Peripheral nerve injuries occur as the result of sudden trauma and lead to reduced quality of *** peripheral nervous system has an inherent capability to regenerate ***,peripheral nerve regeneration following injury is generally slow and incomplete that results in poor functional outcomes such as muscle *** conventional surgical procedures for peripheral nerve injuries present many benefits,there are still several limitations including scarring,difficult accessibility to donor nerve,neuroma formation and a need to sacrifice the autologous *** many years,other therapeutic approaches for peripheral nerve injuries have been explored,the most notable being the replacement of Schwann cells,the glial cells responsible for clearing out debris from the site of *** cultured Schwann cells to the injured sites showed great benefits in promoting axonal regeneration and functional ***,there are limited sources of Schwann cells for extraction and difficulties in culturing Schwann cells in ***,novel therapeutic avenues that offer maximum benefits for the treatment of peripheral nerve injuries should be *** review focused on strategies using mesenchymal stem cells to promote peripheral nerve regeneration including exosomes of mesenchymal stem cells,nerve engineering using the nerve guidance conduits containing mesenchymal stem cells,and genetically engineered mesenchymal stem *** present the current progress of mesenchymal stem cell treatment of peripheral nerve injuries.

Single-cell network analysis reveals gene expression programs for Arabidopsis root development and metabolism

Single-cell RNA-sequencing datasets of Arabidopsis roots have been generated,but related comprehensive gene co-expression network analyses are *** conducted a single-cell gene co-expression network analysis with publicly available scRNA-seq datasets of Arabidopsis roots using a SingleCellGGM *** analysis identified 149 gene co-expression modules,which we considered to be gene expression programs(GEPs).By examining their spatiotemporal expression,we identified GEPs specifically expressed in major root cell types along their developmental *** GEPs define gene programs regulating root cell development at different stages and are enriched with relevant developmental *** examples,a GEP specific for the quiescent center(QC)contains 20 genes regulating QC and stem cell niche homeostasis,and four GEPs are expressed in sieve elements(SEs)from early to late developmental stages,with the early-stage GEP containing 17 known SE developmental *** also identified GEPs for metabolic pathways with cell-type-specific expression,suggesting the existence of cell-type-specific metabolism in *** the GEPs,we discovered and verified a columellaspecific gene,NRL27,as a regulator of the auxin-related root gravitropism *** analysis thus systematically reveals GEPs that regulate Arabidopsis root development and metabolism and provides ample resources for root biology studies.

Emotion and executive control are often conceptualized as two distinct modes of human brain ***,however,is known about how the dynamic organization of large-scale functional brain networks that support flexible emotion processing and executive control,especially their *** amygdala and prefrontal systems have long been thought to play crucial roles in these *** advances in human neuroimaging studies have begun to delineate functional organization principles among the large-scale brain networks underlying emotion,executive control,and their ***,we propose a dynamic brain network model to account for interactive competition between emotion and executive control by reviewing recent resting-state and task-related neuroimaging studies using network-based *** this model,dynamic interactions among the executive control network,the salience network,the default mode network,and sensorimotor networks enable dynamic processes of emotion and support flexible executive control of multiple processes;neural oscillations across multiple frequency bands and the locus coeruleus−norepinephrine pathway serve as communicational mechanisms underlying dynamic synergy among large-scale functional brain *** model has important implications for understanding how the dynamic organization of complex brain systems and networks empowers flexible cognitive and affective functions.

The influence of a mechanical structure's volume increment on the volume power density (VPD) of triboelectric nanogenerators (TENGs) is often neglected when considering surface charge density and surface power density. This paper aims to address this gap by introducing a standardized VPD metric for a more comprehensive evaluation of TENG performance. The study specifically focuses on 2 frequency-up mechanisms, namely, the integration of planetary gears (PG-TENG) and the implementation of a double-cantilever structure (DC-TENG), to investigate their impact on VPD. The study reveals that the PG-TENG achieves the highest volume average power density, measuring at 0.92 W/m^(3). This value surpasses the DC-TENG by 1.26 times and the counterpart TENG by a magnitude of 69.9 times. Additionally, the PG-TENG demonstrates superior average power output. These findings introduce a new approach for enhancing TENGs by incorporating frequency-up mechanisms, and highlight the importance of VPD as a key performance metric for evaluating TENGs.

Transcriptome and proteome analysis reveals the characteristics of carbon metabolism in the mycelial development of Gomphus purpuraceus

Gomphus purpuraceus is an ancient wild edible mushroom. However， the niche and lignocellulose degradation system of G. purpuraceus have remained cryptic. In the current study， transcriptome and proteome sequencing of G. purpuraceus in four mediums was performed. A total of 3489 differentially expressed genes (deGs) and 891 differentially expressed proteins (dePs) were screened out. For the lignocellulose degradation system， a total of 61 expressed genes were annotated， including 19 genes encoding cellulase， 8 genes encoding hemicellulase， 5 genes encoding pectinase， 22 genes encoding lignin oxidase， and 7 lignin degrading auxiliary enzyme. From two-omics， morphology， and enzyme activities， we speculated that G. purpuraceus might have a limited capacity to degrade polysaccharides (starch and lignocellulose). In addition， trace amount of fumonisins (FB1 and FB2) were found in the G. purpuraceus. These results provided a new perspective for understanding the key pathways and hub genes involved in carbon metabolism of G. purpuraceus development.

