Evaluation of the Kinetic change of the Immunogenicity of Dengue-2 DNA Vaccine in Mice Administered by Different Administration Routes

A plasmid DNA vaccine is able to induce both humoral and cellular immune responses;however, the kinetic change of the Th1/Th2 response, antibody avidity, cytokine secretion, and neutralization activity after different priming and boosting strategies have not been evaluated. A plasmid DNA, designated pCBD2 and previously shown to efficiently induce an immune response very similar to that by a wild type virus, was evaluated kinetically in this study. Our results suggest that a DNA vaccine delivered by the gene gun (gg) route produced higher and longer DENV-2-specific antibody titers than those induced through the intramuscular (im) route. Although the gg group induced a Th2 response and im delivery induced a Th1 response, priming by gg delivery, followed by a boosting by im delivery, did not shift the immune response from a Th2 to Th1 response. Furthermore, the antibody avidity (AI) measured by ELISA demonstrated a gradual increase of AI from low (AI range from 6.8% - 9.6%) on day 42 to high (AI value > 30) on day 119 in all but the gene-gun immunization group, in which an AI value of 23 was observed. Although there was lower avidity in the gg group, the mice sera from all three groups of mice demonstrated significant neutralization activity. This is the first report about the kinetics of immunogenicity of a DNA vaccine through different administration strategies, which suggests that gene gun delivery of a DNA vaccine can induce an immune response containing both neutralizing and nonneutralizing antibodies at high titers important for neutralization.

BACKGROUND A new nomenclature of metabolic associated steatotic liver disease(MASLD)was proposed in 2023,thus expanding the diagnostic name of“MASLD combined with other etiologies”.AIM To investigate the clinical profiles of patients with concurrent MASLD and ch-ronic hepatitis B virus(HBV)*** This study included participants from the Taiwan *** diagnostic cri-teria of MASLD encompassed hepatic steatosis and any cardio-metabolic risk *** hepatitis B surface antigen was considered indicative of chronic HBV *** etiology was defined as MASLD combined with chronic HBV infection(MASLD-HBV).Fibrosis 4(FIB-4)score determined the severity of liver fibrosis,and athero-sclerosis was diagnosed by the presence of carotid plaques on duplex *** In a total of 18980 participants(mean age,55.18±10.35 years;males,30.42%),there were 7654(40.3%)MASLD patients and 2128(11.2%)HBV *** propensity score matching for age and gender,HBV carriers had a lower percentage of MASLD than healthy *** with dual etiology had higher aspartate aminotrans-ferase,alanine aminotransferase(ALT),and FIB-4 levels,but lower gamma glutamyl transferase(GGT)levels than MASLD *** contrast,those with dual etiology had higher ALT and GGT levels,but lower FIB-4 than“HBV alone”*** risk of atherosclerosis was similar among these three *** MASLD-HBV patients have worse liver fibrosis severity than MASLD patients,but better liver fibrosis stage than“HBV alone”patients,suggesting a complex interaction between MASLD and chronic HBV infection.

The build-up of amyloid-β（Aβ） peptides in the brain has been linked to Alzheimer's disease pathogenesis and may represent a key target for *** radix（the root of Astragalus mongholicus Bunge or *** Bunge） is used mainly as a tonic agent in traditional Chinese ***（AST）,the major component of Astragali radix,can reduce the infarction volume after *** attempted to ascertain whether AST acts through the MAPK and PI3 K/AKT pathway to protect against b-amyloid pathways in cortical *** cortical neurons were cultured and exposed to 2 mM Ab in the absence or presence of AST（50-400 mg/ml）.In cultured cortical neurons,hyperphosphorylation of Tau,mitochondrial membrane potential,synaptotoxicity,apoptosis and activation of caspase-3 determined after Ab *** Ab-treated rats displayed severe memory impairments in passive avoidance test and both markedly tau hyperphosphorylation and increased numbers of degenerative neurons in the cerebral *** these effects were prevented or substantially attenuated by AST *** of cultured cortical neurons with AST attenuated the Ab-induced cytotoxicity,apoptosis,synaptotoxicity,and mitochondrial *** beneficial effect of AST in reducing Ab-induced cytotoxicity,apoptosis,and mitochondrial dysfunction in cortical cells can be blocked by inhibition of phosphoinositide 3-kinase（PI3 K）-dependent protein kinase B（PKB,as known as AKT） activation by *** addition,inhibition of extracellular protein kinase（ERK） with U0126 shares with the AST the same beneficial effects in reducing Ab-induced *** prevents Ab-induced Alzheimer's disease conditions by a mechanism involving the control of both the MAPK（or ERK）and the PI3 K/AKT pathways in injured cortical neurons.

