Field Trials of CpGV Virus Isolates Overcoming Resistance to CpGV-M

The Cydia pomonella granulovirus (CpGV) has been used for many years as biological agent for codling moth control in apple orchards. Resistance to the Mexican strain of CpGV was detected in orchards in Germany, France and Italy. A laboratory insect colony was started from insects collected in a French resistant orchard. It was named RGV. Various virus isolates were identified as active against this resistant insect colony. Field tests were carried out in 2007 to test if the two virus isolates CpGV-I12 and NPP-R1 were effective in the field. Although these virus isolates were not able to reduce insect caused fruit damages, they significantly reduced the overwintering insect populations. NPP-R1 was subjected to eight passages on RGV larvae (NPP-R1.8) that improved its biological activity on RGV larvae. 2008 field trials were set up to test this improved virus strain, compared to CpGV-I12 and Madex plus active on RGV. These tests confirmed the ability to control both in susceptible and resistant insect populations.

Growth depression of Rosa plants at sites previously used to cultivate the same or closely related species is a typical symptom of rose replant disease(RRD).Currently,limited information is available on the causes and the etiology of RRD compared to apple replant disease(ARD).Thus,this study aimed at analyzing growth characteristics,root morphology,and root metabolites,as well as microbial communities in the rhizosphere of the susceptible rootstock Rosa corymbifera‘Laxa’grown in RRD-affected soil from two sites(Heidgraben and Sangerhausen),either untreated or disinfected byγ-*** a greenhouse bioassay,plants developed significantly more biomass in theγ-irradiated than in the untreated soils of both *** plant metabolites detected in ***‘Laxa’roots were site-and *** aloesin was recorded in significantly higher concentrations in untreated than inγ-irradiated soils from Heidgraben,the concentrations of phenylalanine were significantly lower in roots from untreated soil of both *** microbial communities of 8-week-old plants were studied by sequencing of 16S rRNA,ITS,and cox gene fragments amplified from total community *** by microscopic observations,sequences affiliated to the bacterial genus Streptomyces and the fungal genus Nectria were identified as potential causal agents of RRD in the soils *** relative abundance of oomycetes belonging to the genus Pythiogeton showed a negative correlation to the growth of the ***,the RRD symptoms,the effects of soil treatments on the composition of the rhizosphere microbial community revealed striking similarities to findings related to ARD.

Impacts of 1-Aminocyclopropane-1-Carboxylic Acid as a Late Post-Bloom Thinner on Fruit Set, Yield, and Fruit Quality in “Gala” and “Fuji” apples

A few post-bloom thinners are available for apple (Malus domestica Borkh.) and the prospects for additional thinners appear to be limited. Application of blossom thinners may present a risk of overthining in the areas where weather is less predictable. Thus, we studied the impacts of various rates of 1-aminocyclopropane-1-carboxylic acid (ACC) in three strains of “Fuji” apple and different rates of ACC and one rate of Ethrel in “Buckeye Gala” apple, when fruitlet diameter was about 20 mm, on fruit set, yield and fruit quality attributes at harvest, and return bloom in southwest Idaho in the Intermountain West region, USA. In 2013, application of Ethrel at 300 mg·L-1 did not affect fruit set, fruit weight, diameter (D), length(L), or L/D ratio 34 days after application, or yield, fruit weight, color, russet, and starch degradation pattern (SDP) at harvest, while application of ACC at 150 mg·L-1 or higher reduced fruit set by 19% to 34% in “Buckeye Gala” apple. In this cultivar, application of ACC at 350 mg·L-1 significantly increased fruit weight, diameter and length 34 days after application, and increased fruit weight, color, and SDP at harvest time. Application of ACC at all rates reduced total yield per tree in “Buckeye Gala”. Application of ACC at 300 mg·L-1 significantly reduced fruit set but applications of 150 mg·L-1 or 300 mg·L-1 ACC did not affect yield or quality attributes of “Sun Fuji” apple in Sunny Slope area in 2013. Application of ACC reduced fruit set and slightly increased fruit size in “Top Export Fuji” in 2013. Application of ACC at 600 mg·L-1 significantly reduced fruit set in “Aztec Fuji” apple in 2014. Application of ACC in a season never reduced bloom density (return bloom) of the next season. Overall, we conclude that ACC is an excellent tool as a late-season post-bloom fruit thinner and can be effective when applied

