METTL5 promotes gastric cancer progression via sphingomyelin metabolism

BACKGROUND The treatment of gastric cancer(GC)has caused an enormous social burden worldwide.Accumulating studies have reported that N6-methyladenosine(m6A)is closely related to tumor progression.METTL5 is a m6A methyltransferase that plays a pivotal role in maintaining the metabolic stability of cells.However,its aberrant regulation in GC has not been fully elucidated.AIM To excavate the role of METTL5 in the development of GC.METHODS METTL5 expression and clinicopathological characteristics were analyzed via The Cancer Genome Atlas dataset and further verified via immunohistochemistry,western blotting and real-time quantitative polymerase chain reaction in tissue microarrays and clinical samples.The tumor-promoting effect of METTL5 on HGC-27 and AGS cells was explored in vitro by Cell Counting Kit-8 assays,colony formation assays,scratch healing assays,transwell assays and flow cytometry.The tumor-promoting role of METTL5 in vivo was evaluated in a xenograft tumor model.The EpiQuik m6A RNA Methylation Quantification Kit was used for m6A quantification.Next,liquid chromatography-mass spectrometry was used to evaluate the association between METTL5 and sphingomyelin metabolism,which was confirmed by Enzyme-linked immunosorbent assay and rescue tests.In addition,we investigated whether METTL5 affects the sensitivity of GC cells to cisplatin via colony formation and transwell experiments.RESULTS Our research revealed substantial upregulation of METTL5,which suggested a poor prognosis of GC patients.Increased METTL5 expression indicated distant lymph node metastasis,advanced cancer stage and pathological grade.An increased level of METTL5 correlated with a high degree of m6A methylation.METTL5 markedly promotes the proliferation,migration,and invasion of GC cells in vitro.METTL5 also promotes the growth of GC in animal models.METTL5 knockdown resulted in significant changes in sphingomyelin metabolism,which implies that METTL5 may impact the development of GC via sphingomyelin metabolism.In addition,high METTL5 expr

Herein,we report the facile synthesis of a highly strained hexabenzocoronene-containing carbon nanoring,cyclo[4]-paraphenylene[2]-2,11-hexabenzocoronenylene([4,2]CPHBC),as the segment of a[10,10]single-walled carbon nanotube([10,10]SWNT).[4,2]CPHBC was synthesized based on the platinummediated assembly of diborylbiphenyl and diborylhexabenzocoronene,forming a tetranuclear platinum complex,followed by reductive elimination.This nanoring molecule was confirmed by NMR and HRMS,and its photophysical properties were studied using steady-state and time-resolved spectroscopies.Moreover,the selective supramolecular host-guest interaction between[4,2]CPHBC and C_(60) was also investigated.

design of a rapid-cycling synchrotron for flash proton therapy

The purpose of this study was to design a rapid-cycling synchrotron, making it capable of proton beam ultrahigh dose rate irradiation, inspired by laser accelerators. The design had to be cheap and simple. We consider our design from six aspects: the lattice, injection, extraction, space charge effects, eddy current effects and energy switching. Efficiency and particle quantity must be addressed when injected. The space charge effects at the injection could affect particles' number. The eddy current effects in the vacuum chambers would affect the magnetic field itself and generate heat, all of which need to be taken into account. Fast extraction can obtain 10^(10) protons/pulse, equal to instantaneous dose rate up to 10~7 Gy/s in a very short time, while changing various extraction energies rapidly and easily to various deposition depths. In the further research, we expect to combine a delivery system with this accelerator to realize the FLASH irradiation.

Natural gas characteristics and gas-source comparisons of the Lower Triassic Feixianguan Formation,Eastern Sichuan Basin,China

There is great controversy regarding the origin and source of natural gas in the Lower Triassic Feix-ianguan Formation in the Eastern Sichuan Basin.This seriously restricts the study of natural gas dy-namics in the Feixianguan Formation and thus hampers natural gas exploration in the region,so further study is urgently required.Using experimental tests of natural gas composition,stable isotopes,and noble gas isotopes with gas chromatography(GC)and mass spectrometry(MS)studies of source rock and reservoir asphalt saturated hydrocarbons,the natural gas geochemical characteristics,the genetic identification and a gas-source comparison of the Feixianguan Formation were studied.Then,con-strained by the thermal history,the histories of gas generation and expulsion were restored by basin simulation technology.Finally,a gas accumulation model was established for the Feixianguan Formation.The results showed that(1)the H_(2)S-rich and H2S-poor gas reservoirs of the Feixianguan Formation are distributed on the east and west sides of the Kaijiang-liangping trough in the Eastern Sichuan Basin,respectively.The carbon and hydrogen isotope compositions of the natural gas in the gas reservoirs are generally heavy and have typical characteristics of high-maturity dry gas reservoirs.(2)The natural gas of the Feixianguan Formation is organic thermogenic gas,which is mainly oil-type gas generated by the secondary cracking of crude oil.The gas-generating parent material is mainly type II kerogen.(3)The natural gas of the Feixianguan Formation in the Eastern Sichuan Basin was mainly generated by argil-laceous source rocks of the Upper Permian Longtan Formation.(4)Natural gas accumulation occurred as follows:the paleo-structure heights were filled with crude oil in the Early Jurassic,and paleo-oil res-ervoirs were formed in the Feixianguan Formation;during the Middle-Late Jurassic,the paleo-oil res-ervoirs were cracked when the reservoir temperatures rose above 160 C,and paleo-gas reservoirs were formed.Since the end of the Late Jurassi

