clostridioides difficile(***)is the most common pathogen causing health care-associated *** TcdA and TcdB have been shown to activate enteric neurons;however,what population of these cells is more profoundly influenced and the mechanism underlying these effects remain *** To characterize a specific population of TcdA-affected myenteric neurons and investigate the role of the P2X7 receptor in TcdA-induced ileal inflammation,cell death,and the changes in the enteric nervous system in *** swiss mice were used to model TcdA-induced ileitis in ileal loops exposed to TcdA(50μg/Loop)for 4 *** investigate the role of the P2X7 receptor,Brilliant Blue G(50 mg/kg,i.p.),which is a nonspecific P2X7 receptor antagonist,or A438079(0.7μg/mouse,i.p.),which is a competitive P2X7 receptor antagonist,were injected one hour prior to TcdA *** samples were collected to analyze the expression of the P2X7 receptor(by quantitative real-time polymerase chain reaction and immunohistochemistry),the population of myenteric enteric neurons(immunofluorescence),histological damage,intestinal inflammation,cell death(terminal deoxynucleotidyltransferasemediated dUTP-biotin nick end labeling),neuronal loss,and s100B synthesis(immunohistochemistry).REsULTs TcdA upregulated(Pssion of the P2X7 receptor gene in the ileal tissues,increasing the level of this receptor in myenteric neurons compared to that in control *** with the control mice indicated that TcdA promoted(Pss of myenteric calretinin+(calr)and choline acetyltransferase+neurons and increased the number of nitrergic+and calr+neurons expressing the P2X7 *** of the P2X7 receptor decreased TcdAinduced intestinal damage,cytokine release[interleukin(IL)-1β,IL-6,IL-8,and tumor necrosis factor-α],cell death,enteric neuron loss,and s100B synthesis in the mouse *** Our findings demonstrated that TcdA induced the upregulation of the P

Interaction of pulsed transversely excited atmospheric （TEA） cO2-1aser radiation at 10.6 μm with nanoporous activated carbon cloth was investigated. Activated carbon cloth of different adsorption characteristics was used. Activated carbon cloth modifications were initiated by laser pulse intensities from 0.5 to 28 MW/cm^2, depending on the cloth adsorption characteristics. cO2 laser radiation was effectively absorbed by the used activated carbon cloth and largely converted into thermal energy. The type of modification depended on laser power density, number of pulses, but mostly on material characteristics such as specific surface area. The higher the surface area of activated carbon cloth, the higher the damage threshold.

Unravelling the competitive effect of microstructural features on the fracture toughness and tensile properties of near beta titanium alloys

The competitive effect of microstructural features including primaryα(α_(p)),secondaryα(α_(s)),grain boundaryα(α_(GB)) and β grain size on mechanical properties of a near β Ti alloy were studied with two heat treatment *** relative effect of β grain size and sta(solution treatment and ageing)processing parameters on mechanical properties were quantitatively explored by the application of Taguchi *** results were further explained via correlating microstructure with the fracture toughness and tensile *** was found that large numbers of fine as precipitates and continuous α_(s) played greater roles than other features,resulting in a high strength and very low ductility(sta process *** β grain size had a negative correlation with fracture *** the samples prepared by BAscA( β anneal slow cooling and ageing)process,improved ductility and fracture toughness were obtained due to a lower density ofα;precipitates,a basket-weave structure and zigzag morphology of α_(GB).For this heat treatment,an increase in prior β grain size had an observable positive effect on fracture *** contradictory effect of β grain size on fracture toughness found in literature was for the first time *** was shown that the microstructure obtained from different processes after β solution has complex effect on mechanical *** complexity derived from the competition between microstructure features and the overall sum of their effect on fracture toughness and tensile properties.A novel table was proposed to quasi-quantitatively unravel these competitive effects.

Objective:To identify the prevalence of soil-transmitted helminthiasis(sTH)among school-age children in the cagayan valley,the Philippines,assess their level of awareness on the disease,and determine predisposing factors of the ***:A total of 478 GradesⅢ-Ⅴschool-age children in Pamplona and sanchez-Mira school Districts in the cagayan valley answered the questionnaire assessing their knowledge,attitude,and practices on sTH,subjected to anthropometric measurements,and provided faecal samples for parasitologic assessment(direct smear,Kato-Katz,and formol-ether concentration techniques).Results:The participants of the study,with 55.86%females,were 8 to 14 years *** nutritional status was assessed‘normal’(84.31%),‘severely wasted’(6.49%),‘wasted’(5.23%),‘overweight’(2.72%),and‘obese’(1.26%).The prevalence of infection with at least 1 sTH species was 25.99%in Pamplona and 19.40%in ***,the prevalence of heavy intensity was 7.11%for Ascaris lumbricoides and 1.67%for Trichuris *** hookworm infections had light *** majority of the school-age children had a low score in the KAP *** knowledge of sTH,‘stunted growth as a symptom of infection’was associated with a lower risk of Ascaris lumbricoides infection(OR 0.448;95%cI 0.212,0.945;P=0.035)while‘playing with soil as a mode of transmission’was associated with an increased risk of Ascaris lumbricoides infection(OR 2.067;95%cI 1.014,4.212;P=0.046).In attitude towards sTH,‘I think I have intestinal worm now’was associated with a higher risk of Ascaris lumbricoides infection(OR 1.681;95%cI 1.061,2.662;P=0.027).conclusions:The prevalence rate of Ascaris lumbricoides among the school-age children in the cagayan valley shows the need to further intensify intervention in the area to meet the threshold set by the World Health *** identified predictors of infection,which concerns the school-age children’s knowledge and attitude toward sTH,can be used in augmenting

