An injectable and coagulation-independent Tetra-PEG hydrogel bioadhesive for post-extraction hemostasis and alveolar bone regeneration

Effective control of post-extraction hemorrhage and alveolar bone resorption is critical for successful extraction socket treatment,which remains an unmet clinical ***,an injectable Tetra-PEG hydrogel that possesses rapid gelation,firm tissue adhesion,high mechanical strength,suitable degradability,and excellent biocompatibility is developed as a sutureless and coagulation-independent bioadhesive for the management of extraction *** results demonstrate that the rapid and robust adhesive sealing of the extraction socket by the Tetra-PEG hydrogel can provide reliable protection for the underlying wound and stabilize blood clots to facilitate tissue *** vivo experiments using an anticoagulated rat tooth extraction model show that the hydrogel significantly outperformed clinically used cotton and gelatin sponge in hemostatic efficacy,wound closure,alveolar ridge preservation,and in situ alveolar bone *** evaluations reveal the mechanisms for accelerated bone repair through suppressed long-term inflammation,elevated collagen deposition,higher osteoblast activity,and enhanced ***,our study highlights the clinical potential of the developed injectable Tetra-PEG hydrogel for treating anticoagulant-related post-extraction hemorrhage and improving socket healing.

CFD Investigations of Ship Maneuvering in Waves Using naoe-FOAM-SJTU Solver

Ship maneuvering in waves includes the performance of ship resistance, seakeeping, propulsion, and maneuverability. It is a complex hydrodynamic problem with the interaction of many factors. With the purpose of directly predicting the behavior of ship maneuvering in waves, a CFD solver named naoe-FOAM-SJTU is developed by the Computational Marine Hydrodynamics Lab(CMHL) in Shanghai Jiao Tong University. The solver is based on open source platform OpenFOAM and has introduced dynamic overset grid technology to handle complex ship hull-propeller-rudder motion system. Maneuvering control module based on feedback control mechanism is also developed to accurately simulate corresponding motion behavior of free running ship maneuver. Inlet boundary wavemaker and relaxation zone technique is used to generate desired waves. Based on the developed modules, unsteady Reynolds-averaged Navier-Stokes(RANS) computations are carried out for several validation cases of free running ship maneuver in waves including zigzag, turning circle, and course keeping maneuvers. The simulation results are compared with available benchmark data. Ship motions, trajectories, and other maneuvering parameters are consistent with available experimental data, which indicate that the present solver can be suitable and reliable in predicting the performance of ship maneuvering in waves. Flow visualizations, such as free surface elevation, wake flow, vortical structures, are presented to explain the hydrodynamic performance of ship maneuvering in waves. Large flow separation can be observed around propellers and rudders. It is concluded that RANS approach is not accurate enough for predicting ship maneuvering in waves with large flow separations and detached eddy simulation(DES) or large eddy simulation(LES) computations are required to improve the prediction accuracy.

Numerical Prediction of Wall Effect on Propeller in Restricted Channel

In restricted channel, the hydrodynamic performance of propeller is affected by the wall. In the present work, two cylindrical channels with different diameters being 1.8D and 5.0D are adopted to study the influence of wall on the hydrodynamic performance and wake field of the propeller model DTMB4119. The numerical simulations are carried out by the single-phase solver pimpleDyMFoam in open source platform OpenFOAM. The Reynolds Averaged Navier-Stokes equations (RANS) are adopted to solve the flow field. The arbitrary mesh interface (AMI) method is used to simulate the rotation of propeller. The designed advance ratio, J = 0.833, is applied in all the computations. For the 5.0 D case, the predicted results of open water performance are in good agreement with experiment data. In restricted channel, the predicted results of thrust and torque coefficients are larger than the open water case. The pressure on the wall of restricted channel downstream increases and approaches the results in open water gradually. Due to the flux conservation, higher negative induced velocity is investigated in the flow field of the propeller in restricted channel.

