BACKGROUND Peripheral blood stem cells(PBSC)are commonly cryopreserved awaiting clinical use for hematopoietic stem cell *** term cryopreservation is commonly defined as five years or longer,and limited data exists regarding how long PBSC can be cryopreserved and retain the ability to successfully *** programs,stem cell banks,and regulatory and accrediting agencies interested in product stability would benefit from such ***,we assessed recovery and colony forming ability of PBSC following long-term cryopreservation as well as their ability to engraft in NOD/SCID/IL-2 Rγnull(NSG)*** To investigate the in vivo engraftment potential of long-term cryopreserved PBSC *** PBSC units which were collected and frozen using validated clinical protocols were obtained for research use from the Cellular Therapy Laboratory at Indiana University *** units were thawed in the Cellular Therapy Laboratory using clinical standards of practice,and the pre-freeze and post-thaw characteristics of the units were *** function was assessed using standard colony-forming ***34-selected cells were transplanted into immunodeficient mice to assess stem cell *** Ten PBSC units with mean of 17 years in cryopreservation(range 13.6-18.3 years)demonstrated a mean total cell recovery of 88%±12%(range 68%-110%)and post-thaw viability of 69%±17%(range 34%-86%).BFU-e growth was shown in 9 of 10 units and CFU-GM growth in 7 of 10 units *** mice were transplanted with CD34-selected cells from four randomly chosen PBSC *** mice demonstrated long-term engraftment at 12 wk with mean34%±24%human CD45+cells,and differentiation with presence of human CD19+,CD3+and CD33+*** bone marrow from all mice demonstrated growth of erythroid and myeloid *** We demonstrated engraftment of clinically-collected and thawed PBSC following cryopreservation up to 18 years in NSG mice,sign

The constitutively-expressed cyclooxygenase 1（COX-1） and the inducible COX-2 are both involved in the conversion of arachidonic acid（AA） to prostaglandins（PGs）.However,the functional roles of COX-1 at the cellular level remain *** hypothesized that by comparing differential gene expression and eicosanoid metabolism in lung fibroblasts from wild-type（WT） mice and COX-2^（-/-） or COX-1^（-/-） mice may help address the functional roles of COX-1 in inflammation and other cellular *** to WT,the number of specifically-induced transcripts were altered descendingly as follows：COX-2^（-/-）〉 COX-1^（-/-）〉 WT ＋ IL-1β.COX-1^（-/-） or COX-2^（-/-） cells shared about 50%of the induced transcripts with WT cells treated with IL-1β,*** interactive ＂anti-inflammatory,proinflammatory,and redox-activated＂ signature in the protein-protein interactome map was observed in COX-2^（-/-） *** augmented COX-1mRNA（in COX-2^（-/-） cells） was associated with the upregulation of mRNAs for glutathione S-transferase（GST）,superoxide dismutase（SOD）,NAD（P）H dehydrogenase quinone 1（NQO1）,aryl hydrocarbon receptor（AhR）,peroxiredoxin,phospholipase,prostacyclin synthase,and prostaglandin e synthase,resulting in a significant increase in the levels of PGe2,PGD2,leukotriene B4（LTB4）,PGF（1α）,thromboxane B2（TXB2）,and PGF（2α）.The COX-1 plays a dominant role in shifting AA toward the LTB4 pathway and anti-inflammatory *** to WT,the upregulated COX-1 mRNA in COX-2^（-/-） cells generated an ＂eicosanoid storm＂.The genomic characteristics of COX-2^（-/-） is similar to that of proinflammatory cells as observed in IL-1β induced WT ***-1^（-/-） and COX-2^（-/-） cells exhibited compensation of various eicosanoids at the genomic and metabolic levels.