context: There is no established pharmacological treatment for the core sympto ms of chronic fatigue syndrome cORE JOURNALs IN cLINIcAL NEUROLOGY cORE vIEW (cF s). Galantamine hydrobromide, an acetyl cholesterone inhibitor, has pharmacologi cal properties that might benefit patients with cFs. Objective: To compare the e fficacy and tolerability of galantamine hydrobromide in patients with cFs. Desig n, setting, and Patients: Randomized, double blind trial conducted June 1997 th rough July 1999 at 35 outpatient centers in the United Kingdom(n=17), United sta tes (n=14), the Netherlands (n=2), sweden (n=1), and Belgium (n=1) involving 434 patients with a clinical diagnosis of cFs (modified Us centers for Disease cont rol and Prevention criteria). Interventions: A total of 89 patients were randoml y assigned to receive 2.5 mg of galantamine hydrobromide; 86 patients, 5.0 mg; 9 1 patients, 7.5 mg; and,86 patients, 10 mg (these patients received medicine in the tablet form 3 times per day); a total of 82 patients received matching place bo tablets 3 times per day. Main Outcome Measures: The primary efficacy variable was the global change on the clinician Global Impression scale after 4, 8, 12, and 16 weeks of treatment. secondary outcomes were changes in core symptoms of cFs on the chalder Fatigue Rating scale, the Fibromyalgia Impact Q uestionnaire, and the Pittsburgh sleep Quality Index; changes in quality of life on the Nottingham Health Profile; and assessment of plasma free cortisol level s and cognitive performance on a computer based battery of tests. Results: Afte r 16 weeks, there were no statistically significant differences between any of t he galantamine or placebo groups in clinical condition on the clinician Global I mpression scale, or for any of the secondary end points. Fracture energy evaluation on 7075-T651 aluminum alloy welds determined by instrumented impact pendulum

By using an instrumented impact pendulum, the force versus time curves of 7075-T651 aluminum welds were obtained from standard charpy-v samples. considering the force-time curves and constant impact velocity, the fracture energies for different zones were quantified. A fracture energy improvement for the HAZ(33.6 J) was observed in comparison with the weld metal(7.88 J), and base metal(5.37 J and 7.37 J for longitudinal and transverse directions, respectively). This toughness increment was attributed to the microstructural transformation caused by the thermodynamic instability of η′ precipitates during the welding. Fracture energy for weld metal was higher than that for base metal, probably due to pores created during solidification. Regarding the dynamic yielding force obtained from the force-time curves, an approximation to the dynamic yield strength for weld, HAZ and base metal was determined. Fracture surfaces revealed an intergranular failure for base metal in longitudinal direction, whereas a predominately brittle failure(cleavage) with some insights of ductile characteristics was observed for the transverse direction. In contrast, a ductile failure was observed for weld metal and HAZ.

The use of tyrosine kinase inhibitors (TKIs) has resulted in successful treatm ent of KIT-positive neoplasms. carcinosarcoma is a very aggressive neoplasm. co nsequently, c-kit expression may have significant clinical implications for thi s tumor. The purpose of the present study was to assess ckit expression in the c arcinomatous and sarcomatous element of carcinosarcoma to identify if KIT repres ents a therapeutic target for treatment of this neoplasm. Immunohistochemical st aining for c-KIT was performed on paraffin-embedded tissue blocks of 20 consec utive uterine specimens with carcinosarcoma, 40 with endometrial carcinoma, and 12 with atrophic endometrium. Two pathologists assessed the scoring index of sta ining. In the stromal element of carcinosarcoma, no immunohistochemically c-KIT stained cells were observed and in the epithelial element, a low scoring index of staining was present. In endometrial endometrioid carcinoma, the scoring inde x was high in only 15%of the specimens. In the atrophic endometrium, a low scor ing index was seen in all specimens. Our results seem to suggest that c-kit pro bably plays no major role in the pathogenesis of the majority of these tumors wh ile it may be involved in the pathogenesis of some endometrial carcinomas. In vi ew of the multiple molecular targets that are activated by imatinib mesylate, no definitive conclusions can be drawn with regard its effectiveness in carcinosar comas based on the present study. Further studies of c-kit expression in larger series of carcinosarcomas in order to settle the controversial issues with rega rd to frequency of expression, prognostic, and clinical value are warranted.
