The ORF9b protein,derived from the nucleocapsid’s open-reading frame in both SARS-CoV and SARS-CoV-2,serves as an accessory protein crucial for viral immune evasion by inhibiting the innate immune *** its significance,the precise regulatory mechanisms underlying its function remain *** the present study,we unveil that the ORF9b protein of SARS-CoV-2,including emerging mutant strains like delta and Omicron,can undergo ubiquitination at the K67 site and subsequent degradation via the proteasome pathway,despite certain mutations present among these ***,our investigation further uncovers the pivotal role of the translocase of the outer mitochondrial membrane 70(TOM70)as a substrate receptor,bridging ORF9b with heat shock protein 90 alpha(HSP90α)and Cullin 5(CUL5)to form a *** this complex,CUL5 triggers the ubiquitination and degradation of ORF9b,acting as a host antiviral factor,while HSP90αfunctions to stabilize ***,treatment with HSP90 inhibitors such as GA or 17-AAG accelerates the degradation of ORF9b,leading to a pronounced inhibition of SARS-CoV-2 ***-cell sequencing data revealed an up-regulation of HSP90αin lung epithelial cells from COVID-19 patients,suggesting a potential mechanism by which SARS-CoV-2 may exploit HSP90αto evade the host *** study identifies the CUL5-TOM70-HSP90αcomplex as a critical regulator of ORF9b protein stability,shedding light on the intricate host–virus immune response dynamics and offering promising avenues for drug development against SARS-CoV-2 in clinical settings.

Analysis method and experimental study of ice accumulation detection signal based on Lamb waves

A quantitative identification method for in-flight icing has the capability to significantly enhance the safety of aircraft *** guided waves have the unique advantage of detecting icing in a relatively large area,but quantitative identification of ice layers is a *** this paper,a quantitative identification method of ice accumulation based on ultrasonic guided waves is ***,a simulation model for the wave dynamics of piezoelectric coupling in three dimensions is established to analyze the propagation characteristics of Lamb waves in a structure consisting of an aluminum plate and an ice *** wavelet transform method is utilized to extract the Time of Flight(ToF)or Time of delay(ToD)of S_(0)/B_(1) mode waves,which serves as a characteristic parameter to precisely determine and assess the level of ice ***,an experimental system is developed to evaluate the feasibility of Lamb waves-based icing real-time detection in the presence of spray ***,a combination of the Hampel median filter and the moving average filter is developed to analyze ToF/ToD *** simulation results reveal a positive correlation between geometric dimensions(length,width,thickness)of the ice layer and ToF/ToD of B1 mode waves,indicating their potential as indicators for quantifying ice *** results of real-time icing detection indicate that ToF/ToD will reach greater peak values with the growth of the arbitrary-shaped ice layer until saturation to effectively predict the simulation *** study lays a foundation for the practical application of quantitative icing detection via ultrasonic guided waves.

Objective This study aimed to comprehensively analyze and compare the clinicopathological features and prognosis of Chinese patients with human epidermal growth factor receptor 2(HER2)-low early breast cancer(BC)and HER2-IHC0 *** Patients diagnosed with HER2-negative BC(N=999)at our institution between January2011 and december 2015 formed our study *** characteristics,association between estrogen receptor(ER)expression and HER2-low,and evolution of HER2 immunohistochemical(IHC)score were ***-Meier method and log-rank test were used to compare the long-term survival outcomes(5-year follow-up)between the HER2-IHC0 and HER2-low *** HER2-low BC group tended to demonstrate high expression of ER and more progesterone receptor(PgR)positivity than HER2-IHC0 BC group(Ponger overall survival(OS)in HER2-low BC group than in HER2-IHC0 group(P=0.007)in the whole cohort and the hormone receptor(HR)-negative *** were no significant differences between the two groups in terms of disease-free survival(DFS).The discordance rate of HER2 IHC scores between primary and metastatic sites was 36.84%.Conclusion HER2-low BC may not be regarded as a unique BC group in this population-based study due to similar clinicopathological features and prognostic roles.

