The present work is to determine the concentration of norepinephrine (NE) in rat hypothalamus by linear sweep voltammetry (LSV) using the ultramicrocarbon fiber electrode for study of acupuncture analgesia *** were performed on 220--250g wistar albino rats. After therat was put on the operation table inserting the fiber electrod into the HVMand other neighbouring nuclei of the rat's brain, then we determined directlythe concentration of NE, in order to observe the change of norepinephrineconcentration before and after electroacupuncture. Vivo is determined every 5minutes for keeping the fiber electrode stablize.

The mechanism of methanol adsorption induced by oxygen and the effect of water on the methanol oxidation on electrolytic silver have been studied by ultra-high vacuum temperature programmed reaction spectroscopy(TPRS), transient response method and isotopic exchange experiment. It has been found that oxygen adsorbed on silver can greatly promote methanol adsorption and can also react with methanol to produce water. Experimental results show that oxygen in the water comes from the adsorbed oxygen and hydrogen from the methyl and the hydroxyl in methanol. It has also been found that there exists competitive adsorption between water and oxygen on silver and water can increase the selectivity of methanol oxidation to formaldehyde. This result is consistent with the catalytic activities.

To study the transcriptional regulation of urokinase receptor (uPAR) in high- (95D) and low-metastatic (95C) human lung cancer cells, we performed PCR to amplify 2238 bp uPAR promoter from 95C and 95D cells. According to the results of sequencing, five different bases are found in uPAR promoter between 95C and 95D cells. The results of luciferase activity assay showed that these differences have no significant effect on the uPAR promoter activity. Based on a normal uPAR promoter, progressive truncated mutants were constructed. The transient transfection/reporter assay showed that the promoter region from –136 to +9 may interact with relevant nuclear factors, which result in different levels of uPAR expression between 95C and 95D cells.

It is well documented that the glycosylation of E-cadherin is correlated with cancer metastasis, but whether E- cadherin could be core fucosylated remains largely unknown. We found that E-cadherin was core fucosylated in highly metastatic lung cancer cells while absent in lowly metastatic lung cancer cells. Since α-1,6 Fucosyltransferase (α-1,6 FucT) is known to catalyze the reaction of core fucosylation, we investigated the biological function of core fucosylation on E-cadherin by α-1,6 FucT targeted RNAi and transfecting α-1,6 FucT expression vector. As a result, calcium dependent cell-cell adhesion mediated by E-cadherin was strengthened with the reduction of core fucosylation on E- cadherin after RNAi and was weakened with the elevated core fucosylation on E-cadherin after α-1,6 FucT over expression. Our data indicated that α-1,6 FucT could regulate E-cadherin mediated cell adhesion and thus play an important role in cancer development and progression. Computer modeling showed that core fucosylation on E-cadherin could significantly impair three-dimensional conformation of N-glycan on E-cadherin and produce conformational asym- metry so as to suppress the function of E-cadherin. Furthermore, the relationship between the expression of core fucosylated E-cadherin and clinicopathological background of lung cancer patients was explored in lung cancer tissue of patients. It turns out to demonstrate that core fucosylated E-cadherin could serve as a promising prognostic indicator for lung cancer patients.

The low-abundantly expressed genes composed the majorities of the mRNAs expressed in the central nervous system (CNS), and were thought to be important for the normal brain functions. Through differential screening a low-abundance cDNA sublibrary with mRNA from neuropathic pain of chronic constriction injury (CCI) model, we have identified a novel rat gene, rat spinal-cord expression protein 4 gene (RSEP4). The total length of RSEP4 cDNA is 2006 bp, with a 501 nucleotide open reading frame (ORF) that encodes a 167 amino acid polypeptide. Northern blot revealed that RSEP4 was expressed specifically in the CNS. In situ hybridization showed that the mRNA of RSEP4 was strongly expressed in the CA1, CA2, CA3 and DG regions of hippocampus, the Purkinje cells of cerebellum, and the small sensory neurons of dorsal horn and large motor neurons of ventral horn of spinal cord. Over-expression of RSEP4-EGFP fusion protein in the human embryonic kidney 293T cells showed that RSEP4 protein was mainly localized in the cell cytoplasm. These results suggest that RSEP4 may play some roles in the CNS.