Effects of tacrolimus (FK506) on sciatic nerve regeneration in rats

BACKGROUND： Tacrolimus （FK506） protects peripheral nerves located in damaged regions by inhibiting T lymphocyte proliferation and activation. OBJECTIVE： To evaluate the effect of FK506 on promoting regeneration of rat sciatic nerve. DESIGN, TIME AND SETTING： A randomized, controlled, animal study was performed at the Laboratory of the Department of Orthopedic Surgery, Dalian Medical University, China, from September 2007 to September 2008. MATERIALS： A total of 60 adult, male, Sprague-Dawley rats were equally and randomly divided into model, local administration and systemic administration groups. All rats received a neurotomy of bilateral sciatic nerves to establish models of nerve regeneration chambers. The powder and injection of FK506 were supplied by Fujisawa Pharmaceutical, Japan. METHODS： The regeneration chambers of the model group were infused with 0.2 mL saline. The systemic group were injected with 0.2 mL saline, followed by daily subcutaneous injections of FK506 （1 mg/kg）, for 14 days. The local administration group was infused with 0.2 mL FK506 （1 μg/mL）. MAIN OUTCOME MEASURES： Local immune response was observed using hematoxylin-eosin staining. Myelinated nerve fiber number, myelin sheath and nerve fiber thickness were observed using toluidine blue staining. Wet weight of gastrocnemius was evaluated. Compound muscle action potential amplitude, latency, and conduction time were recorded, and motor nerve conduction velocity was calculated using electrophysiology. RESULTS： The total number of myelinated nerve fibers in the local and systemic administration groups was significantly higher than in the model group. The density of myelinated nerve fibers, myelin sheath thickness and mean axon diameter were significantly increased in the systemic administration group compared with the model group （P 〈 0.05）. An anti-senescence hydrogel with pH-responsive drug release for mitigating intervertebral disc degeneration and low back pain

Oxidative stress and aging lead to progressive senescence of nucleus pulposus(NP)cells,resulting in intervertebral disc(IVD)degeneration(IVDD).In some cases,degenerative IVD can further cause low back pain(LBP).Several studies have confirmed that delaying and rejuvenating the senescence of NP cells can attenuate ***,the relatively closed tissue structure of IVDs presents challenges for the local application of anti-senescence ***,we prepared an anti-senescence hydrogel by conjugating phenylboronic acid-modified gelatin methacryloyl(GP)with quercetin to alleviate IVDD by removing senescent NP *** hydrogel exhibited injectability,biodegradability,prominent biocompatibility and responsive release of quercetin under pathological *** vitro experiments demonstrated that the hydrogel could reduce the expression of senescence markers and restore the metabolic balance in senescent NP *** vivo studies validated that a single injection of the hydrogel in situ could maintain IVD tissue structure and alleviate sensitivity to noxious mechanical force in the rat models,indicating a potential therapeutic approach for ameliorating IVDD and *** approach helps prevent potential systemic toxicity associated with systemic administration and reduces the morbidity resulting from repeated injections of free drugs into the IVD,providing a new strategy for IVDD treatment.

Association study of GRIK4 polymorphisms and major depressive disorder in the Han Chinese population

Objective Major depressive disorder(MDD) is a common and complex mental disorder characte rized by depression. Recent studies found that glutamate ionotropic receptor kainate type subunit 4(GRIK4) is associated with the MDD and the gene GRIK4, therefore, may be a candidate gene f or many mental disorder especially MDD. In this study, we investigated whether single nucleotide polymorphisms(SNPs) in GRIK4 were associated with *** In the case-control study, we recruited 568 unrelated major depressive disorder patients and 846 healthy controls. All patients were diagnosed on the basis of DSM-IV criteria. We had ge notyped five SNPs using the matrix-assisted laser desorption/ionization time of flight(MALDI-TOF) mass spectrometer on Mass ARRAY? Analyzer 4 platform(Sequenom, San Diego, CA). Design of probes and primers was by using the Mydequenom online software--the accompanying Assay Design Suite v2.0. Several softwares was conducted to do the statistic analysis. Result The results showed that rs56275759 demonstrated statistically significant difference betw een MDD patients and control subjects both in allelic frequencies(P value = 0.011449) and genot ypic frequencies(P value = 0.029024). Rs4582985 was deviated from the Hard-Weinberg equilibr ium so we excluded it from further analysis. Strong LD was found among rs4582985, rs6589847 and rs56275759, and the significant association between this block and MDD was found. Conclusion In summary, our results indicated that rs56275759 in GRIK4 might be correlated with MDD in the Han Chinese population. MDD, a severe and complex psychiatric disorder, is affecte d by many elements such as genetic, psychological and environmental factors. The study may pr ovide a reference for future studies of MDD. The SNPs we studied may be used as a marker for p redicting the MDD in the disease diagnose.
