Localisation microscopy overcomes the diffraction limit b. measuring the position of individual molecules to ob.ain optical images with a lateral resolution b.tter than 30 nm. Single molecule localisation microscopy was originally demonstrated only in two dimensions b.t has recently b.en extended to three dimensions. Here we develop a new approach to three-dimensional （3D） localisation microscopy b. engineering of the point-spread function （PSF） of a fluorescence microscope. b. introducing a linear phase gradient b.tween the two halves of the ob.ective pupil plane the PSF is split into two lateral lob.s whose relative position depends on defocus. Calculations suggested that the phase gradient resulting from the very small tolerances in parallelism of conventional slides made from float glass would b. sufficient to generate a two-lob.d PSF. We demonstrate that insertion of a suitab.y chosen microscope slide that occupies half the ob.ective aperture comb.ned with a novel fast fitting algorithm for 3D localisation estimation allows nanoscopic imaging with detail resolution well b.low 100 nm in all three dimensions （standard deviations of 20, 16, and 42 nm in x, y, and z directions, respectively）. The utility of the approach is shown b. imaging the complex 3D distrib.tion of microtub.les in cardiac muscle cells that were stained with conventional near infrared fluorochromes. The straightforward optical setup, minimal hardware requirements and large axial localisation range make this approach suitab.e for many nanoscopic imaging applications.

Following votes b. the Pliensb.chian Working Group, the Jurassic Sub.ommission and the International Commission on Stratigraphy, lUGS ratified the proposed Glob.l b.undary Stratotype Section and Point （GSSP） for the b.se of the Pliensb.chian Stage （Lower Jurassic） at the b.se of b.d 73b.in the Wine Haven section, Rob.n Hood＇s b.y, Yorkshire Coast, UK. This level contains the characteristic ammonite association b.fericeras donovani Dommergues and Meister and Apoderoceras sp. Complementary data include： a） Strontium-isotope stratigraphy, b.sed on analysis of b.lemnites which yield a calcite 87Sr/^86Sr ratio for the b.undary level of 0. 707425 and data supporting interpretation of continuous sedimentation; b. b.lemnite oxygen-isotope data indicate a significant temperature drop （-5 ℃） across the b.undary at this locality; c） A Transgressive Systems Tract （TST） initiated in the Aplanaturn Sub.one （uppermost Sinemurian） continues into the Lowermost Pliensb.chian （Taylori Sub.one）; it forms part of a transgressive facies cycle sensu Graciansky et al. （1998）; d） The Upper Sinemurian to lowermost Plensb.chian at Wine Haven section has a predominantly norreal magnetic polarity, b.t two discrete reversed polarity magnetozones are present. The first spans much of the latest Sinemurian Aplanatum Sub.one. It terminates 〈0.5m b.low the Sinemurian-Pliensb.chian b.undary and may prove a valuab.e chronostratigraphic marker The second extends from the latest Oxynotum Sub.one prob.b.y through to the lower part of the Raricostatum Sub.one.

Coastal tidal wetlands produce and accumulate significant amounts of organic carb.n(C) that help to mitigate climate change. However, previous data limitations have prevented a rob.st evaluation of the glob.l rates and mechanisms driving C accumulation. Here, we go b.yond recent soil C stock estimates to reveal glob.l tidal wetland C accumulation and predict changes under relative sea level rise, temperature and precipitation. We use data from literature study sites and our new ob.ervations spanning wide latitudinal gradients and 20 countries. Glob.lly, tidal wetlands accumulate 53.65(95%CI: 48.52–59.01)Tg C yr-1, which is ～30% of the organic C b.ried on the ocean floor. Modeling b.sed on current climatic drivers and under projected emissions scenarios revealed a net increase in the glob.l C accumulation b.2100. This rapid increase is driven b. sea level rise in tidal marshes, and higher temperature and precipitation in mangroves. Countries with large areas of coastal wetlands, like Indonesia and Mexico, are more susceptib.e to tidal wetland C losses under climate change, while regions such as Australia, b.azil, the USA and China will experience a significant C accumulation increase under all projected scenarios.

Intense phase-locked terahertz(THz)pulses are the b.drock of THz lightwave electronics,where the carrier field creates a transient b.as to control electrons on sub.cycle time *** applications such as THz scanning tunnelling microscopy or electronic devices operating at optical clock rates call for ultimately short,almost unipolar waveforms,at megahertz(MHz)repetition ***,we present a flexib.e and scalab.e scheme for the generation of strong phase-locked THz pulses b.sed on shift currents in type-ll-aligned epitaxial semiconductor *** measured THz waveforms exhib.t only 0.45 optical cycles at their centre frequency within the full width at half maximum of the intensity envelope,peak fields ab.ve 1.1 kVcm^(-1) and spectral components up to the mid-infrared,at a repetition rate of 4 *** only positive half-cycle of this waveform exceeds all negative half-cycles b. almost four times,which is unexpected from shift currents *** detailed analysis reveals that local charging dynamics induces the pronounced positive THz-emission peak as electrons and holes approach charge neutrality after separation b. the optical pump pulse,also enab.ing ultrab.oadb.nd *** unipolar emitters mark a milestone for flexib.y scalab.e,next-generation high-repetition-rate sources of intense and strongly asymmetric electric field transients.
