Assessment of the binding interactions of SARS-CoV-2 spike glycoprotein variants

Severe acute respiratory syndrome-associated coronavirus 2 is a major global health issue and is driving the need for new *** surface spike protein,which plays a central role in virus infection,is currently the target for vaccines and neutralizing *** emergence of novel variants with multiple mutations in the spike protein may reduce the effectiveness of neutralizing antibodies by altering the binding activity of the protein with angiotensin-converting enzyme 2(ACE2).To understand the impact of spike protein mutations on the binding interactions required for virus infection and the effectiveness of neutralizing monoclonal antibody(mAb)therapies,the binding activities of the original spike protein receptor binding domain(RBD)sequence and the reported spike protein variants were investigated using surface plasmon *** addition,the interactions of the ACE2 receptor,an antispike mAb(mAb1),a neutralizing mAb(mAb2),the original spike RBD sequence,and mutants D614G,N501Y,N439K,Y453F,and E484K were *** to the original RBD,the Y453F and N501Y mutants displayed a significant increase in ACE2 binding affinity,whereas D614G had a substantial reduction in binding *** mAb-RBD mutant proteins displayed a reduction in binding affinities relative to the original RBD,except for the E484K-mAb1 *** potential neutralizing capability of mAb1 and mAb2 was ***,mAb1 failed to inhibit the ACE2-RBD interaction while mAb2 inhibited the ACE2-RBD interactions for all RBD mutants,except mutant E484K,which only displayed partial blocking.

Automatic recognition of human emotions in a continuous dialog model remains challenging where a speaker’s utterance includes several sentences that may not always carry a single *** work with standalone speech emotion recognition(SER)systems proposed for continuous speech only has been *** the recent decade,various effective SER systems have been proposed for discrete speech,i.e.,short speech *** would be more helpful if these systems could also recognize emotions from continuous ***,if these systems are applied directly to test emotions from continuous speech,emotion recognition performance would not be similar to that achieved for discrete speech due to the mismatch between training data(from training speech)and testing data(from continuous speech).The problem may possibly be resolved if an existing SER system for discrete speech is ***,in this work the author’s existing effective SER system for multilingual and mixed-lingual discrete speech is enhanced by enriching the cepstral speech feature set with bi-spectral speech features and a unique functional set of Mel frequency cepstral coefficient features derived from a sine filter *** augmentation is applied to combat skewness of the SER system toward certain *** using random forest is *** enhanced SER system is used to predict emotions from continuous speech with a uniform segmentation *** to data scarcity,several audio samples of discrete speech from the SAVEE database that has recordings in a universal language,i.e.,English,are concatenated resulting in multi-emotional speech ***,fear,sad,and neutral emotions,which are vital during the initial investigation of mentally disordered individuals,are selected to build six categories of multi-emotional *** results demonstrate the suitability of the proposed method for recognizing emotions from continuous speech as well as from discrete speech.

Dynamic Study of Effect of Low Dose Infliximab in Ankylosing Spondylitis

Objectives: To study the effect of low dose infliximab in patients with ankylosing spondylitis（AS） in3different aspects simultaneously. The first is clinical disease activity of AS measuredby Bath Ankylosing Spondylitis Disease Activity Index （BASDAI） and Bath AnkylosingSpondylitis Metrology Index （BASMI）, the second is its relationship with routine laboratoryparameters predicting the disease activity, like Erythrocyte Sedimentation Rate （ESR）, C-reactive protein （CRP）, platelet etc and the third is serum level of cytokines like TNF-α and IL-6. Method: Thirty-four patients with AS treated with infliximab were included in this longitudinalcohort study. All patients fulfilled the modified New York criteria for AS. Low dose infliximabwas given intravenously （3mg/kg body weight） at0,2,6and every8weeks thereafter for aperiod of22weeks. Clinical disease activity was assessed by BASDAI score and BASMI scoreand clinical improvement was recorded according to Bath Ankylosing Spondylitis DiseaseActivity Index20%（BASDAI20）, Bath Ankylosing Spondylitis Disease Activity Index50%（BASDAI50） and Bath Ankylosing Spondylitis Disease Activity Index70%（BASDAI70）.Laboratory measures like ESR, CRP, total platelet count and serum albumin were recorded ateach visit. Serum TNF-α level and IL-6were analyzed in13patients for4visits. Radiologicalassessment was done by computed tomography （CT） scan of sacro-iliac joint before the patientstarted therapy. Therapeutic safety of the drug was monitored at each visit by assessing completeblood count, liver function test and renal function test. Result: Thirty-four patients were enrolled in the study. The mean age of the patients was32.5years and mean weight was72.2kg.88.2%of the patients were male and11.8%patients werefemale.41.2%（14/34） of the patients were smoker. Dynamics and Small Signal Stability Analysis of PMSG-based Wind Farm with an MMC-HVDC System

The permanent magnet synchronous generator (PMSG)-based wind farm with a modular multilevel converter (MMC) based HVDC system exhibits various oscillations and can experience dynamic instability due to the interactions between different controllers of the wind farm and MMC stations, which have not been properly examined in the existing literatures. This paper presents a dynamic modeling approach for small signal stability analysis of PMSG-based wind farms with a MMC- HVDC system. The small signal model of the study system is validated by the comprehensive electromagnetic transient (EMT) simulations in PSCAD/EMTDC. Then the eigenvalue approach and participation factors analysis are utilized to comprehensively evaluate the impact of different controllers, system’s parameters and the circulating current suppressing controller (CCSC) on the small signal stability of the entire system. From eigenvalue analysis, it is revealed that as the output active power of the wind farm increases within the rated range, the overall system will exhibit a sub-synchronous oscillation (SSO) instability mode, an extremely weak damping mode, and a low frequency oscillation instability mode. From participation factors analysis, it is observed that the SSO mode and weak damping mode are primarily related to the internal dynamics of the MMC, which can be suppressed or improved by CCSC. It is determined that the low frequency oscillation mode is primarily caused by the interactions between the phase locked loop (PLL) control of the wind farm and the voltage and frequency (V-F) control of the MMC station. The analysis also depicts that the larger proportional gain value of the V-F control of the MMC station and smaller PLL bandwidth of the wind farm can enhance the small signal stability of the entire system.
