Stress enhances expression of calcium-binding proteins and NMDAR subunit genes in the rat hippocampus

Backgrounds:Oxidative stress impairs the function of calcium-binding proteins and deregulates calcium signaling in living *** have previously explored the overexpression of calcium-binding protein genes in a reactive oxygen and nitrogen species-induced in vitro cell model of stress that leads to ***,in in vivo models,low levels of stress leads to depressive-like ***,we aimed to analyze gene expression of major calcium-binding proteins(calcineurin,calmodulin,calsyntenin,synaptotagmin,and calreticulin)and N-methyl-D-aspartic acid(NMDA)receptor subunits(glutamate receptor ionotropic[GluN]GluN1,GluN2A,and GluN2B)in the hippocampus of stress-induced ***:Six-week-old male Wistar rats were assigned to two stress induction groups and a control group without stress(n=6).Stress was induced by using H_(2)O_(2)(3%in water)or by immobilization(using a sticky mat)over a period of 30 *** of calcium-binding protein genes in the hippocampus,antioxidant assays,structural alterations in hippocampal neurons,and depressive-like behavior were ***:Expression of genes encoding calcium-binding proteins calcineurin,calsyntenin,synaptotagmin and NMDA receptor subunit GluN1 was enhanced in both chemical and physical stress-induced rats compared with control rats(4.25±0.05 vs.1.03±0.02,ppppppced rats,neurons in the cA2 region of the hippocampus were fewer and appeared disorganized compared with control ***,stressinduced rats showed decreased mobility and lower sucrose preference in behavioral studies compared with control ***:Lower levels of reactive oxygen and nitrogen species(RONS)can also lead to stress in rats by affecting their calcium signaling,buffering capacity in the neurons leading to depressive symptoms.

Background Our previous study has reported that supplementation of oligosaccharide-based polymer enhances gut health and disease resistance of pigs infected with enterotoxigenic ***(ETEc)F18 in a manner similar to *** objective of this study was to investigate the impacts of oligosaccharide-based polymer or antibiotic on the host metabolic profiles and colon microbiota of weaned pigs experimentally infected with ETEc *** Multivariate analysis highlighted the differences in the metabolic profiles of serum and colon digesta which were predominantly found between pigs supplemented with oligosaccharide-based polymer and *** relative abundance of metabolic markers of immune responses and nutrient metabolisms,such as amino acids and carbohydrates,were significantly differentiated between the oligosaccharide-based polymer and antibiotic groups(qchange>2.0).In addition,pigs in antibiotic had a reduced(pce of Lachnospiraceae and Lactobacillaceae,whereas had greater(pclostridiaceae and Streptococcaceae in the colon digesta on d 11 post-inoculation(pI)compared with d 5 *** The impact of oligosaccharide-based polymer on the metabolic and microbial profiles of pigs is not fully understood,and further exploration is ***,current research suggest that various mechanisms are involved in the enhanced disease resistance and performance in ETEc-challenged pigs by supplementing this polymer.

Spinal cord injury(ScI)is a devastating and disabling medical condition generally caused by a traumatic event(primary injury).This initial trauma is accompanied by a set of biological mechanisms directed to ameliorate neural damage but also exacerbate initial damage(secondary injury).The alterations that occur in the spinal cord have not only local but also systemic consequences and virtually all organs and tissues of the body incur important changes after ScI,explaining the progression and detrimental consequences related to this ***(pNIE)is a growing area of research aiming to integrate and explore the interactions among the different systems that compose the human organism,considering the mind and the body as a *** initial traumatic event and the consequent neurological disruption trigger immune,endocrine,and multisystem dysfunction,which in turn affect the patient's psyche and *** the present review,we will explore the most important local and systemic consequences of ScI from a pNIE perspective,defining the changes occurring in each system and how all these mechanisms are ***,potential clinical approaches derived from this knowledge will also be collectively presented with the aim to develop integrative therapies to maximize the clinical management of these patients.

BAcKGROUND Metabolic dysfunction-associated steatotic liver disease(MASLD),characterised by hepatic lipid accumulation,causes inflammation and oxidative stress accompanied by cell damage and *** injury(LI)is also frequently reported in patients hospitalised with coronavirus disease 2019(cOVID-19),while preexisting MASLD increases the risk of LI and the development of cOVID-19-associated *** of injury at the cellular level remain unclear,but it may be significant that severe acute respiratory syndrome coronavirus 2(SARS-coV-2)which causes cOVID-19,uses angiotensin-converting expression enzyme 2(AcE2),a key regulator of the‘anti-inflammatory’arm of the renin-angiotensin system,for viral attachment and host cell *** To determine if hepatic AcE2 levels are altered during progression of MASLD and in patients who died with severe *** AcE2 protein levels and localisation,and histological fibrosis and lipid droplet accumulation as markers of MASLD were determined in formalin-fixed liver tissue sections across the MASLD pathological spectrum(isolated hepatocellular steatosis,metabolic dysfunction-associated steatohepatitis(MASH)+/-fibrosis,end-stage cirrhosis)and in post-mortem tissues from patients who had died with severe cOVID-19,using AcE2 immunohistochemistry and haematoxylin and eosin and picrosirius red staining of total collagen and lipid droplet areas,followed by quantification using machine learning-based image pixel *** AcE2 staining is primarily intracellular and concentrated in the cytoplasm of centrilobular hepatocytes and apical membranes of bile duct ***,AcE2 protein levels are elevated in non-fibrotic MASH compared to healthy controls but not in the progression to MASH with fibrosis and in ***2 protein levels and histological fibrosis are not associated,but AcE2 and liver lipid droplet content are significantly correlated across the MASLD *** A

The SARS-coV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOc)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOcD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUc¼0.91,95%cI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.