The landscape of gastrointestinal endoscopy continues to evolve as new technologies and techniques become *** advent of image-enhanced and magnifying endoscopies has highlighted the step toward perfecting endoscopic screening and diagnosis of gastric ***,with the development of convolutional neural network,artificial intelligence(AI)has made unprecedented breakthroughs in medical imaging,including the ongoing trials of computer-aided detection of colorectal polyps and gastrointestinal *** the past demi-decade,applications of AI systems in gastric cancer have also *** AI’s efficient computational power and learning capacities,endoscopists can improve their diagnostic accuracies and avoid the missing or mischaracterization of gastric neoplastic *** far,several AI systems that incorporated both traditional and novel endoscopy technologies have been developed for various purposes,with most systems achieving an accuracy of more than 80%.However,their feasibility,effectiveness,and safety in clinical practice remain to be seen as there have been no clinical trials ***,AI-assisted endoscopies shed light on more accurate and sensitive ways for early detection,treatment guidance and prognosis prediction of gastric *** review summarizes the current status of various AI applications in gastric cancer and pinpoints directions for future research and clinical practice implementation from a clinical perspective.

Modified preoperative score to predict disease-free survival for hepatocellular carcinoma patients with surgical resections

BACKGROUND No prognostic models specific to hepatocellular carcinoma patients receiving surgical resection have been considered strong and convincing enough for survival prediction thus far,and there are no models including only preoperative *** derived a nomogram to predict disease-free survival in a previous *** To simplify our score and compare research outcomes among other scoring *** We retrospectively reviewed data from 1106 patients with hepatocellular carcinoma who underwent liver resection at the Linkou chang Gung Memorial Hospital between April 2003 and December *** analyses were conducted to identify the significant survival ***,Harrell’s C-index,and Akaike information criterion were compared between our score,AJCC 8^(th)edition,Tokyo score,and Taipei Integrated Scoring System(TTV-CTPAFP model).RESULTS Among the 1106 patients,731(66.1%)had tumor recurrence at a median followup of 83.9 *** risk factors were identified:platelet count,albumin level,indocyanine green retention rate,multiplicity,and radiologic total tumor *** were divided into three risk groups,and the 5-year survival rates were 61.7%,39%,and 25.7%,*** C-index was 0.617,which was higher than the Tokyo score(0.613)and the Taipei Integrated Scoring System(0.562)and equal to the value of the AJCC 8th edition(0.617).CONCLUSION The modified score provides an easier method to predict *** treatment can be planned preoperatively by dividing patients into risk groups.

Polychlorinated dibenzo-p-dioxins and dibenzofurans(PCDD/Fs)are a group of compounds of major environmental *** emitted into the atmosphere,PCDD/Fs undergo photochemical reactions and enter other environmental compartments via wet and dry *** this study,atmospheric PCDD/F depositions were collected via an automated PCDD/F deposition sampler and traditional cylindrical vessels,respectively,in northern,central and southern Taiwan from 2008 to *** automated PCDD/F precipitation sampler used in this study can prevent both re-suspension and photodegradation of the PCDD/Fs collected and also effectively separates the PCDD/F samples into dry and wet *** results indicate that the average atmospheric PCDD/F concentrations collected by the high-volume sampling trains were:13.6±10(n=10),15.6+5.2(n=7)and 10.9+6.3(n=6)fg I-TEQ/m in northern,central and southern Taiwan,*** addition,the results also indicate that the PCDD/F deposition flux collected with an automated PCDD/F sampler(1.84±0.90.68±5.1 pg I-TEQ/m/day,n=23)is significantly higher than that sampled with cylindrical vessels(1.11±0.69.64±5.2 pg I-TEQ/m/day,n=23).Based on the Mann-Whitney statistical analysis,the p value(0.037)of PCDD/F deposition flux between those two samplers measurement is lower than *** difference is attributed to the fact that part of the PCDD/F depositions collected by traditional cylindrical vessels is photo degraded and re-volatilization.
