The cerebellum receives a wide variety of sensory inputs and generates motor-related outputs according to internal rules of *** climbing fiber(CF) and mossy fiber(MF) are condidered to transfer sensory information into cerebellar cortex,the CFs activate the Purkinje cells(PCs) directly by discharging complex spikes,whereas the mossy fibers excite them indirectly via the granule cells by discharging simple *** sensory stimulus precisely evokes responses in cerebellar PCs,the cellular mechanisms of sensory information processing in cerebellar cortex are ***,we studied the synaptic responses of cerebellar molecular layer interneurons(MLIs) and PCs evoked by air-puff stimulation of the ipsilateral whisker pad by in vivo whole-cell patch-clamp recording techniques in urethane-anesthetized ICR *** found that air-puff stimulus failed to provoke either simple or complex spikes in PCs,but evoked inhibitory postsynaptic potentials(IPSPs) in the somas of *** voltage-clamp,the stimulus evoked a sequence of transient inward currents followed by strong outward currents in the somas and dendrites in *** outward currents,revealed excitatory postsynaptic currents(EPSCs) in somas and a temporal summation of parallel fiber EPSCs in PC ***,the stimulus evoked spike firing and rapid EPSCs in MLIs,and the highest amplitude,shortest duration EPSCs were recorded from MLIs deep in the molecular layer,which expressed basket-type synaptic *** MLI to PC responses,time to peak of PC IPSP was significantly slower than MLI recorded *** GABA receptors uncovered larger EPSCs in PCs whose time to peak,half-width and 10-90%rising time were also significantly slower than in *** labeling indicated that the MLIs(but not PCs) are *** findings indicated that tactile face stimulation evoked rapid excitation in MLIs and inhibition of PCs in mouse cerebellar cortex via the MF-granule

Overdose intake of ethanol can impair cerebellar cortical neurons to integrate and transfer external information, resulting in a dysfunction of cerebellar motor regulation or cerebellar ataxia. We recently investigated the effects of ethanol on sensory stimulation-evoked responses in the cerebellar cortex of urethane-anesthetized mice, by electrophysiological and pharmacological methods. We found that ethanol dosedependently inhibited the facial stimulation evoked field potential responses in cerebellar granule cell layer, whereas ethanol bidirectional modulate the sensory stimulation-evoked responses in molecular layer via GABAA receptor. In vivo patch-clamp recordings showed that ethanol dose-dependently inhibited the facial stimulation-evoked outward currents of cerebellar Purkinje cells（PCs）. The ethanol-induced inhibition of facial stimulation-evoked outward currents were significantly abrogated by cannabinoid receptor 1（CB1） antagonists and agonist. The ethanol-induced inhibition of facial stimulation-evoked outward currents was prevented by cerebellar surface perfusion of the PKA inhibitors H-89, but not by intracellular administration of the PKA inhibitor PKI. Our present results indicate that ethanol inhibits the facial stimulation-evoked outward currents by activating presynaptic CB1 receptors via the PKA signaling pathway, suggesting that ethanol overdose impairs sensory information processing, at least in part, by inhibiting GABA release from molecular layer interneurons onto PCs. In addition, ethanol inhibited the spontaneous CS-evoked pause in simple spike firing and in the amplitude of the afterhyperpolarization via activation of CB1 receptors in vivo in mice, suggesting that excessive ethanol intake inhibits climbing fiber–PC synaptic transmission by modulating CB1 receptors in the cerebellar cortex.

Background: It has been reported that the mechanisms of morphine addiction is the homeostasis dysregulation of brain. The cerebellum is involved in control of motor coordination and receives sensory inputs to execute complex motor tasks. Increasing evidence has involved that addiction is related to a disruption in cerebellar processing and connectivity with other brain regions. However, the changes of information processing in cerebellum in morphine addiction is unknown. In present research, our aim is to observe the effects of morphine dependence and withdrawal on sensory information processing in cerebellar granule cell layer in vivo in mice. Methods and Main Results: Forty male KM mice were randomly divided into four groups（n=10）: the morphine dependence group, morphine withdraw 1 day group, morphine withdrawal 7 days group, and the saline control group. The mice in dependence group and withdrawal group were administered with morphine by subcutaneous injection through the method of "increasing addiction for 7 days", and the saline group were injected saline. Mice administered morphine exhibited opioid-like responses including Straub tail and hyperlocomotion in behavior. Electrophysiological methods were used in our research to record the field potential of granule cell layer（GCL）. Air-puff stimulation of the ipsilateral whisker-pad evoked stimulus-on（N1） and stimulus-off responses（N2） in GCL of cerebellar Crus II. The amplitude of N1 and N2 in dependence group, withdrawal 1 day group and withdrawal 7 day group are obviously decreased compared with control group（p＜0.05）, and withdrawal 1 day group shows a marked drop compared with dependence group and withdrawal 7 day group（p＜0.05）. The onset latency of the sensory stimulationevoked responses has no change. Conclusion: These results indicate that morphine dependence and withdrawal affect the homeostatic regulation of sensory information processing in cerebellar granule cell layer.

