Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor IlT2(synonyms:lIR1,MIR7,CD85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(MHC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of MHC I-positive cells of many *** novel compounds antagonized the interaction of CD8 co-receptor with MHC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of CD8^(+)cytotoxic T lymphocytes(CTls)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of CD8^(+)CTl responses,the engineered high affinity IlT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTls and could have potential for the development of novel autoimmunity therapies.

The ionic liquid(Il) tributylmethylammonium bis(trifluoromethylsulfonyl)amide([N4441][NTf2]) was used as neat lubricant and as an additive(1.5 wt%) in a polar oil to study its friction and wear reducing *** tests were completed for 90 minutes at room temperature and 100 °C in a reciprocating configuration at loads of 30 and 70 N,10 Hz-frequency,and 4 mm stroke *** volume was measured by confocal microscopy and the surface-Il interaction determined by *** main findings were that neat Il showed the best tribological behavior;the Il-containing mixture behaved similar to the base oil regarding friction,however outperformed the antiwear behavior of the base oil under higher temperature;surface-Il chemical interaction was found mainly at 100 °C.

Freshwater biota are more comprehensive and direct indicators of biological impacts,and more meaningful to the public than water quality or physical habitat *** biotic data and the multiple biological indicators developed from them offer a much richer array of data for assessing the impacts of pollution controls than a limited set of physical or chemical *** recent decades,assemblage-based assessments by ecologists,environmental scientists,and water quality agencies have been employed globally for determining the condition of,and threats to,freshwater ecosystems.A key step in this advance has been the development of multimetric indices(MMIs)or indices of biotic integrity(IBIs)based on quantitative assessments of algae,macrophyte,macroinvertebrate,fish or riparian bird *** Europe,where biological assemblages are mandated for assessing freshwater ecosystem health,many indices are ***,the proliferation of MMIs globally has not always occurred through the application of rigorous study designs and monitoring protocols,nor have they always effectively incorporated functional metrics,stressor assessments,and statistical ***,in this review,we discuss eleven major concerns with the development and application(including logistical limitations)of multimetric indicators based on freshwater biota to encourage more rigorous and widely applicable(transferable)MMI use and ***,our concerns focus on reference conditions;sampling effort,methods,and season;trophic guild definition;metric comprehensiveness,options,screening and scoring;and MMI *** could also benefit from increased attention to ecological mechanisms and metric development,to further improve our understanding of anthropogenic impacts as well as rehabilitation effects on freshwater ecosystems *** closer attention to study designs,ecological mechanisms and metric development should further improve our understa

目的:用非线性混合效应模型(NONMEM)软件建立利福平做为抗生素治疗在儿童体内的群体药代动力学模型.方法:回顾性收集29例患儿的133份血样的血药浓度及临床数据.患儿年龄0.04-15.9岁(平均8.97岁),体重3-55公斤(平均33.43公斤),利福平口服或者静脉滴注剂量为8.3-21.9mg/kg,服药间隔12小时,采血时间0.75-15.9小时,同时混合血药浓度稳态和非稳态情况。采用反相高效液相色潽紫外荧光法测定利福平的血药浓度。运用NONMEM软件建立药代动力学模型估算药代动力学参数并分析药代动力学变化特点。结果:利福平在儿童体内的药代动力学符合一房室模型。其药物吸收速率常数(Ka)、清除率(Cl)、表观分布容积(V)和生物利用度(F)分别为1.54h-1,9.773l/h,45.1l和0.901。患儿体重对清除率和表观分布容积有显著影响,年龄对清除率有显著影响。性别对给药剂量无显著影响。模型通过自举法(bootstrap),直观预测检验(Visual Predictive Check,VPC)和正态预测分布误差(Normalized predictive distribution error,NPDE)进行验证评估,拥有良好的稳定性及准确的预测性能。结论:本模型可以较好解释利福平在服药间隔为12小时时,在儿童体内药代动力学个体差异的原因,运用次模型可准确预测利福平的血药浓度,促进个体化给药方案的实施。

Viral Acute Respiratory Infections in Central African Republic Children: Epidemiological and Clinical Aspects

Background: Acute respiratory infections (ARI) are recognized as an important cause of morbidity, mortality, and hospitalization among children in developing countries. Objectives: To identify the respiratory viruses circulating in Central African children before the SARS-COV2 pandemic and to assess the clinical manifestations. Methodology: This is a cross-sectional, descriptive, multicenter study, run from March 1, 2019, to March 31, 2020. Children aged 28 days to 15 year-old, with respiratory symptoms ≤10 days had been included. Nasopharyngeal swabs were taken and sent to the Institute Pasteur in Bangui (WHO National Referral Center for influenza). Virus research was done by cell and molecular culture techniques. Data were recorded and processed with Access 2019 software, then analyzed with STATA version 14 software. Chi-square test and ANOVA test were used to compare proportions at the p 0.05 threshold. Results: Out of 659 children included during the study period, viruses were identified in 231 children, for an overall positivity rate of 35.05% (231/659). Rhinoviruses (RV) and influenza viruses were found in 66.23% and 16.88% respectively. Virus-virus co-infections were found in 10 (10/231) children (4.32%). Children under 5 years of age were more represented (78.60%). The main reasons for consultation were: fever (96.20%), cough (95.45%), runny nose (78.5%), and breathing difficulty (30.50%). IlI (Influenza-like Illness) was found in 71.02% versus 28.98% of SARI (Severe Acute Respiratory Infection). There was a statistically significant association between age 5 years and severity of acute respiratory infection (p = 0.001). The outcome was known for the 122 children at the CHUPB site with a mortality rate of 17.21% (n = 21). Conclusion: Viral ARI is common in children in Central African Republic. Care givers should think about it in order to reduce the inappropriate prescription of antibiotics.

AIM:To investigate whether or not bariatric surgeryweight outcomes vary by ethnicity in a large,nationally representative sample of ***:The Bariatric Outcomes longitudinal Database was used for analysis and contains data on surgeries performed on adolescents from 2004 to 2010from 423 surgeons at 360 facilities across the United States Adolescents(n=827)between 11 and 19 years old who underwent either gastric bypass or adjustable gastric banding surgery were included in the *** measures included changes in anthropometric measurements[weight(kg)and body mass index]from baseline to 3(n=739),6(n=512),and 12(n=247)mo after ***:A year after patients underwent either gastric bypass(51%)or adjustable gastric banding(49%)surgery,mean estimated weight loss for all ethnic groups differed by a maximum of only 1.5 kg,being34.3 kg(95%CI:30.0-38.5 kg)for Hispanics,33.8 kg(95%CI:27.3-40.3 kg)for non-Hispanic blacks,and32.8 kg(95%CI:30.9-34.7 kg)for non-Hispanic *** overall pairwise group comparisons were significant,indicating that no ethnic group had better weight loss outcomes than did ***:Bariatric surgery substantially reduces the weight of severely obese adolescents at 1 year post-procedure with little variation by ethnicity and/or *** results suggest that bariatric surgery is a safe and reasonable treatment for all severely obese adolescents with the appropriate indications.