Virus replication relies on complex interactions between viral *** the case of African swine fever virus(ASFV),only a few such interactions have been identified so *** this study,we demonstrate that ASFV protein p72 interacts with p11.5 using co-immunoprecipitation and liquid chromatography-mass spectrometry(LC-MS).It was found that protein p72 interacts specifically with p11.5 at sites amino acids(aa)1–216 of p72 and aa 1–68 of *** assess the importance of p11.5 in ASFV infection,we developed a recombinant virus(ASFVGZΔA137R)by deleting the A137R gene from the ASFVGZ *** with ASFVGZ,the infectious progeny virus titers of ASFVGZΔA137R were reduced by approximately 1.0 *** addition,we demonstrated that the growth defect was partially attributable to a higher genome copies-to-infectious virus titer ratios produced in ASFVGZΔA137R-infected MA104 cells than in those infected with *** finding suggests that MA104 cells infected with ASFVGZΔA137R may generate larger quantities of noninfectious ***,we found that p11.5 did not affect virus-cell binding or ***,we show for the first time the interaction between ASFV p72 and *** results effectively provide the relevant information of the p11.5 *** results extend our understanding of complex interactions between viral proteins,paving the way for further studies of the potential mechanisms and pathogenesis of ASFV infection.

Three New Hydroxytetradecenals from Amomum tsao-ko with Protein Tyrosine Phosphatase 1B and Glycogen Phosphorylase Inhibitory Activity

The fruits of Amomum tsao-ko(Cao-Guo)were documented in Chinese Pharmacopoeia for the treatment of abdominal pain,vomiting,and *** our previous study,a series of diarylheptanes and flavonoids withα-glucosidase and protein tyrosine phosphatase 1B(PTP1B)inhibitory activity have been reported from the middle-polarity part of ***-ko,whereas the antidiabetic potency of the low-polarity constituents is still *** this study,three new hydroxytetradecenals,(2E,4E,8Z,11Z)-6R-hydroxytetradeca-2,4,8,11-tetraenal(1),(2E,4E,8Z)-6R-hydroxytetradeca-2,4,8-trienal(2)and(2E,4E)-6R-hydroxytetradeca-2,4-dienal(3)were obtained from the volatile oils of *** structures of compounds 1–3 were determined using spectroscopic data involving 1D and 2D nuclear magnetic resonance(NMR),high-resolution mass spectra(HRMS),and specific rotation([α]D).Their hypoglycemic activity was evaluated against glycogen phosphorylase(GPa)and *** 1 and 2 displayed moderate activity against PTP1B with inhibition rates of 33.8%−50.3%at 100 and 200μ***,compound 1 exhibited an obvious inhibitory effect on GPa(IC50=31.7μM),whereas compound 2 was *** study demonstrates hydroxytetradecenals as the characteristic components of ***-ko with therapeutic potential in diabetes.

Glycogen metabolism plays a key role in the development of hepatoellular carcinoma(HCC),but the function of glycogen metabolism genes in the tumor microenvironment(TME)is still to be *** cell RNA-seq data were obtained from ten HCC tumor samples totaling 64,545 cells and 65 glycogen metabolism genes were analyzed bya nonnegative matrix factorization(NMF).The prognosis and immune response of new glycogen TME cell dusters were predicted by using HCC and immunotherapy cohorts from public *** single cell analysis was divided into fibroblasts,NT T cells,macrophages,endothelial clls,and B cells,which were separately divided into new cell clusters by glycogen metabolism gene *** temporal trajectory analysis demonstrated the temporal differentiation trajectory of different glycogen subtype cell *** communication analysis revealed extensive interactions between endothelial cells with glycogen metabolizing TME *** subtypes and diferent glycogen subtype cell *** analysis of transcription factors upstream of TME cell clusters with different glycogen *** addition,TME cell dusters of glycogen metabolism were found to be enriched in expression in CAF subtypes,CD8 depleted,M1,and M2 *** seq analysis showed the prognostic signifcance of glycogen *** TME cell dusters in HCC,while a significant immune response was found in the immunotherapy cohort in patients treated with immune checkpoint blockade(ICB),especially for CAFs,T cells,and macrophages In summary,our study reveals for the first time that glycogen metabolism mediates intercellular communication in the hepatocellular carcinoma microenvironment while elucidating the anti-tumor mechanisms and immune prognostic responses of different subtypes of cell dusters.

INTRODUCTION Sleep disturbance is one of the most common nonmotor symptoms in Parkinson's disease (PD).Sleep disturbance affects 40-98% of PD patients in the world. In China, the prevalence of PD patients with sleep disturbance ranges from 47.66% to 89.10%. Sleep disturbance usually has adverse impact on the quality of life of PD patients. Apossible pathogenesis of PD with sleep disturbance include thalamocortical pathway degeneration and changes of neurotransmitter systems. The etiology of sleep disturbance is multifactorial,involving degeneration of areas regulating sleep,sleep structure affected by drugs,sleep disturbance induced by drug,and sleep fragmentation by multiple factors.