Aligned Solidification Structure of the MnBi Phase in Semisolidified Bi-Mn Alloy with a Static Magnetic Field

The solidification structure of Bi-3 wt pct Mn alloy grown up in the semisolid zone under the influence of a staticmagnetic field (up to 1.0 T) and the relation of the magnetic property with the solidification structure have beeninvestigated experimentally. It was shown that the primary phase MnBi crystals in the alloy aligned and oriented alongthe direction of the applied magnetic field. The orientating tendency and the average length of the elongated MnBicrystals increased with the increase of the applied field and the solidification time. Moreover, the remanence of thealloy along the aligned direction of the MnBi phase in the case of solidification with a magnetic field was apparentlyanisotropic and nearly double of that without the magnetic field. This indicated that the MnBi crystals orientedand aligned along their easy magnetization axis. A model was proposed to explain the alignment and orientationgrowth of the MnBi crystals in a magnetic field in terms of the magnetic anisotropy of the crystals and the magneticinteraction between them.

The adsorption and desorption of CO on the Ag-rich and Pd-rich surface of Ag-Pd al-loy have been investigated with work function measurement, Auger spectroscopy （AES）,and thermal desorption spectroscopy （TDS） in an ultrahigh vacuum system, *** showed that the surface of the alloy (X***=40% at.) was rich in Ag after an-ncaling as would normally be expected and was rich in Pd after Ar+ bombardment possiblycaused by different sputtering rate on account of their different surface binding *** of CO the work functions of both surfaces （Pd-rich and Ag-rich） are raised. Theresults of AES and work function measurement confirmed that the Ag-rich alloy becamecnriched with Pd upon prolonged contact with CO at 293 K. With TDS, one singledesorption peak of adsorbed CO was observed throughout the whole range of exposure forthe Ag-Pd system. This result implied that Pd atom in the surface were acting as individualchemisorption centers and that the Ag atoms were merely acting as a

Preparation and characterization of some surface negatively charged residue mutants of cytochrome b_5

Site-directed mutagenesis was used to obtain seven variants of tryptic fragment of bovine liver cytochrome bs (cyt bs), in which the negatively charged residues around the heme exposed edge of cyt bs were replaced by hydropho-bic amino acid alanine. Double-site mutants, triple-site mutants and even quadruple-site mutants were obtained. DNA sequencing and molecular weight measurements of the mutant proteins both confirmed that these site-directed muta-genesises were successfully performed. Spectroelectrochem-istry of these mutant proteins revealed that the apparent redox potentials of these mutant proteins caused a positive shift of 2-10 mV. The global structure of these mutant proteins did not show much difference from that of the wild type cyt bs, providing a solid base for the further study on the roles of the proteins’ surface charges.

The conflation of geographic datasets is one of the key technologies in the realm of spatial data capture and integration in geographic information system (GIS). Map conflation is a complex process of matching and merging spatial data. Due to various reasons such as errors in original data related to map data discrepancies,a great amount of uncertainties exists during the process and this will result in errors in featuring matching,especially point feature. Thus,it is vital to develop the method to detect the errors in feature matching and further the conflation results will not be affected. In this paper,error matching detection and robust estimation adjustment methods are proposed for map conflation. The characteristics of errors in feature matching are first analyzed,then a new approach for map conflation based on the least-squares adjustment is presented,and a robust estimation adjustment method is further proposed to detect and process matching errors. The results of the map conflation test show that the proposed method not only determines the errors in feature matching,but also obtains the optimal merging results in map conflation.