Objective To study the properties and possible mechanisms of the long-term synaptic plasticity induced by the facial tactile stimulation at mossy fiber-granule cell（MF-GC） synapses in urethane-anesthetized mice, by field potential recording technique and pharmacological methods. Methods Adult ICR mice（6–8-week-old） were anesthetized with urethane, and tracheotomized to avoid respiratory obstruction. The craniotomy was drilled to expose the cerebellar surface corresponding to Crus II. Results Our results showed that 4 Hz facial stimulation evoked long-term depression（LTD） of MF-GC synaptic transmission. However, 20 Hz facial stimulation evoked long-term potentiation（LTP） at MF-GC synapses. The MF-GC LTP was prevented by N-methyl-Daspartic acid receptor（NMDA） receptor antagonist, and could be pharmacological induced by application of NMDA. Notably, blockade of protein kinase C（PKC） pathway, 20 Hz facial stimulation failed to induce the MF-GC LTP. In addition, the MF-GC LTP could be enhanced by blocking γ-aminobutyrate A（GABAA） and cannabinoids I（CB1） receptors activities, but was decreased by inhibiting of nitric oxide synthesis. These results indicated that high frequency facial stimulation induced an NMDA receptor dependent, GABAA receptor and NO involved, and PKC signaling required LTP of MFGC synaptic transmission in vivo in mice. Conclusion Our results suggest that the sensory stimulation evoked MF-GC synaptic plasticity might be involved in motor learning in living animals.

Objective To investigate the effect of L-NMMA, an inhibitor of nitric oxide synthase（NOS）, in the hippocampal dentate gyrus（DG） on spatial learning and memory in a rat model of Alzheimer’s disease（AD）. Methods AD model was prepared by ovariectomy combined with intraperitoneal injection of D-galactose. The spatial learning and memory abilities of rats were assessed by Morris water maze（MWM）;the extracellular level of Glu and amplitude of f EPSP in the DG were determined by in vivo microdialysis, HPLC and electrophysiological recording. Results（1） Microinjection of LNMMA into the DG improved the impairment of spatial learning and memory in AD rats.（2） In sham and AD+L-NMMA groups, the concentrations of Glu were significantly increased on the 2 nd and 3 th days of MWM test, and then returned to before-training levels;but in AD group, Glu levels were significantly increased throughout the MWM test period.（3） In sham and AD+L-NMMA groups, the f EPSP amplitudes of DG were significantly increased from the 2 nd to the 5 th days;in AD group, fEPSP amplitude only slightly increased on the 2 nd day. Conclusion Inhibition of NOS in the hippocampal DG improves the spatial learning and memory impairment in AD rats, in part by recovering the responses of Glu and synaptic efficiency in the DG during spatial learning.

Objective The magnocellular neurosecretory cells(MNCs) of the hypothalamic paraventricular nucleus(PVN) integrate incoming signals to secrete oxytocin(OT) and vasopressin(VP) from their nerve terminals in the posterior pituitary *** has been demonstrated that high frequency stimulation(HFS) of afferent fibers induced an NMDA receptor-dependent LTP of excitatory glutamatergic inputs in the supraoptic nucleus(SON) MNCs of rat hypothalamic slices,therefore, the N-methyl-D-aspartic acid(NMDA) receptors-dependent plasticity in PVN MNCs may be involved in nitric oxide(NO) production and PKA signaling ***, the NO and PKA signaling cascades in the long-term plasticity in PVN MNCs is currently *** we used whole-cell patch-clamp recording, single-cell reverse transcription-multiplex polymerase chain reaction(SC-RT-mPCR), biocytin histochemistry and pharmacological methods to examine the mechanism of high frequency stimulus(HFS, 100 Hz)-induced long-term potentiation(LTP) at glutamatergic synapses in the PVN MNCs of juvenile male *** HFS-induced LTP at glutamatergic synapses was accompanied by a decrease in the paired-pulse ratio(PPR) of the PVN *** these MNCs, HFS-induced LTP persisted in the presence of a group 1 metabotropic glutamate receptor(mGluR1) antagonist;however, it was abolished by an NMDA receptor ***, HFS-induced LTP in the PVN MNCs was completely prevented by a nitric oxide synthase(NOS) *** application of an NO donor not only induced the LTP of excitatory glutamatergic inputs in the PVN MNCs, but also occluded the HFS-induced LTP in these ***, HFS-induced LTP in the PVN MNCs was also abolished by a specific protein kinase A(PKA) inhibitor, *** Our results indicate that a HFS can induce an NMDA receptor and NO cascades dependent on presynaptic glutamatergic LTP in the PVN MNCs via a PKA signaling pathway.
