An Exome-seq Based Tool for Mapping and Selection of Candidate Genes in Maize Deletion Mutants

Despite the large number of genomic and transcriptomic resources in maize, there is still much to learn about the function of genes in developmental and biochemical processes. Some maize mutants that were generated by gamma-irradiation showed clear segregation for the kernel phenotypes in B73 ? Mo17 F2 ears. To better understand the functional genomics of kernel development,we developed a mapping and gene identi?cation pipeline, bulked segregant exome sequencing(BSEx-seq), to map mutants with kernel phenotypes including opaque endosperm and reduced kernel size. BSEx-seq generates and compares the sequence of the exon fraction from mutant and normal plant F2 DNA pools. The comparison can derive mapping peaks, identify deletions within the mapping peak, and suggest candidate genes within the deleted regions. We then used the public kernel-speci?c expression data to narrow down the list of candidate genes/mutations and identi?ed deletions ranging from several kb to more than 1 Mb. A full deletion allele of the Opaque-2 gene was identi?ed in mutant 531, which occurs within a $200-kb deletion. Opaque mutant 1486 has a 6248-bp deletion in the mapping interval containing two candidate genes encoding RNA-directed DNA methylation 4(RdDM4) and AMP-binding protein, respectively. This study demonstrates the ef?-ciency and cost-effectiveness of BSEx-seq for causal mutation mapping and candidate gene selection,providing a new option in mapping-by-sequencing for maize functional genomics studies.

Fast and sensitive antigen detection is important in biomedical research and development. Despite, being invented 48 years ago, the enzyme-linked immunosorbent assay(ELISA) remains one of the most successful and widely employed bioanalytical techniques in research and clinical diagnostics due to its reliability and simplistic design. Recently, nanotechnology has offered efficient signal reporting. In spite of some improvements in these systems, there are typically increased material costs involved and the need for expensive equipment or complicated chemical processes, thus negating any possible benefits over ELISA. Herein, we communicate a simple Cu-DNAzyme system for signal transduction of a CuO nanoparticle-labeled immunoassay. The reported immunoassay amplifies signal generation similar to traditional ELISA and is fast, simple, cost-effective, and sensitive, holding promise for biomedical applications and point-of-care testing.

Structural development of vegetation on rehabilitated North Stradbroke Island:Above/belowground feedback may facilitate alternative ecological outcomes

Incorporation of Biochar Carbon into Stable Soil Aggregates:The Role of Clay Mineralogy and Other Soil Characteristics

Aggregation and structure play key roles in water-holding capacity and stability of *** this study,the incorporation of carbon(C) from switchgrass biochar into stable aggregate size fractions was assessed in an Aridisol(from Colorado,USA) dominated by 2:1 clays and an Alfisol(from Virginia,USA) containing weathered mixed 1:1 and 2:1 mineralogy,to evaluate the effect of biochar addition on soil *** biochar was applied at 4 levels,0,25,50,and 100 g kg^(-1),to the soils grown with wheat in a growth chamber *** changes in soil strength and water-holding capacity using water release curves were *** the Colorado soil,the proportion of soil occurring in large aggregates decreased,with concomitant increases in small size *** changes in aggregate size fractions occurred in the Virginia *** the Colorado soil,C content increased from 3.3 to 16.8 g kg^(-1),whereas in the2 000 μm *** greatest increase(from 6.2 to 22.0 g kg^(-1)) occurred in the 53–250 μm *** results indicated that C was incorporated into larger aggregates in the Virginia soil,but remained largely unassociated to soil particles in the Colorado *** addition had no significant effect on water-holding capacity or strength *** biochar to more weathered soils with high native soil organic content may result in greater stabilization of incorporated C and result in less loss because of erosion and transport,compared with the soils dominated by 2:1 clays and low native soil organic content.

＜正＞Maternal cigarette smoking and prenatal nicotine exposure increase the risk for sudden infant death syndrome （SIDS） by 2- to 4-fold,yet despite adverse publicity,nearly one of four pregnant women smoke tobacco in *** who succumb to SIDS typically experience a severe bradycardia that precedes or is accompanied by centrally mediated life-threatening apneas and *** the causes of the apnea and bradycardia prevalent in SIDS victims are unknown,it has been hypothesized that these fatal events are exaggerated cardiorespiratory responses to hypoxia or *** in heart rate are primarily determined by the activity of cardiac vagal neurons（CVNs） in the brainstem. Nicotinic receptors play an important role in modulating the activity of parasympathetic CVNs in the *** of neostigmine（10μM） significantly increased the holding current,amplitude,and frequency of miniature excitatory postsynaptic current（mEPSC） glutamatergic events in *** unexposed animals,the nicotine-evoked facilitation of mEPSC frequency,but not mEPSC amplitude or holding current,was blocked byα-bungarotoxin（100 nM）.Prenatal nicotine exposure significantly exaggerated and altered the types of nicotinic receptors involved in these *** prenatal nicotine-exposed animals,α-bungarotoxin only partially reduced the increase in mEPSC *** addition,in prenatal nicotine-exposed animals,the increase in holding current was partially dependent onα-7 subunit-containing nicotinic receptors,in contrast to unexposed animals in which a-bungarotoxin had no ***/hypercapnia did not elicit an increase in excitatory neurotransmission to CVNs in unexposed animals,but in animals that were exposed to nicotine in the prenatal period,hypoxia/hypercapnia recruited an excitatory neurotransmission to ***- β- erythroidine at a concentration of 100μM totally blocked the exaggerated action of CVNs induced by hypoxia/hypercapnia in animals that were expose

