Previously,we employed a proteomics-based 2-D gel electrophoresis assay to show that exposure to 10 u.M benzo(a)pyrene(BaP) during a 24 h frame can lead to changes in nuclear protein expression and alternative *** further expand our knowledge about the DNA damage response(DDR) induced by BaP,we investigated the nuclear protein expression profiles in HeLa cells treated with different concentrations of BaP(0.1,1,and 10μM) using this proteomics-based 2-D gel electrophoresis *** found 125 differentially expressed proteins in BaP-treated cells compared to control *** them,79(63.2%) were down-regulated,46(36.8%) were up-regulated;8 showed changes in the 1 u.M and 10 u.M BaP-treated groups,2 in the 0.1μM and 10μM BaP-treated groups,4 in the 0.1μM and 1μM BaP-treated groups,and only one showed changes in all three *** protein spots were chosen for liquid chromatography-tandem mass spectrometry(LC-MS/MS) identification,and of these,39 were identified,including subunits of the 26S proteasome and Annexin *** functions of some identified proteins were further examined and the results showed that they might be involved in BaP-induced *** together,these data indicate that proteomics is a valuable approach in the study of environmental chemical-host interactions,and the identified proteins could provide new leads for better understanding BaP-induced mutagenesis and carcinogenesis.

In south-eastern Australia,the same baited,round traps(comprising 50–57-mm mesh netting)are used to target giant mud,Scylla serrata and blue swimmer crabs,Portunus armatus in spatially separated *** fisheries are characterised by the common,problematic discarding of undersized portunids(<85 and 65 mm carapace length;CL for *** and ***)and fish(yellowfin bream,Acanthopagrus australis).This poor selectivity was addressed here in two experiments assessing the utility of(1)traps partially or completely covered in larger mesh(91 mm to match the minimum legal size of the smaller ***),and then(2)any cumulative benefits of fitting species-specific escape *** experiment 1,there were no differences among catches of legal-sized portunids associated with either partial,or complete trap coverage with larger *** of mesh coverage,both designs of 91-mm traps also retained significantly fewer(by up to 42%)undersized *** and *** experiment 2,replicate traps completely covered in 91-mm mesh were tested against conventional traps comprising 56-mm mesh,and traps with the same mesh sizes,but also three escape gaps configured for either ***(46×120 mm)or ***(36×120 mm)(*** treatments in total).All modified traps maintained catches of legal-sized ***,and only the 91-mm traps with escape gaps caught fewer legal-sized *** undersized ***,*** and ***(mean catches reduced by up to 49%)were retained in all larger-meshed than small-meshed traps,and in all of those traps with escape gaps(by up to 95%)than *** there were no significant cumulative benefits of escape gaps in largermeshed traps(measured by a statistical interaction),there was a trend of fewer unwanted catches *** data support configuring portunid traps with mesh sizes matching the morphology of the smallest legal-sized target ***,simply retroactively fitting escape gaps in existing,smaller-m

AIM: To test the hypotheses that diffusion weighed (DW)and transcatheter intraarterial perfusion (TRIP)magnetic resonance imaging (MRI) can each be used to assess regional differences in tumor function in an animal pancreatic cancer ***: VX2 tumors were implanted in pancreata of 6 rabbits. MRI and digital subtraction angiography (DSA) were performed 3 wk following implantation. With a 2-French catheter secured in the rabbit's gastroduodenal artery, each rabbit was transferred to an adjacent 1.5T MRI scanner. DWand TRIP-MRI were performed to determine if necrotic tumor core could be differentiated from viable tumor periphery. For each, we compared mean differences between tumor core/ periphery using a 2-tailed paired t-test (α = 0.05). Imaging was correlated with histopathology. RESULTS: Tumors were successfully grown in all rabbits, confirmed by necropsy. On DW-MRI, mean apparent diffusion coeffi cient (ADC) value was higher in necrotic tumor core (2.1 ± 0.3 mm2/s) than in viable tumor periphery (1.4 ± 0.5 mm2/s) (P < 0.05). On TRIP-MRI, mean perfusion values was higher in tumor periphery (110 ± 47 relative units) than in tumor core (66 ± 31 relative units) (P < 0.001). CONCLUSION: Functional MRI can be used to differentiate necrotic from viable tumor cells in an animal pancreatic cancer model using ADC (DW-MRI) and perfusion (TRIP-MRI) values.

Feasibility of pancreatectomy following high-dose proton therapy for unresectable pancreatic cancer

AIM To review surgical outcomes for patients undergoing pancreatectomy after proton therapy with concomitant capecitabine for initially unresectable pancreatic *** From April 2010 to September 2013,15 patients with initially unresectable pancreatic cancer were treated withproton therapy with concomitant capecitabine at 1000 mg orally twice daily. All patients received 59.40 Gy(RBE) to the gross disease and 1 patient received 50.40 Gy(RBE) to high-risk nodal targets. There were no treatment interruptions and no chemotherapy dose reductions. Six patients achieved a radiographic response sufficient to justify surgical exploration,of whom 1 was identified as having intraperitoneal dissemination at the time of surgery and the planned pancreatectomy was aborted. Five patients underwent resection. Procedures included:Laparoscopic standard pancreaticoduodenectomy(n = 3),open pyloris-sparing pancreaticoduodenectomy(n = 1),and open distal pancreatectomy with irreversible electroporation(IRE) of a pancreatic head mass(n = 1). RESULTS The median patient age was 60 years(range,51-67). The median duration of surgery was 419 min(range,290-484),with a median estimated blood loss of 850 cm^3(range,300-2000),median ICU stay of 1 d(range,0-2),and median hospital stay of 10 d(range,5-14). Three patients were re-admitted to a hospital within 30 d after discharge for wound infection(n = 1),delayed gastric emptying(n = 1),and ischemic gastritis(n = 1). Two patients underwent R0 resections and demonstrated minimal residual disease in the final pathology specimen. One patient,after negative pancreatic head biopsies,underwent IRE followed by distal pancreatectomy with no tumor seen in the specimen. Two patients underwent R2 resections. Only 1 patient demonstrated ultimate local progression at the primary site. Median survival for the 5 resected patients was 24 mo(range,10-30).CONCLUSION Pancreatic resection for patients with initially unresectable cancers is feasible after
