Taq1B cETP Polymorphism and cardiovascular Risk in an Endogamous Pop-ulation of Diabetic Men: A Study in Santa Rosa Del conlara, San Luis, Argentina

Objectives: Type 2 diabetes mellitus (T2DM) patients are at increased risk of cardiovascular diseases (cVDs). Several polymorphisms in the cholesterol ester transfer protein (cETP) gene have been reported. The aim of this study was to determine the distribution and effect of the Taq1B polymorphism in the cETP gene on clinical and biochemical indicators of cVD risk in a population of endogamous-T2DM men. Methods: 102 men (57.5 ± 9.3 years old) inhabitants of Santa Rosa del conlara, San Luis, Argentina, were recruited and assigned into two groups (22 control and 80 T2DM). Further, these two groups were subdivided according to their Taq1B cETP gene genotypes (i.e., B1B1, B1B2 and B2B2). clinical and fasting-plasma biochemical indicators of cVD risk were measured and their association with the B1 allele was determined. Results: compared to control, T2DM men had more central obesity, hypertension, atherogenic index, insulin resistance and poorly controlled diabetes. compared to T2DM men having the B2 allele, those T2DM men having the B1 allele have increased risk of cVD as assessed by systolic blood pressure (156 ± 16.0 vs 135.8 ± 19.2, p = 0.015), atherogenic index (6.15 ± 1.3 vs 4.4 ± 0.7, p = 0.0008), HDL-c levels (38.9 ± 5.3 vs 64.4 ± 8.2, p ± 3.0 vs 2.4 ± 0.78, p = 0.004). Interestingly, only body mass index (r = ﹣0.559, p = 0.01) and HDL-c concentration (r = ﹣0.492, p = 0.02) negatively correlated with cVD risk in the endogamous population of B1B1 and B1B2 T2DM men. conclusion: The B1 allele of the cETP gene predicts cardiovascular complications in an endogamous population of T2DM men.

Seafloor and buried reliefs occur along continental margin of the Ross Sea(Antarctica).These features are several kilometres wide and tens of metres high,exhibiting cone or flat-top dome *** studies have proposed a volcanic or glacial origin for these formations,but these hypotheses do not account for all the available *** this study,we use morpho-bathymetric data,intermediate resolution multichannel seismic and high resolution chirp profiles,as well as magnetic lines to investigate these clusters of *** employing targeted processing techniques to enhance the geophysical characterization of the seafloor and buried reliefs,and to understand the underlying geological features,we propose that the reliefs are mud *** of these formations appear to be associated with a plumbing system,as indicated by acoustic anomalies linked to sediment containing *** formations are likely fed by clayey source rocks of Miocene ***,other reliefs might be the result of mud mobilisation caused by gravity instability and fluid overpressure.

Reduced nitrogen availability during growth improves quality in red oak lettuce leaves by minimizing nitrate content, and increasing antioxidant capacity and leaf mineral content

Overuse of N in lettuce production can lead to environmental problems caused by leaching and the accumulation of harmful nitrates in edible tissues. This study investigated the effect of applied nitrogen (N) concentrations between 40 and 2400 mg·L–1 on growth, nitrate accumulation, mineral leaf content, and antioxidant capacity in Oak Leaf lettuce cv. “Shiraz” grown under hydroponic conditions in Australia. Yield (g FW) increased with nitrogen (N) application rate up to 1200 mg·L–1, as did leaf N content, while c:N declined. Nitrogen Utilization Efficiency (NUtE) increased rapidly from 40 to 75 mg·L–1 applied N, leveling at 150 mg·L–1 with no subsequent effect of N concentrations between 400 and 2400 mg·L–1. Nitrate content rose significantly with increased N, particularly at 1200 and 2400 mg·L–1. Leaf total plant phenolic content (TPP) and antioxidant capacity (measured by ferric reducing antioxidant power—FRAP) were both maximal at 75 and 400 mg·L–1 applied N, while highest oxygen radical absorption capacity (ORAc) values were found in leaves supplied with low N (40 to 400 mg·L–1). Applied N as calcium nitrate also significantly affected leaf mineral content as B, Mg, Mn, and Zn significantly decreased with increasing N. These results indicate that N applications of 1200 mg·L–1 or higher can result in reduced antioxidant capacity and mineral content in lettuce leaves.

