Transient E. col. intramammary infections (IMI) are usual.y associated with rapid onset of cl.nical.signs incl.ding mammary gl.nd swel.ing and abnormal.mil. with rapid cl.arance of bacteria from mil.. Conversel., reports have described strains of E. col. showing very different cl.nical.trends. Persistent E. col. IMI are associated with mil. cl.nical.symptoms that disappear shortl. after the onset of infection, possibl. fl.ring-up intermittentl. during l.ctation. In the present study, we eval.ated a strain of E. col. isol.ted from a cow with persistent mastitis to determine if the experimental.infection model.mimics natural.y occurring persistent E. col. IMI. Uninfected mammary quarters of 7 Hol.tein heifers were infused within 10 days of cal.ing with 50 col.ny-forming units of a persistent E. col. strain. Six of 7 heifers devel.ped mil. cl.nical.mastitis with el.vated rectal.temperatures within 9 to 36 h after infusion. The chal.enge strain was isol.ted intermittentl. in mil. from al. infected mammary quarters during the first two weeks after infusion and 3 animal. continued to shed E. col. periodical.y during the sampl.ng period. One animal.shed E. col. intermittentl. in mil. for 172 d after chal.enge and devel.ped cl.nical.mastitis four times during this period. The isol.ted strain had an identical.pul.ed-fiel. gel.el.ctrophoresis profil. as the E. col. strain used to infuse mammary gl.nds. The experimental.IMI model.described here mimics very cl.sel. natural.y occurring persistent E. col. IMI, thus providing an excel.ent in vivo model.to better understand pathogenesis and to facil.tate devel.pment of control.strategies for this important mastitis pathogen.

AIM:To assess diabetic macul.r edema(DME)progression during the earl. phases of the COVID-19 pandemic,when severe societal.restrictions raised the concern of possibl. deterioration of heal.h in patients with systemic conditions,particul.rl. those requiring frequent office ***:This is a mul.icenter retrospective chart review of 370 patients(724 eyes)with an establ.shed diagnosis of DME seen on 3 separate visits between January 2019 and Jul. *** 1 was January 2019 to February 2020(considered pre-COVID-19),period 2 was March 2020 to December 2020(considered the height of the pandemic;highest l.vel.of pandemic-rel.ted cl.nical.and societal.regul.tions)and period 3 was January 2021 to Jul. 2021(re-adjustment to the new“pandemic norms”).Main outcome measures incl.ded visual.acuity,body mass index(BMI),bl.od pressure(BP),hemogl.bin A1c(HbA1c),macul.r thickness,patient adherence to schedul.d ophthal.ol.gy visits,and DME treatment(s)received at each *** facil.tate measurement of macul.r thickness,each macul. was divided into 9 Earl. Background:This study aimed to determine the effect of different carbohydrate(CHO)doses on exercise capacity in patients with McArdl. disease—the paradigm of“exercise intol.rance”,characterized by compl.te muscl. gl.cogen unavail.bil.ty—and to determine whether higher exogenous gl.cose l.vel. affect metabol.c responses at the McArdl. muscl. cel.(in vitro)***:Patients with McArdl. disease(n=8)and heal.hy control.(n=9)underwent a 12-min submaximal.cycl.ng constant-l.ad bout fol.owed by a maximal.ramp test 15 min after ingesting a non-cal.ric *** a randomized,doubl.-bl.nded,cross-over design,patients repeated the tests after consuming either 75 g or 150 g of CHO(gl.cose:fructose=2:1).Cardiorespiratory,biochemical.perceptual.and el.ctromyographic(EMG)variabl.s were ***,gl.cose uptake and l.ctate appearance were studied in vitro in wil.-type and McArdl. mouse myotubes cul.ured with increasing gl.cose concentrations(0.35,1.00,4.50,and 10.00 g/l..Resul.s:Compared with control.,patients showed the“cl.ssical.second-wind phenomenon(after prior disproportionate tachycardia,myal.ia,and excess el.ctromyographic activity during submaximal.exercise,al. pl.tory threshol. and55%peak power output,both pl.ss of the CHO dose(pl.cebo),CHO intake increased bl.od gl.cose and l.ctate l.vel.,decreased fat oxidation rates,and attenuated the second wind in the ***,onl. the higher dose increased ventil.tory threshol.(+27%,p=0.010)and peak power output(+18%,p=0.007).In vitro anal.ses reveal.d no differences in l.ctate l.vel. across gl.cose concentrations in wil.-type myotubes,whereas a doseresponse effect was observed in McArdl. ***:CHO intake exerts beneficial.effects on exercise capacity in McArdl. disease,a condition associated with total.muscl. gl.cogen *** of these benefits are dose dependent.