Informing Primary School Nutritional Policy: Effects of Mid-Morning Snacks on Appetite and Energy Control

The purpose of this research was to inform primary school nutritional policy by identifying which mid-morning snack would be more beneficial to consume from an appetite control perspective. During morning break 14 girls and 11 boys were provided with 160 ml of semi-skimmed milk or153 gof apple in a randomised crossover manner. Visual analogue scales were used to record hunger, prospective food consumption and fullness, immediately before and after breakfast, immediately before and after the mid-morning snack, and every 60 min until 21:00 on each day. School dinner/packed lunch energy intakes were assessed 90 min following the mid-morning snacks, in addition to evening energy intake. Children felt less hungry and could eat less when apple was consumed, however lunch and evening energy intakes were not different. Fluctuations in appetite did not translate into differences in energy intake therefore both milk and fruit should be promoted as mid-morning snacks in primary schools.

Genomewide selection for fruit quality traits in apple: breeding insights gained from prediction and postdiction

Excised Leaf Bioassay: Phenotypic Expression of Natural Resistance in apple against the Obliquebanded Leafroller

Different approaches were assessed in the development of a bioassay method that would provide data necessary to evaluate resistance of apple genotypes to the obliquebanded leafroller (OBLR) Choristoneura rosaceana (Harris). A leaf disc method used to evaluate the impact of pesticides on leafrollers resulted in high levels of leaf-abandonment of first and second instar larvae. Inserting leaf petioles into water soaked Oasis foam resulted in a significant reduction in overall leaf weight and diminished the appearance of leaf quality. When leaf petioles were inserted into water-filled Eppendorf vials, leaf weight increased and overall leaf quality appeared to be good. We opted to adopt this latter whole-leaf bioassay method, which though very time consuming, provided a robust assessment of OBLR life history parameters against different apple genotypes. Across all apple genotypes evaluated, male larvae developed faster than females to pupation as well as to adulthood, while females developed faster as pupae and were heavier than male pupae. OBLR larvae reared on leaves from different apple genotypes displayed significant variability in development time, pupal weight, larval survivorship, and number of offspring produced. We utilized three indices to characterize host acceptance, sub-lethal effects, and lethal effects of OBLR to apple genotypes. Our results indicated that ‘Lady’, ‘Viking’, and ‘Northern Spy’ show some of the highest levels of resistance against OBLR, while ‘Granny Smith’ was quite susceptible to OBLR and could be used in future studies identifying apple genotypes with relative resistance to OBLR.

A 48-year-old woman presented with extensive facial ulceration of 1 year in duration. Based on the combination of the clinical appearance and a “nondiagnostic”biopsy taken elsewhere, the patient was started on oral prednisone at a dose of 40 mg/day, with a working diagnosis of pyoderma gangrenosum. According to the patient, the ulceration worsened over the 2 months whilst on prednisone and pain control was a major issue, controlled for the most part with oral oxycodone. One month prior to our evaluation of the patient, she was started on dapsone at 50 mg/day with no added benefit. Approximately 2 years prior to our evaluation, she developed raised, reddish, skin lesions on the abdomen and legs which recurred, but healed spontaneously each time after a few weeks. Her past medical history was remarkable for the remote use of intravenous drugs (which she had stopped for the past 20 years) and the past and continued use of alcohol on a daily basis. She had recently been tested elsewhere and was found to be positive for hepatitis C, but not human immunodeficiency virus (HIV). Examination revealed several ulcerations of the face and forehead, with an “apple jelly”coloration to the periphery and necrotic center. There was complete erosion of the nasal sidewall with apparent involvement of the septum(Fig.1). There were also scattered, smaller, nonulcerated, reddish to purplish lesions on the abdomen. Otolaryngologic examination revealed ulceration of the right ala and erosion of the nasal septum, but was otherwise unremarkable. No cervical or submental lymphadenopathy was noted. Routine laboratory tests showed the patient to be anemic, with a hemoglobin level of 11 g/dL (normal, 12-15.5 g/dL) and a white blood count of 13,300 (normal,3500-10,500) The total bilirubin was normal at 1.0 mg/dL, but several liver function tests were elevated by 2-4 times the upper limit of normal, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), and γ-glutamyl transpep
