gPCR-gs mediates the protective effects of ginsenoside Rb1 against oxygen-glucose deprivation/re-oxygenation-induced astrocyte injury

Objectives:To investigate whether the protective actions of ginsenoside Rb1(Rb1)on astrocytes are mediated through the g_(s)-type g-protein-coupled receptor(gPCR-g_(s)).Methods:Primary astrocyte cultures derived from neonatal mouse brain were *** injury was induced via oxygen-glucose deprivation/re-oxygenation(OgD/R).Cell morphology,viability,lactate dehydrogenase(LDH)leakage,apoptosis,glutamate uptake,and brain-derived neurotrophic factor(BDNF)secretion were assessed to gauge cell survival and *** blot was used to investigate the cyclic adenosine monophosphate(cAMP)and protein kinase B(Akt)signaling ***-g_(s)-specific inhibitors and molecular docking were used to identify target ***:Rb1 at concentrations ranging from 0.8 to 5μM did not significantly affect the viability,glutamate uptake,or BDNF secretion in normal ***/R reduced astrocyte viability,increasing their LDH leakage and apoptosis *** also decreased glutamate uptake and BDNF secretion by these ***1 had protective effects of astrocytes challenged by OgD/R,by improving viability,reducing apoptosis,and enhancing glutamate uptake and BDNF ***,Rb1 activated the cAMP and Akt pathways in these *** the gPCR-g_(s) inhibitor NF449 was introduced,the protective effects of Rb1 completely disappeared,and its activation of cAMP and Akt signaling pathways was significantly ***:Rb1 protects against astrocytes from OgD/R-induced injury through gPCR-g_(s) mediation.

Effects of warming and drought stress on the growth characteristics,photosynthetic-transpiration rates,and yield of winter wheat

Climate change has limited crop productivity *** crop response to global climate changes is vital to maintaining agricultural sustainable development.A two-year experiment was conducted to investigate the effects of warming and drought on crop growth and winter wheat yield *** results showed that both warming and drought shortened the crop growth period,reduced the leaf area index,and increased winter wheat biomass *** sufficient water supply conditions,warming would increase photosynthetic and transpiration rates and water use efficiency,while under water deficit conditions,the opposite was *** warming conditions,the grain yield of the water deficit treatment was 8.9%lower than that of the sufficient water supply *** non-warming conditions,the grain yield of water deficit treatment was 12.4%lower than that of the sufficient water *** the conditions of water-sufficient supply,the grain yield of the warming treatment was 4.4%lower than that of the non-warming treatment,and under the conditions of water deficit,the grain yield of the warming treatment was 1.3%lower than that of the non-warming *** tends to decrease wheat growth and grain yield,but sufficient water supply could improve winter wheat’s water use efficiency and reduce the warming limitation on wheat production.

Quantile Regression under Truncated, Censored and dependent Assumptions

In this paper, we focus on the problem of nonparametric quantile regression with left-truncated and right-censored data. Based on Nadaraya-Watson(NW) Kernel smoother and the technique of local linear(LL) smoother, we construct the NW and LL estimators of the conditional quantile. Under strong mixing assumptions, we establish asymptotic representation and asymptotic normality of the estimators. Finite sample behavior of the estimators is investigated via simulation, and a real data example is used to illustrate the application of the proposed methods.

Bone metastasis secondary to breast cancer negatively impacts patient quality of life and *** treatment of bone metastases is challenging since many anticancer drugs are not effectively delivered to the bone to exert a therapeutic *** improve the treatment efficacy,we developed Pluronic P123(P123)-based polymeric micelles dually decorated with alendronate(ALN)and cancer-specific phage protein DMPgTVLP(DP-8)for targeted drug delivery to breast cancer bone ***(DOX)was selected as the anticancer drug and was encapsulated into the hydrophobic core of the micelles with a high drug loading capacity(3.44%).The DOX-loaded polymeric micelles were spherical,123 nm in diameter on average,and exhibited a narrow size *** in vitro experiments demonstrated that a pH decrease from 7.4 to 5.0 markedly accelerated DOX *** micelles were well internalized by cultured breast cancer cells and the cell death rate of micelle-treated breast cancer cells was increased compared to that of free DOX-treated *** binding of the micelles to hydroxyapatite(HA)microparticles indicated their high affinity for bone.P123-ALN/DP-8@DOX inhibited tumor growth and reduced bone resorption in a 3D cancer bone metastasis *** vivo experiments using a breast cancer bone metastasis nude model demonstrated increased accumulation of the micelles in the tumor region and considerable antitumor activity with no organ-specific histological damage and minimal systemic *** conclusion,our study provided strong evidence that these pH-sensitive dual ligand-targeted polymeric micelles may be a successful treatment strategy for breast cancer bone metastasis.

Introduction:Chemokine-like factor 1(CKLF1)is a chemokine that is overexpressed in several *** previousﬁndings revealed a signiﬁcant increase in CKLF1 expression in the ischemic brain,suggesting its potential as a therapeutic target for ischemic ***:In this study,we examined the expression dynamics of CKLF1 in both in vivo and in vitro models of ischemic cardiac *** infarction(MI)was induced in vivo by ligation of the left anterior descending artery(LAD)of the rat *** levels of CKLF1,Creatine Kinase MB Isoenzyme(CK-MB),and Lactate dehydrogenase(LDH)in the serum were detected using Enzyme-linked immunosorbent assay(ELISA).The expression of CKLF1 in the infarcted area was detected by immunohistochemistry,immunoﬂuorescence,quantitative PCR(qPCR),and Western blotting(WB).H9C2 and AC16 cardiomyocytes cultured in vitro were subjected to oxygen and glucose deprivation(OgD).LDH was used to detect cell damage,and CKLF1 expression was detected by qPCR and ***:CKLF1 mRNA and protein expression were signiﬁcantly increased in h9c2 cells at 1.5 h and in AC16 cells at 4 h after *** serum CK-MB in rats increased signiﬁcantly on theﬁrst day after infarction,while the LDH concentration increased signiﬁcantly on the third day after ***1 blood levels signiﬁcantly increased on theﬁrst day following MI in ***1 expression notably increased in the infarct area on days 1,3,and 7 *** MI tissue,CKLF1 colocalizes with cardiomyocytes,macrophages,and ***:CKLF1 was substantially expressed during myocardial ischemia injury both in vivo and in vitro and was colocalized with macrophages and neutrophils,indicating that CKLF1 is expected to be a biomarker and a drug target for the treatment of myocardial infarction.