土体所经历的干湿循环和应力历史,对其孔隙结构和持水性能影响明显。为进一步深入研究二者耦合作用的影响,开展了先竖向固结再循环脱吸湿以及先气压脱湿再反复竖向加卸载两种不同加载路径的水分变化测量试验,探索土样持水性能和变形能力的演化规律。研究结果表明:(1)脱湿与吸湿路径对比,土体的持水能力具有明显的差异性,且土体越密实,这种差异性越强烈;(2)随着脱吸湿循环次数的增加,土样的进气值略有增大,减湿段和吸湿段的斜率都会减小,但影响趋势随循环次数增多而减弱;(3)对于气压减湿后的非饱和土样,其前期固结压力随气压值增加而增大,而加卸载形成的滞回圈的面积却随之减小,土体的持水性能和变形能力也降低;(4)重塑土样从不同加载路径首次达到同一应力状态时,先固结再脱湿路径下土样的体积收缩更多,而先施加气压再加载路径下土样持水性能减弱的更多。

Over the last two decades, the hypothesis of a magnocellular deficit in dyslexia has raised considerable interest and controversy. Using an electrophysiological procedure (visual evoked potentials, VeP), we compared magnocellular and parvocellular contrast and spatial frequency-response functions between phonological dyslexics (n = 16) and a typical reading group (n = 12) matched for age and socioeconomic background. No significant differences were found between the two groups in the amplitude of the VeP components associated with either magnocellular or parvocellular responses. However, topographic analyses revealed a group difference in the distribution of amplitude in the right frontal and left temporal regions, which appeared to be underactivated in dyslexics. These results suggest a deficit in the higher-level cortical regions involved in phonological and/or linguistic processing, and calls into question the notion of a magnocellular involvement in dyslexia.

Isolation and activation of collagenase from fish processing waste

Collagenase was isolated from fish waste (a mixture of haddock, herring, ground fish and flounder) using a Tris-buffer system. The proteins in the crude extract were precipitated using ammonium sulfate (40% - 80%) and purified with gel-filtration chromatography using Sephadex G-100. The results showed that the collagenase enzyme was produced as a latent enzyme and was activated with bovine trypsin and potassium thiocyanate (KSCN). The enzyme activity was affected by pH and temperature. Optimal enzyme activities were found at 35?C and a pH of 7.5 when insoluble collagene type I was used as substrate and the liberated amino acids were measured in relation to L-leucine in the presence of ninhydrin. The enzyme activity was completely inhibited by the action of ethylenediaminetetraacetic acid (eDTA) suggesting that the collagenase enzyme isolated from the fish waste is a metalloproteinase enzyme requiring metal ions for enzyme activity. Dialysis against KSCN decreased the enzyme total activity and increased its specific activity. Sodium dodecyl sulphate polyacryla-mide gel electrophoresis (SDS-PAGe) indicated that the purified procollagenase enzyme have only one band at molecular weight of 50 kilodaltons (kDa). When the enzyme was cleaved with trypsin, it was found to consist of two subunits: a large unit with a molecular weight of 50 kDa and a small unit with a molecular weight of 10 kDa.

Studies on interacting Blends of Acrylated epoxy resin based Poly(ester-Amide)s and Vinyl esterResin

epoxy resin based Unsaturated poly(ester-amide) resins (UPeAs) can be prepared by many methods but here these were prepared by reported method [1]. These UPeAs were then treated with acrylotl chloride to afford acrylated UPeAs resin (i.e. AUPeAs). Interacting blends of equal proportional AUPeAs and vinyl ester epoxy (Ve) resin were prepared. APeAs and AUPeAs were characterized by elemental analysis, molecular weight determined by vapour pressure osmometer and by IR spectral study and by thermogravimetry. The curing of interacting blends was monitored on differential scanning calorimeter (DSC). Based on DSC data in situ glass reinforced composites of the resultant blends have been prepared and characterized for mechanical, electrical and chemical properties. Unreinforced blends were characterized by thermogravimetry (TGA).

Temporal and elevation trends in rainfall erosivity on a 149 km2watershed in a semi-arid region of the American Southwest
