Non-coding landscapes of colorectal cancer

For two decades Vogelstein's model has been theparadigm for describing the sequence of molecular changes within protein-coding genes that would lead to overt colorectal cancer(CRC). This model is now too simplistic in the light of recent studies, which have shown that our genome is pervasively transcribed in RNAs other than m RNAs, denominated non-coding RNAs(nc RNAs). The discovery that mutations in genes encoding these RNAs [i.e., micro RNAs(mi RNAs), long non-coding RNAs, and circular RNAs] are causally involved in cancer phenotypes has profoundly modified our vision of tumour molecular genetics and pathobiology. By exploiting a wide range of different mechanisms, nc RNAs control fundamental cellular processes, such as proliferation, differentiation, migration, angiogenesis and apoptosis: these data have also confirmed their role as oncogenes or tumor suppressors in cancer development and progression. The existence of a sophisticated RNA-based regulatory system, which dictates the correct functioning of protein-coding networks, has relevant biological and biomedical consequences. different mi RNAs involved in neoplastic and degenerative diseases exhibit potential predictive and prognostic properties. Furthermore, the key roles of nc RNAs make them very attractive targets for innovative therapeutic approaches. Several recent reports have shown that nc RNAs can be secreted by cells into the extracellular environment(i.e., blood and other body fluids): this suggests the existence of extracellular signalling mechanisms, which may be exploited by cells in physiology and pathology. In this review, we will summarize the most relevant issues on the involvement of cellular and extracellular nc RNAs in disease. We will then specifically describe their involvement in CRC pathobiology and their translational applications to CRC diagnosis, prognosis and therapy.

Introduction:This paper presents a case study of the application of an ecosystem services framework to support land-use *** district of Koboko,in northwestern Uganda,formed the study *** overall purpose was to improve the understanding of links between human actions,their impact on ecosystems and the services they provide,and,ultimately,consequences for human *** framework developed by the Millennium Ecosystem Assessment was used as a main conceptual *** analysis comprised four main stages:(1)identification of the main ecosystems and their services;(2)analysis of human wellbeing components,focusing on poverty issues;(3)assessment of the interrelations between ecosystem services and human wellbeing;(4)identification of the main drivers of change and development of future spatial development ***:The scenarios were developed starting from the main drivers of change and show the dependency of human wellbeing on ecosystems services,identifying how spatial development strategies can affect that *** information was not easily available,therefore scenario development made use of both quantitative and qualitative *** results of qualitative and quantitative approaches were consistent,proving the strength and flexibility of the ***:Scenario results aim to provide guidance for local government land-use planning,focusing on the promotion of sustainability through ecosystem services *** spatially explicit analysis illustrated how different policies affect urban development;it was clearly shown that even with very different demographic outcomes,the impact of sectorial policies guaranteed a good level of suitability for new residential areas.

Proof of concept of a self-tightening needle-less suture using a NiTi shapememory alloy

Among surgical procedures,suturing is considered ***,in some situations,suturing is not easy or ***,we present proof of concept of a new self-tightening needle-less suture made of a NiTi *** new device is designed to overcome two demanding steps of traditional suturing:needle introduction and manual *** novel suture is composed only of a short NiTi wire that is able to change shape by exploiting a thermoelastic phase *** shape change is achieved by increasing the temperature from that of the operating room(e.g.,20-22℃)to 32℃using warm *** consists of two steps:suture introduction,in which the suture is in an open configuration,and shape recovery(i.e.,closed configuration).The closed configuration is maintained at human body temperature thanks to peculiar pseudoelastic properties of the NiTi ***,thermal and functional characterization verified the simplicity and effectiveness of the proposed device.