chloroplast Proteins without cleavable Transit Peptides： Rare Exceptions or a Major constituent of the chloroplast Proteome？

Most chloroplast proteins （cp proteins） are nucleus-encoded, synthesized on cytosolic ribosomes as precursor proteins containing a presequence （cTP）, and post-translationally imported via the Tic/Toc complex into the organelle, where the cTP is removed. Only a few unambiguous instances of cp proteins that do not require cTPs （non-canonical cp proteins） have been reported so far. However, the survey of data from large-scale proteomic studies presented here suggests that the fraction of such proteins in the total cp proteome might be as large as -30%. To explore this discrepancy, we chose a representative set of 28 putative non-canonical cp proteins, and used in vitro import and Red Fluorescent Protein （RFP）-fusion assays to determine their sub-cellular destinations. Four proteins, including embryo defective 1211, glycolate oxidase 2, protein disulfide isomerase-like protein （PDII）, and a putative glutathione S-transferase, could be unambiguously assigned to the chloroplast. Several others （＇potential cp proteins＇） were found to be imported into chloroplasts in vitro, but failed to localize to the organelle when RFP was fused to their c-terminal ends. Extrapolations suggest that the fraction of cp proteins that enter the inner compartments of the organelle, although they lack a cTP, might be as large as 11.4% of the total cp proteome. Our data also support the idea that cytosolic proteins that associate with the cp outer membrane might account for false positive cp proteins obtained in earlier studies.

利用静电纺丝技术制备了热塑性聚氨酯/多壁碳纳米管(TPU/MWNTs)复合纳米纤维,并通过平行板接收装置实现纤维的定向排布。测试了纤维压阻敏感性随MWNTs含量的变化规律,与浇注法制备的TPU/MWNTs膜的压阻敏感性进行对比分析,结果表明MWNTs含量仅为2.91%(质量分数)时,纤维就表现出了最佳的压阻特性(Gauge factor≈11.4),比同质量分数下膜的压阻特性(Gauge factor≈2.6)有较大的提高,同时在8%的拉伸应变内电阻的相对变化量与应变值维持良好的线性关系,线性度优于复合膜。

Milk: An Alternative Beverage for Hydration?

The hydration status of a physically active individual or athlete can influence his/her physical or mental performance. The degree of hypohydration may lead to serious health problems, and consequently to impair athletic performance. The consumption of water or carbohydrate-electrolyte beverages before, during and after exercise is essential to maintain fluid balance and prevent dehydration. Besides these drinks, research findings have pointed using bovine milk as an alternative to sports drinks to optimize hydration levels in athletes and physically active individuals. The nutritional composition of cow’s milk promotes post-exercise rehydration process, due to the natural presence of water and electrolytes such as sodium and potassium. Although most studies have confirmed that bovine milk is a viable alternative for rehydration, recent studies have not reported significant differences between milk and some sports drinks for hydration process. Thus, milk consumption could be used as an alternative beverage for the hydration process.

Effect of discounting on estimation of benefits determined by hepatitis c treatment

The combination of either boceprevir or telaprevir with ribavirin and interferon (triple therapy) has been shown to be more effective than ribavirin+interferon (dual therapy) for the treatment of genotype 1 hepatitis c. Since the benefit of these treatments takes place after years, simulation models are needed to predict long-term outcomes. In simulation models, the choice of different values of yearly discount rates (e.g., 6%, 3.5%, 2%, 1.5% or 0%) influences the results, but no studies have specifically addressed this issue. We examined this point by determining the long-term benefits under different conditions on the basis of standard modelling and using quality-adjusted life years (QALYs) to quantify the benefits. In our base case scenario, we compared the long-term benefit between patients given a treatment with a 40% sustained virologic response (SVR) (dual therapy) and patients given a treatment with a 70% SVR (triple therapy), and we then examined how these specific yearly discount rates influenced the incremental benefit. The gain between a 70% SVR and a 40% SVR decreased from 0.45 QA-LYs with a 0% discount rate to 0.22 QALYs with a 6% discount rate (ratio between the two values = 2.04).Testing the other discounting assumptions confirmed that the discount rate has a marked impact on the magnitude of the model-estimated incremental benefit. In conclusion, the results of our analysis can be helpful to better interpret cost-effectiveness studies evaluating new treatment for hepatitis c.

