kidney(HK)with renal stones is challenging for urologists.Although both retroperitoneal and transperitoneal laparoscopic approaches have been reported in some case reports,the therapeutic outcome of retroperitoneal compared with transperitoneal laparoscopic lithotripsy is unknown.AIM To assess the efficacy of laparoscopic lithotripsy for renal stones in patients with HK.METHODS This was a retrospective study of 12 patients with HK and a limited number(n≤3)of 20-40 mm renal stones treated with either retroperitoneal or transperitoneal laparoscopic lithotripsy(June 2012 to may 2019).The perioperative data of both groups were compared including operation time,estimated blood loss,postoperative fasting time,perioperative complications and stone-free rate(SFR).RESULTS No significant difference was observed for age,gender,preoperative symptoms,body mass index,preoperative infection,hydronephrosis degree,largest stone diameter,stone number and isthmus thickness.The mean postoperative fasting time of the patients in the retroperitoneal group and the transperitoneal group was 1.29±0.49 and 2.40±0.89 d,respectively(P=0.019).There was no significant difference in operation time(194.29±102.48 min vs 151.40±39.54 min,P=0.399),estimated blood loss(48.57±31.85 m L vs 72.00±41.47 m L,P=0.292)and length of hospital stay(12.14±2.61 d vs 12.40±3.21 d,P=0.881)between the retroperitoneal and transperitoneal groups.All patients in both groups had a complete SFR and postoperative renal function was within the normal range.The change in estimated glomerular filtration rate(e GFR)from the preoperative stage to postoperative day 1 in the retroperitoneal group and the transperitoneal group was-3.86±0.69 and-2.20±2.17 m L/(min·1.73 m2),respectively(P=0.176).From the preoperative stage to the 3-mo follow-up,the absolute change in e GFR values for patients in the retroperitoneal group and the transperitoneal group was-3.29±1.11 and-2.40±2.07 m L/(min·1.73 m2),respectively(P=0.581).CONCLUSION Both retroperitoneal and trans

Post-stroke depression(PSD)is a serious and common complication of stroke,which seriously afects the rehabilitation of stroke patients.To date,the pathogenesis of PSD is unclear and efective treatments remain unavailable.Here,we established a mouse model of PSD through photothrombosis-induced focal ischemia.By using a combination of brain imaging,transcriptome sequencing,and bioinformatics analysis,we found that the hippocampus of PSD mice had a signifcantly lower metabolic level than other brain regions.RNA sequencing revealed a signifcant reduction of miR34b-3p,which was expressed in hippocampal neurons and inhibited the translation of eukaryotic translation initiation factor 4E(eIF4E).Furthermore,silencing eIF4E inactivated microglia,inhibited neuroinfammation,and abolished the depression-like behaviors in PSD mice.Together,our data demonstrated that insufcient miR34b-3p after stroke cannot inhibit eIF4E translation,which causes PSD by the activation of microglia in the hippocampus.Therefore,miR34b-3p and eIF4E may serve as potential therapeutic targets for the treatment of PSD.

The positive regulation of bone-forming osteoblast activity and the negative feedback regulation of osteoclastic activity are equally important in strategies to achieve successful alveolar bone regeneration. Here, a molybdenum(Mo)-containing bioactive glass ceramic scaffold with solid-strut-packed structures(Mo-scaffold) was printed, and its ability to regulate pro-osteogenic and antiosteoclastogenic cellular responses was evaluated in vitro and in vivo. We found that extracts derived from Mo-scaffold(Moextracts) strongly stimulated osteogenic differentiation of bone marrow mesenchymal stem cells and inhibited differentiation of osteoclast progenitors. The identified comodulatory effect was further demonstrated to arise from Mo ions in the Mo-extract,wherein Mo ions suppressed osteoclastic differentiation by scavenging reactive oxygen species(ROS) and inhibiting mitochondrial biogenesis in osteoclasts. Consistent with the in vitro findings, the Mo-scaffold was found to significantly promote osteoblastmediated bone formation and inhibit osteoclast-mediated bone resorption throughout the bone healing process, leading to enhanced bone regeneration. In combination with our previous finding that Mo ions participate in material-mediated immunomodulation, this study offers the new insight that Mo ions facilitate bone repair by comodulating the balance between bone formation and resorption. Our findings suggest that Mo ions are multifunctional cellular modulators that can potentially be used in biomaterial design and bone tissue engineering.

Various uncertainties arising during acquisition process of geoscience data may result in anomalous data instances(i.e.,outliers)that do not conform with the expected pattern of regular data instances.With sparse multivariate data obtained from geotechnical site investigation,it is impossible to identify outliers with certainty due to the distortion of statistics of geotechnical parameters caused by outliers and their associated statistical uncertainty resulted from data sparsity.This paper develops a probabilistic outlier detection method for sparse multivariate data obtained from geotechnical site investigation.The proposed approach quantifies the outlying probability of each data instance based on mahalanobis distance and determines outliers as those data instances with outlying probabilities greater than 0.5.It tackles the distortion issue of statistics estimated from the dataset with outliers by a re-sampling technique and accounts,rationally,for the statistical uncertainty by Bayesian machine learning.Moreover,the proposed approach also suggests an exclusive method to determine outlying components of each outlier.The proposed approach is illustrated and verified using simulated and real-life dataset.It showed that the proposed approach properly identifies outliers among sparse multivariate data and their corresponding outlying components in a probabilistic manner.It can significantly reduce the masking effect(i.e.,missing some actual outliers due to the distortion of statistics by the outliers and statistical uncertainty).It also found that outliers among sparse multivariate data instances affect significantly the construction of multivariate distribution of geotechnical parameters for uncertainty quantification.This emphasizes the necessity of data cleaning process(e.g.,outlier detection)for uncertainty quantification based on geoscience data.
