The bio-active components of the Mongolian medicine Horcha-6 and therapeutic mechanism in the rat migraine model

Background:The active components of Horcha-6 were identified using liquid chromatography with tandem mass ***,we investigated the potential mechanisms that explain why Horcha-6 may be effective in treating migraines through the use of network pharmacology and a rat migraine ***:After identifying the active components of Horcha-6,the corresponding genes of the active components’target were obtained from the Universal Protein database,and a“compound-target-disease”network was constructed using Cytoscape 3.9.0 *** the in vivo experiments,nitroglycerin was injected intraperitoneally into rats to create a migraine ***-treatment with Horcha-6 was administered orally for 14 days,and rats were subjected to migraine-related behavior *** sequencing was performed to identify the gene expression regulated by Horcha-6 in the trigeminal ***:A total of 903 chemical components of Horcha-6 have been collected in the liquid chromatography with tandem mass *** discovered 55 of the Horcha-6 bio-active components that were evaluated based on their Percent Human Oral Absorption(≥30%)and DL values(≥0.185)on the traditional Chinese medicine systems pharmacology ***“compound-target-disease”network contained 163 intersection targets with the migraine *** Ontology analysis indicated that these components significantly regulated the immune response,vascular function,oxidative stress,*** Kyoto Encyclopedia of Genes and Genomes enrichment analysis was performed,we observed that most of the target genes were significantly enriched in the inflammation and neuro-related signaling pathway,toll-like receptor signaling pathway,neuroactive ligand-receptor interaction,*** predictions were further demonstrated via in vivo animal model *** RNA sequencing results showed that 41 genes were down-regulated(P<0.05)and 86 genes were up-regulated(P<0.05)in the Horcha-6 treated group compared with the untreat

Promotion of reactive oxygen species activated by nanosilver surface engineering for resistant bacteria-infected skin tissue therapy

Nanosilver has been regarded as a promising alternative to traditional antibiotics for fighting pathogenassociated infections due to its efficacy toward a broad spectrum of ***,bacterial resistance to nanosilver has emerged *** this contribution,a surface engineering strategy based on N-halamine chemistry to address bacterial resistance to nanosilver was *** 1,3-dichloro-5,5-dimethylhydantoin(DCDMH)as an N-halamine source,AgCI nanodots were deposited on the surface of Ag nano wires(Ag NWs)via in situ redox reaction to prepare AgCl-on-Ag *** in vitro and in vivo tests,AgCl-on-Ag NWs effectively inactivated two antibiotic-resistant bacteria,ampicillinresistant Escherichia coli(AREC)and methicillin-resistant Staphylococcus aureus(MRSA)with the minimum bactericidal concentration(MBC)as low as 10μg·ml~(-1)and exhibited good biosafety against normal *** experimental and theoretical tests demonstrated that AgCl-onAg NWs worked on AREC and MAS A by generating high level of reactive oxygen species under visible light irradiation,coupled with the sustained Ag ion ***,the antibacterial mechanism of AgCl-on-Ag NWs against MRSA was verified at the gene level by transcriptome analysis(RNA sequencing).Moreover,the fullthickness defect model verified that AgCl-on-Ag NWs reduced inflammatory cell infiltration and dramatically accelerated wound *** work provides a synergistic mechanism based on nanosilver surface engineering to eradicate the resistant bacteria that can alleviate drug resistance and develop an innovative approach for the treatment of bacterial infections.

Flexible thermoelectric materials are presented with potential applications in electronic devices and energy conversion due to their convenient preparation,good flexibility,and various ***,as ductility is rarely observed in inorganic semiconductors and ceramic insulators,reports on applications of inorganic oxide materials in flexible thermoelectric materials are ***,we report a new method for the synthesis of a flexible Na_(1.4)Co_(2)O_(4) thermoelectric material based on Na_(1.4)Co_(2)O_(4) bulk materials,which are prepared by a self-flux method and painted on print *** coefficient and power factor of the obtained thermoelectric material are 78-102 μVK^(-1) and 159e223 mWm^-(1)K^(-2),respectively,in a temperature range of 303-522 K,which are superior to those values of other conductive polymers and their *** interestingly,the n-type Na_(1.4)Co_(2)O_(4) flexible material is obtained in the painting process at higher pressure with Seebeck coefficients of109 to183 μVK^(-1) in a temperature range of 303-522 *** convenient preparation method of these novel flexible thermoelectric materials may be expanded to the synthesis of other flexible thermoelectric materials,which will be the focus of future work.

