One in three Americans is at risk for developing chronic kidney disease （CKD） and end-stage renal disease （ESRD）, leading to the need for dialysis or a kidney transplant. Small-molecule drugs have been proposed as therapies to manage kidney diseases, but high dosages are often required to achieve therapeutic efficacy, generating off-target side effects, some of which are lethal. To address these limitations, we developed a novel kidney-targeting multimodal micelle （KM） system for drug delivery applications. SpecificaU~ we incorporated the kidney-targeting peptide （Lysine-Lysine-Glutamic acid-Glutamic acid-Glutamic acid）3-Lysine）（（KKEEE）3K） into micelles. This peptide binds to megalin, a multi-ligand cell surface receptor present on renal tubule cells. When incubated with human kidney proximal tubule cells, KMs were found to be biocompatible in vitro. In vivo, KMs showed higher accumulation in the kidneys as compared to a non-targeted （NT） control upon intravenous injection in wild-type C57BL/6j mice. histological evaluation showed no signs of tissue damage, while blood urea nitrogen （BUN） and creatinine levels were within normal ranges, validating the preservation of kidney health upon micelle administration. To our knowledge, this is the first utilization of （KKEEE）BK in a nanoparticle formulation, and our study offers strong evidence that this novel nanoparticle platform can be used as a candidate drug delivery carrier to direct therapeutics to diseased tissue in CKD.

To monitor the progression of lamivudine therapy by demonstrating hBV DNAs and RNAs in the serum of achronically infected patient. Viral nucleic acids were exracted from serum samples taken during a 14-week treatmentperiod and analyzed via competitive PCR and RT/PCR. The dynamic changes in the amount as well as in the structurescan monitor therapy process very well. Our study indicated the level of polyadenylated viral RNA can be a marker forexpression in general, and the appearance of replicative intermediates as a mirror of the mode of action of an antiviraldrug.

Objectives: This study sought to compare tissue Doppler imaging(TDI) with velocity-encoded(VE) magnetic resonance imaging(MRI) for left ventricular(LV) dyssynchrony assessment. Background: Cardiac resynchronization therapy(CRT) is proposed for patients with heart failure, depressed LV function, and a wide QRS complex. Selection is based mainly on electrocardiogram criteria, but recent data suggest that intraventricular dyssynchrony may be preferred for selection. An LV dyssynchrony can adequately be assessed with TDI, but this has not been compared directly with other imaging modalities. A VE MRI potentially allows direct myocardial wall motion measurements similar to TDI. Methods: Twenty patients with heart failure, systolic LV dysfunction, and a wide QRS complex were included, as well as 10 normal individuals with normal QRS duration and LV function. The TDI and VE MRI data were acquired to study intraventricular dyssynchrony. Results: Left ventricular dyssynchrony was not observed in normal individuals(mean dyssynchrony-2±15 ms on TDI; mean-5±17 ms on MRI, p=NS). In patients, mean LV dyssynchrony was 55±37 ms on TDI; 49±38 ms on MRI(p=NS). Good correlation between both modalities was observed(linear regression TDI=0.99×MRI-5, n=30, r=0.98, phe MRI showed a small, nonsignificant underestimation of 5±8 ms compared with TDI. Agreement between MRI and TDI for classification according to severity of LV dyssynchrony(minimal, intermediate, and extensive) was excellent(κ±SE=0.96±0.07, ph 95%of patients classified identical. Conclusions: Both MRI and TDI yield comparable information on LV dyssynchrony; MRI is useful in the selection of patients for CRT.

Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor ILT2(synonyms:LIR1,MIR7,CD85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(MhC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of MhC I-positive cells of many *** novel compounds antagonized the interaction of CD8 co-receptor with MhC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of CD8^(+)cytotoxic T lymphocytes(CTLs)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of CD8^(+)CTL responses,the engineered high affinity ILT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTLs and could have potential for the development of novel autoimmunity therapies.

Background: ALS is believed to be multifactorial in origin with modifying genes affecting its clinical expression. Childhood-onset spinal muscular atrophy (SMA) is an autosomal recessive disorder of motor neurons, caused by mutations of the survival motor neuron (SMN) gene. The SMN gene exists in two highly homologous variants: SMN1, the causative gene responsible for the production of the majority of functional SMN protein, and SMN2, responsible for the production of less protein but sufficient for modifying the SMA phenotype. Objective: To test whether SMN genotypes are associated with susceptibility to and severity of sporadic ALS. Methods: We performed competitive quantitative PCR analysis for both SMN1 and SMN2 genes in 242 clinically well-defined ALS patients and 175 controls. The combined determination of SMN1 and SMN2 copies also allowed for an estimation of the level of SMN for each patient (estimated SMN protein level = SMN1 copy number +0.20×SMN2 copy number). Results: One copy of SMN1 was associated with an increased risk of developing ALS (odds ratio = 4.1, 95%CI = 1.2 to 14.2, p = 0.02) and ALS patients carried fewer SMN2 copy numbers (p had an estimated protein SMN level ≤2.2 vs only 36%of controls (p = 0 .0000004). Multivariate Cox regression analyses showed that lower SMN2 copy numb ers and lower levels of estimated SMN protein (hazard ratio = 1.3, 95%CI = 1.1 to 1.6, p = 0.03) were associated with an increased mortality rate. Conclusions: SMN genotypes producing less SMN protein increase susceptibility to and severit y of ALS.

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large hadron Collider,as Standard Model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the jet *** in tagging efficiency between data and simulation are provided for jets with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.

Objectives:Dyslipidemia is strongly associated with raised plasma glucose levels and insulin resistance(IR),and genome-wide association studies have identified 95loci that explain a substantial proportion of the variance in blood ***,the loci’s effects on glucose-related traits are largely *** current research aims to investigate relationship between the lipid loci and glucose-related *** and methods:We have studied these lipid loci and tested their association collectively and individually with fasting plasma glucose(FPG),glycated hemoglobin(hbA1c),and IR in two independent cohorts:10,995 subjects from LifeLines Cohort Study and 2,438subjects from Prevention of Renal and Vascular Endstage Disease(PREVEND)***:In contrast to the positive relationship between dyslipidemia and glucose traits,the genetic predisposition to dyslipidemia showed a pleiotropic lowering effect on glucose ***,the genetic risk score related to higher triglyceride level was correlated with lower levels of FPG(P=9.6×10(-10)and P=0.03 in LifeLines and PREVEND,respectively),hbA1c(P=4.2×10(-7)in LifeLines),and hOMA of estimated IR(P=6.2×10(-4)in PREVEND),after adjusting for blood lipid *** the single nucleotide polymorphism level,15 lipid loci showed a pleiotropic association with glucose traits(Phich eight(CETP,MLXIPL,PLTP,GCKR,APOB,APOE-C1-C2,CYP7A1,and TIMD4)had opposite allelic directions of effect on dyslipidemia and glucose ***:For the first time,we have systematically assessed the pleiotropic effect of lipid-associated SNPs on glucose-related ***,at a clinical level,dyslipidemia is associated with elevated plasma glucose levels and insulin resistance,we observed that the genetic predisposition to dyslipidemia is related to lower levels of fasting plasma glucose,hbA1c,and *** findings suggest a complex genetic regulation and metabolic interplay between lipids and glucose.