Toxaemia secondary to pyloric foreign body obstruction in two African lion(Panther leo) cubs

A case of toxaemia secondary to pyloric foreign body obstruction in two four-month-old African lion cubs were presented in this article. The lion cubs were presented to the school of veterinary medicine with a complaint of weight loss and stunted growth despite having a normal appetite and seizures. Defi nitive diagnosis was made based on gross pathology after attempting various symptomatic treatments. This article therefore is meant to discourage the use of blankets as bedding in holding enclosures for warmth and comfort post-weaning in captive lion cubs and indeed wild cats in general as they tend to eat bedding that has been soiled with food.

Implications of step-chilling on meat color investigated using proteome analysis of the sarcoplasmic protein fraction of beef longissimus lumborum muscle

In order to improve beef color and color stability, step-chilling （SC） was applied on excised bovine Iongissimus lumborum muscle, with chilling starting at 0-4℃ for 5 h, then holding the temperature at 12-18℃ for 6 h, followed by 0-4℃ again until 24 h post-mortem, pH and temperature were measured during rigor on SC loins as well as those subjected to routine chilling （RC, 0-4℃, till 24 h post-mortem）. Color L＊, a＊, b＊ values, metmyoglobin （MetMb） content, MetMb reducing ability （MRA） and NADH content were determined on samples aged for 1, 7, and 14 d. Sarcoplasmic proteome analysis was only conducted on d 1 samples. The results showed muscles subjected to SC maintained a temperature at around 15℃ for 5 to 10 h post-mortem, and exhibited a slow temperature decline, but rapid pH decline. Beef steaks treated with SC had higher L＊, a＊, b＊ and chroma values than those of RC samples at 1 and 7 d chilled storage （0-4℃）, while showing no significant difference for a＊, b＊ and chroma values at d 14. The SC samples also exhibited a lower relative content of surface MetMb, higher MRA and NADH content, compared with RC beef steaks during storage, indicating the SC-treated beef showed an improved color stability. Eleven differential protein spots/nine proteins were identified by two-dimensional gel electrophoresis and mass spectrometry, and those proteins were mainly involved in redox, chaperone binding, metabolic and peroxidase activity. Oxidoreductases play a role in decreasing the oxidation-induced myoglobin oxidation and benefiting the production of NADH, and finally improving the colour of beef. Of these, pyruvate dehydrogenase E1 component subunit beta showed a positive correlation with color L＊, a＊, b＊ values and accounted for more than 60% of the variation in color values; this protein can be considered as a potential beef color biomarker. Grip Force Using an Artificial Limb in a Congenital Amputee

While much attention is being given in the application of advanced technologies to improve upper extremity prostheses, traditional body-powered prostheses still remain the most popular by people with an amputation. A body-powered prosthesis provides the user with a reasonable solution for limb loss given their simple design, lower maintenance and initial cost. The two major types of body-powered prosthesis use either voluntary opening or voluntary closing control of the terminal device (or prehensor) used for holding and manipulating objects. What differentiates these two types of control is the relationship between the muscular force used to apply tension on the cable attached to the prehensor and the force produced by the prehensor. It has been argued that the voluntary closing prosthesis has more optimal compatibility between the muscle force and grip force of the prehensor. As a result, it may provide an advantage to the user in tasks requiring the control of grip force. To determine the effectiveness of the voluntary closing and voluntary opening prosthesis, we asked a person with a congenital quadruple limb deficiency who is right hand dominant, and that uses voluntary opening prostheses to participate in a study investigating grip force control. The participant was required to match different target grip forces displayed on a computer monitor by manipulating the pressure exerted on a hand dynamometer using either a voluntary closing or voluntary opening prosthesis. The participant only had previous experience with a voluntary opening prosthesis. The results showed that in several measures, the participant performed better with the voluntary closing prosthesis. These results provided support for the muscular force-grip force compatibility hypothesis.