Let G be a graph with vertex set V(G),edge set E(G)and maximum degreeΔrespectively.G is called degree-magic if it admits a labelling of the edges by integers{1,2,...,|E(G)|}such that for any vertex v the sum of the labels of the edges incident with v is equal to1+|E(G)|/2·d(v),where d(v)is the degree of v.Let f be a proper edge coloring of G such that for each vertex v∈V(G),|{e:e∈Ev,f(e)≤Δ/2}|=|{e:e∈Ev,f(e)>Δ/2}|,and such an f is called a balanced edge coloring of G.In this paper,we show that if G is a supermagic even graph with a balanced edge coloring and m≥1,then(2m+1)G is a supermagic graph.If G is a d-magic even graph with a balanced edge coloring and n≥2,then n G is a d-magic graph.Results in this paper generalise some known results.

A Class of Antimagic Join Graphs

A labeling f of a graph G is a bijection from its edge set E（G） to the set {1, 2, ..., ｜E（G）｜}, which is antimagic if for any distinct vertices x and y, the sum of the labels on edges incident to x is different from the sum of the labels on edges incident to y. A graph G is antimagic if G has an f which is antimagic. Hartsfield and Ringel conjectured in 1990 that every connected graph other than K2 is antimagic. In this paper, we show that if G1 is an n-vertex graph with minimum degree at least r, and G2 is an m-vertex graph with maximum degree at most 2r - 1 （m ≥ n）, then G1 V G2 is antimagic.

Simulation study of slow extraction for the Shanghai Advanced Proton Therapy facility

The Shanghai Advanced Proton Therapy facility employs third-integer slow extraction. In order to achieve accurate treatment, high-quality spill is needed. Therefore,parameters that may affect slow extraction should be investigated by simulation. A computer model of the synchrotron operation slow extraction was constructed with MATLAB~. By simulating the motion of the circulating protons, we could quantify the influence of machine and initial beam parameters on properties of the extracted beam, such as ripple, uniformity, stability, on-and off-time of the spill and spill width in the synchrotron.Suitable design parameters including the horizontal tunes,power supply ripple, longitudinal RF cavity voltage, RFKO and the chromaticities were determined.

Because of its excellent dose distribution,proton therapy is becoming increasingly popular in the medical application of cancer treatment.A synchrotron-based proton therapy facility was designed and constructed in Shanghai.The synchrotron,beam delivery system,and other technical systems were commissioned and reached their expected performances.After a clinical trial of 47 patients was finished,the proton therapy facility obtained a registration certificate from the National Medical Products Administration.The characteristics of the accelerator and treatment systems are described in this article.

Physical design and multi-particle simulations of a compact IH-DTL with KONUS beam dynamics for proton therapy

A compact room-temperature inter-digital H-mode(IH)drift tube linac(DTL)with Kombinierte Null Grad Struktur beam dynamics is proposed in this paper.The proposed IH-DTL,which operates at 325 MHz,accelerates 18 mA proton particles from 3.0 to 7.0 MeV as part of a proton synchrotron-based therapy system.It is composed of two main sections,namely a bunching section(-30°)and accelerating section(0°).There is no transverse focusing element inside the cavity,which increases the acceleration gradient and reduces the cavity length and power consumption.In our physical designs,LORASR code is utilized for beam dynamics design and multi-particle simulations.The synchronous particle energy,injection phase,and acceleration voltage of each gap are optimized carefully to increase transmission efficiency while minimizing beam emittance growth and beam loss.The total length of the cavity is 0.82 m and the acceleration gradient reaches 4.88 MV/m,resulting in a transmission efficiency of 100%and beam emittance growth less than10%.The details of the specific work undertaken in this study are presented in this paper.

Eddy current effects in a high field dipole

Eddy currents produced by a time-varying magnetic field will introduce time delay and thus affect field quality. This effect leads to drifting of the beam position over time, especially for a compact synchrotron.Simulations and measurements of different dipoles have been performed, to investigate the time delay and field quality. The simulations are conducted using OPERA software. The measurements are conducted using a long coil and Hall sensor. All results show that the magnetic field deviation is up to 0.4% for the dipole with stainless steel endplates. The simulations show that the main sources of eddy current are the field saturation effect and the field component Bz, introduced by the bedstead-type coil. Field correction using a power supply is adopted to reduce the deviation to less than 0.02%.