Background Two studies were designed to determine standard ileal crude protein(CP)and amino acid(AA)digestibility of soybean meal(SBM)from different origins fed to non-pregnant and pregnant *** solvent-extracted SBMs from soybeans produced in the USA,Brazil,and China were *** Exp.1,eight different diets were created:a nitrogen(N)-free diet and 7 experimental diets containing SBM from different origins as the only N *** non-pregnant,multiparous sows were arranged in an 8×8 Latin square design(8 periods and 8 diets).In Exp.2,the diet formula was the same as in *** gestating sows(parity 3)were assigned to 4 different diets in a replicated 4×3 Youden square design(three periods and four diets)in mid-gestation and again in late-gestation *** When fed to non-pregnant and late-gestating sows,the standardized ileal digestibility(SID)of CP and most AAs from different SBM were not significantly different(P>0.05).When fed to mid-gestating sows,the SID values for Arg,His,Lys,Phe,Cys,Gly,Ser,and Tyr in SBM 1 were lower than in SBM 4 and 5(Pong SBM *** SID values for Lys,Met,Val,Ala,Asp,and Tyr in SBM were affected by sow gestation *** findings provide a cornerstone for accurate SBM use in sow diets.

Ultra-thin robust CNT@GC film integrating effective electromagnetic shielding and flexible Joule heating

The demand for lightweight,thin electromagnetic interference(EMI)shielding film materials with high shielding effectiveness(SE),excellent mechanical properties,and stability in complex environments is particularly pronounced in the realm of flexible and portable electronic ***,we developed an ultra-thin film(CNT@GC)in which the glassy carbon(GC)layer wrapped around and welded carbon nanotubes(CNTs)to form a core-shell network structure,leading to exceptional tensile strength(327.2 MPa)and electrical conductivity(2.87×10^(5) S·m^(−1)).The CNT@GC film achieved EMI SE of 60 dB at a thickness of 2µm after post-acid treatment and high specific SE of 3.49×10^(5) dB·cm^(2)·g^(−1),with comprehensive properties surpassing those of the majority of previous shielding ***,the CNT@GC film exhibited Joule heating capability,reaching a surface temperature of 135℃at 3 V with a fast thermal response of about 0.5 s,enabling anti-icing/de-icing *** work presented a methodology for constructing a robust CNT@GC film with high EMI shielding performance and exceptional Joule heating capability,demonstrating immense potential in wearable devices,defense,and aerospace applications.

在云南省干热河谷区,以不同紫花苜蓿(Medicago sativa)品种(系)作为研究对象,应用方差和灰色关联度的分析方法对其生产性能及营养成分进行综合评价。结果表明,8个品种(系)的一级分枝数和水分有明显差异。所有参试材料在2018年和2020年每年可刈割8次,2019年为9次。6号苜蓿品种‘南苜701-特克拉’的年均干草产量最高,达到29589.96 kg·hm^(-2),显著高于5号‘四季旺’和8号‘德钦’(P第2季度>第4季度>第1季度。所有供试材料的粗纤维、粗脂肪、粗灰分、全钙、无氮浸出物差异均不显著(P>0.05)。8号品种‘德钦’的粗蛋白含量(21.43%)、粗灰分(10.16%)和全磷含量(0.25%)均最高,粗纤维含量最低(18.36%)。经灰色关联度分析表明,‘热引41号紫花苜蓿’‘德钦’‘多叶’和‘WL525HQ’的综合表现较好,在该地区具有较高的推广利用价值。

RNA interference(RNAi)is a powerful tool that post-transcriptionally silences target genes in eukaryotic ***,silencing efficacy varies greatly among different insect ***,we met with little success when attempting to knock down genes in the mirid bug Apolygus lucorum via dsRNA *** disappearance of double-stranded RNA(dsRNA)could be a potential factor that restricts RNAi ***,we found that dsRNA can be degraded in midgut fluids,and a dsRNase of ***(AldsRNase)was identified and *** alignment indicated that its 6 key amino acid residues and the Mg2+-binding site were similar to those of other insects’*** signal peptide and endonuclease non-specific domain shared high sequence identity with the brown-winged green stinkbug Plautia stali *** showed high salivary gland and midgut expression and was continuously expressed through the whole life cycle,with peaks at the 4th instar ecdysis in the whole *** purified AldsRNase protein obtained by heterologously expressed can rapidly degrade *** comparing the substrate specificity of AldsRNase,3 specific substrates(dsRNA,small interfering RNA,and dsDNA)were all degraded,and the most efficient degradation is ***,immunofluorescence revealed that AldsRNase was expressed in the cytoplasm of midgut *** cloning and functional study of AldsRNase,the enzyme activity and substrate specificity of the recombinant protein,as well as the subcellular localization of nuclease,the reason for the disappearance of dsRNA was explained,which was useful in improving RNAi efficiency in *** and related species.