Betulin(BE)has exceedingly become a potential natural product,providing multiple pharmacological and biological activi-ties,including anti-cancer,anti-viral,and anti-inflammatory *** research indicated that the solvatomorphism of BE can easily occur through crystallization with different organic *** property of BE can directly affect its extraction,isolation,and preparation *** this study,a system of thermogravimetry(TG)-differential thermal analysis(DTA)coupled with mass spectrometry(MS)with electron ionization(EI)and photoionization(PI)capability,equipped with the skimmer-type interface(i.e.,skimmer-type interfaced TG-DTA-EI/PI-MS system),as a real-time and onsite analysis technique,was ***,four solvatomorphs of BE,namely,with pyridine and water(A),sec-butanol(B),n,n-dimethylformamide(DMF)(C),and isopropanol(V),were analyzed for the first ***,five kinds of the main volatile gaseous species,including H2O,pyridine,sec-butanol,DMF,and isopropanol,were identified ***,the multi-step desolvation processes of the four solvatomorphs of BE were revealed by this system for the first *** system showed great potential for the rapid and accurate analysis of various solvatomorphs of natural products.

OsPDR20 is an ABCG metal transporter regulating cadmium accumulation in rice

Cadmium(Cd)is a non-essential toxic heavy metal,seriously posing high environmental risks to human *** genetic resources relevant to functional genes is important for understanding the metal absorption and accumulation in crops and bioremediation of Cd-polluted *** study investigated a functionally uncharacterized ATP binding cassette transporter G family(ABCG)gene encoding a Pleiotropic Drug Resistance 20(PDR20)type metal transporter which is localized to the plasma membrane of ***20 was transcriptionally expressed in almost all tissues and organs in lifespan and was strongly induced in roots and shoots of young rice under Cd *** expression of OsPDR20 in a yeast mutant ycf1 sensitive to Cd conferred cellular tolerance with less Cd *** of OsPDR20 by RNA interference(RNAi)moderately attenuated root/shoot elongation and biomass,with reduced chlorophylls in rice grown under hydroponic medium with 2 and 10μmol/L Cd,but led to more Cd accumulation.A field trial of rice grown in a realistic Cd-contaminated soil(0.40 mg/kg)showed that RNAi plants growth and development were also compromised compared to wild-type(WT),with smaller panicles and lower spikelet fertility but little effect on yield of ***,OsPDR20 suppression resulted in unexpectedly higher levels of Cd accumulation in rice straw including lower leaves and culm and *** results suggest that OsPDR20 is actively involved in Cd accumulation and homeostasis in rice *** increased Cd accumulation in the RNAi plants has the potential application in phytoremediation of Cd-polluted wetland soils.

Betulin(BE)can be obtained from many plants,such as those belonging Betulaceae family,and pharmacological investigations showed its notable biological properties and good potential for food and pharmaceutical *** investigated the homogeneity,stability,purity,and uncertainty of a newly certified reference material(CRM)of *** certified purity value for the CRM of BE was 99.56%with an extended uncertainty of 0.07%(k=2,P=0.95),as determined by differential scanning calorimetry(DSC).In this study,DSC was used for the first time for purity determination of *** its high accuracy,precision,and reproducibility,DSC can be used as an alternative technique for purity determination of CRMs in the pharmaceutical and food industry.

SNR-adaptive deep joint source-channel coding scheme for image semantic transmission with convolutional block attention module

With the development of deep learning(DL),joint source-channel coding(JSCC)solutions for end-to-end transmission have gained a lot of *** deep JSCC schemes support dynamically adjusting the rate according to different channel conditions during transmission,enhancing robustness in dynamic wireless ***,most of the existing adaptive JSCC schemes only consider different channel conditions,ignoring the different feature importance in the image processing and *** uniform compression of different features in the image may result in the compromise of critical image details,particularly in low signal-to-noise ratio(SNR)*** address the above issues,in this paper,a dual attention mechanism is introduced and an SNR-adaptive deep JSCC mechanism with a convolutional block attention module(CBAM)is proposed,in which matrix operations are applied to features in spatial and channel dimensions *** proposed solution concatenates the pooling feature with the SNR level and passes it sequentially through the channel attention network and spatial attention network to obtain the importance evaluation *** show that the proposed solution outperforms other baseline schemes in terms of peak SNR(PSNR)and structural similarity(SSIM),particularly in low SNR scenarios or when dealing with complex image content.
