This review covers the recent history of a series of very important natural products and their derivatives that are currently in use or under evaluation in the areas of anti-infectives, important cancer treatments that include antibody drug conjugates, followed by a discussion of type 2 diabetes(T2DM) drugs and angiotensin converting enzyme inhibitors. The current structures of the agents are shown, though in the case of some peptides used in T2DM drugs the standard single leTher abbreviation for an amino acid is used.

The development of a multiple level underground mine is a complex task in which geology,engineering,ground control,and unit operations are integrated into a single mine *** components are interdependent and must function cohesively to ensure a profitable underground mining *** optimize reserve recovery,mine planning should begin from the lowest level and progress *** limits any misjudgments or oversights of a given level affecting the underlying levels and ensures the ability to maximize recovery from each *** planning should start with the exploration and characterization of the geologic *** the reserve geology and quality is well understood,then mine planning can begin with respect to the following:(1)orientation of mine works with respect to horizontal stress;(2)access to the reserve;(3)determination of opening widths;(4)selection of back,floor horizons and pillar centers;(5)selection of development and secondary mining heights;(6)appropriate inter-burden thicknesses;and(7)examining the stability of the multiple level mine through numerical *** multiple level mine design process and decisions are presented through a case history *** theme is that there is one opportunity to"get it right"and many chances to overlook a small aspect within the design that will plague the mine throughout all levels and through the entirety of its operating life.

The massively parallel,nonlinear,three-dimensional(3D),toroidal,electrostatic,gyrokinetic,particle-in-cell(PIC),Cartesian geometry UCANcode,with particle ions and adiabatic electrons,has been successfully exercised to identify non-diffusive transport characteristics in present day tokamak *** limitation in applying UCAN to larger scale discharges is the 1D domain decomposition in the toroidal(or z-)direction for massively parallel implementation using MPI which has restricted the calculations to a few hundred ion Larmor radii or gyroradii per plasma minor *** exceed these sizes,we have implemented 2D domain decomposition in UCANwith the addition of the y-direction to the processor *** has been facilitated by use of relevant components in the P2LIB library of field and particle management routines developed for UCLA’s UPIC Framework of conventional PIC *** gyroaveraging specific to gyrokinetic codes is simplified by the use of replicated arrays for efficient charge accumulation and force *** 2D domain-decomposed UCAN2 code reproduces the original 1D domain nonlinear results within *** of UCAN2 on the Cray XC30 Edison at NERSC demonstrate ideal scaling when problem size is increased along with processor number up to the largest power of 2 available,namely 131,072 *** particle weak scaling benchmarks also indicate that the 1 nanosecond per particle per time step and 1 TFlops barriers are easily broken by UCAN2 with 1 billion particles or more and 2000 or more processors.

Currently, and ignoring marine-sourced fatty acids/esters, there are 8 approved drugs(US FDA and Australia) that can be traced back to marine natural products initially isolated from a variety of sources. Six are directed towards cancers of man, one is for pain relief and one is an anti-HIV agent. These have come from the approximately 25,000 compounds currently reported from marine sources from approximately 1965. However,there are a substantial number of "toxic" agents that are either in clinical trials(as the initial compound or a derivative) or are potential "warheads" to be used with specific monoclonal antibodies. The 8 approved agents will be noted with brief comments, but the majority of the available time will be devoted to discussing agents from a variety of marine sources, that are either candidates for warheads, and/or have proceeded to PhaseⅡand Ⅲ trials in man against various cancers. The influence of older compounds that can be modified chemically to provide "interesting leads" will be one of the fundamental areas noted.