Influence of data analysis when exploiting DFN model representation in the application of rock mass classification systems

discrete fracture network(DFN) models have been proved to be effective tools for the characterisation of rock masses by using statistical distributions to generate realistic three-dimensional(3 D) representations of a natural fracture network. The quality of DFN modelling relies on the quality of the field data and their interpretation. In this context, advancements in remote data acquisition have now made it possible to acquire high-quality data potentially not accessible by conventional scanline and window mapping. This paper presents a comparison between aggregate and disaggregate approaches to define fracture sets, and their role with respect to the definition of key input parameters required to generate DFN models. The focal point of the discussion is the characterisation of in situ block size distribution(IBSD) using DFN methods. An application of IBSD is the assessment of rock mass quality through rock mass classification systems such as geological strength index(GSI). As DFN models are becoming an almost integral part of many geotechnical and mining engineering problems, the authors present a method whereby realistic representation of 3 D fracture networks and block size analysis are used to estimate GSI ratings, with emphasis on the limitations that exist in rock engineering design when assigning a unique GSI value to spatially variable rock masses.

A PARALLEL ALGORITHM FOR THE SOLUTION OF LARGE-SCALE NONCONFORMING FLUID-STRUCTURE INTERACTION PROBLEMS IN HEMODYNAMICS

In this work we address the numerical solution of large scale fluid-structure interaction problems when nonconforming grids and/or nonconforming finite elements discretizations are used at the interface separating the fluid and structure physical domains. To deal with nonconforming fluid-structure discretizations we use the INTERNODES method （INTER- polation for NOnconforming DEcompositionS） formerly introduced in [6] for the solution of elliptic PDEs on nonconforming domain decomposition. To cope with the high com- putational complexity of the three dimensional FSI problem obtained after spatial and temporal discretization, we use the block parallel preconditioner FaCSI [7]. A numerical investigation of the accuracy properties of INTERNODES applied to the nonconforming FSI problem is carried out for the simulation of the pressure wave propagation in a straight elastic cylinder. Finally, we study the scalability performance of the FaCSI precondition- er in the nonconforming case by solving a large-scale nonconforming FSI problem in a patient-specific arterial bypass.

New approaches to quantify progressive damage and associated dynamic rock mass blockiness

In the past decade, numerical modelling has been increasingly used for simulating the mechanical behaviour of naturally fractured rock masses. In this paper, we introduce new algorithms for spatial and temporal analyses of newly generated fractures and blocks using an integrated discrete fracture network (DFN)-finite-discrete element method (FDEM) (DFN-FDEM) modelling approach. A fracture line calculator and analysis technique (i.e. discrete element method (DEM) fracture analysis, DEMFA) calculates the geometrical aspects of induced fractures using a dilation criterion. The resultant two-dimensional (2D) blocks are then identified and characterised using a graph structure. Block tracking trees allow track of newly generated blocks across timesteps and to analyse progressive breakage of these blocks into smaller blocks. Fracture statistics (number and total length of initial and induced fractures) are then related to the block forming processes to investigate damage evolution. The combination of various proposed methodologies together across various stages of modelling processes provides new insights to investigate the dependency of structure's resistance on the initial fracture configuration.

Overhanging rock slope by design:An integrated approach using rock mass strength characterisation,large-scale numerical modelling and limit equilibrium methods

Overhanging rock slopes(steeper than 90°) are typically avoided in rock engineering design, particularly where the scale of the slope exceeds the scale of fracturing present in the rock mass. This paper highlights an integrated approach of designing overhanging rock slopes where the relative dimensions of the slope exceed the scale of fracturing and the rock mass failure needs to be considered rather than kinematic release of individual blocks. The key to the method is a simplified limit equilibrium(LE) tool that was used for the support design and analysis of a multi-faceted overhanging rock slope. The overhanging slopes required complex geometries with constantly changing orientations. The overhanging rock varied in height from 30 m to 66 m. Geomechanical modelling combined with discrete fracture network(DFN)representation of the rock mass was used to validate the rock mass strength assumptions and the failure mechanism assumed in the LE model. The advantage of the simplified LE method is that buttress and support design iterations(along with sensitivity analysis of design parameters) can be completed for various cross-sections along the proposed overhanging rock sections in an efficient manner, compared to the more time-intensive, sophisticated methods that were used for the initial validation. The method described presents the development of this design tool and assumptions made for a specific overhanging rock slope design. Other locations will have different geological conditions that can control the potential behaviour of rock slopes, however, the approach presented can be applied as a general guiding design principle for overhanging rock cut slope.

Drosophila is also an advantageous system regarding the animal husbandry and the short generation time and lifespan.