Systemic mastocytosis (SM) is a heterogeneous disease of the bone marrow characterized by abnormal growth, accumulation and activation of clonal mast cells (Mcs). We report a case of SM with multiorgan involvement. A 30-year-old man presented with diarrhea, flushing, maculopapular rash with itching and weight loss. The upper and lower gastrointestinal endoscopies showed macroscopic involvement of stomach and duodenum; mucosal samples from stomach, duodenum, colon and distal ileum showed mucosal inf iltration by large, spindleshaped Mcs with abnormal surface molecule expression (cD2 and cD25), a picture fully consistent with SM, according to the World Health Organization diagnostic criteria. A computed tomography scan showed diffuse lymphadenopathy, hepatosplenomegaly and diffuse small bowel involvement. Bone marrow aspirate and biopsy were diagnostic for SM; serum tryptase levels were increased (209 ng/mL, normal values conclusive diagnosis was smouldering SM. There wereno therapeutic indications except for treatment of symptoms. The patient was strictly followed up because of the risk of aggressive evolution.

Resective surgery for liver tumor: a multivariate analysis of causes and risk factors linked to postoperative complications

BAcKGROUND: In spite of accurate selection of patients eligible for resection, and although advances in surgical techniques and perioperative management have greatly contributed to reducing the rate of perioperative deaths, stress must be placed on reducing the postoperative complication rates reported to be still as high as 50%. This study was designed to analyze the causes and foreseeable risk factors linked to postoperative morbidity on the grounds of data derived from a single-center surgical population. METHODS: From September 1989 to March 2005, 287 consecutive patients, affected either with Hcc or liver metastasis, had liver resection at our department. Among the Hcc series we recorded 98 patients (73.2%) in child- Pugh class A, 32 (23.8%) in class B and 4 in class c (3%). In 104 colorectal metastases, 71% were due to colon cancer, 25% rectal, 3% sigmoid, and 1% anorectal. In 49 non-colorectal metastases, 22.4% were derived from breast cancer, 63.2% gastrointestinal tumors (excluding colon) and 14.4% other cancers. We performed 80 wedge resections, 77 bisegmentectomies and/or left lobectomies, 74 segmentectomies, 22 major hepatectomies, 20 left hepatectomies, and 14 trisegmentectomies. RESULTS: The in-hospital mortality rate in this series was 4.5%, and the morbidity rate was 47.7%, because of pleural effusion (30%), hepatic abscess (25%), hepatic insufficiency (19%), ascites (10%), hemoperitoneum (10%), or biliary fistula (6%). The variables associated with the technical aspects of the surgical procedure thatwere responsible for the complications were: a Pringle maneuver length more than 20 minutes (P=0.001); the type of liver resection procedure, including major hepatectomy (P=0.02), left hepatectomy (P=0.04), trisegmentectomy (P=0.04), bisegmentectomy and/or left lobectomy (P=0,04); and a blood transfusion of more than 600 ml (P=0.04). cONcLUSION: The evaluation of causes and foreseeable risk factors linked to postoperative morbidity during the planning of

Identification of Target Genes and Transcription Factors Implicated in Translation-Dependent Retrograde Sig- naling in Arabidopsis

changes in organellar gene expression （OGE） trigger retrograde signaling. The molecular dissection of OGE-dependent retrograde signaling based on analyses of mutants with altered OGE is complicated by compensatory responses that mask the primary signaling defect and by secondary effects that influence other retrograde signaling pathways. Therefore, to identify the earliest effects of altered OGE on nuclear transcript accumulation, we have induced OGE defects in adult plants by ethanol-dependent repression of PRORS1, which encodes a prolyl-tRNA synthetase located in chloroplasts and mitochondria. After 32 h of PRORS1 repression, the translational capacity of chloroplasts was reduced, and this effect subsequently intensified, while basic photosynthetic parameters were still unchanged at 51 h. Analysis of changes in whole-genome transcriptomes during exposure to ethanol revealed that induced PRORS1 silencing affects the expression of 1020 genes in all. Some of these encode photosynthesis-related proteins, including several down-regulated light-harvesting chlorophyll a/b binding （LHc） proteins. Interestingly, genes for presumptive endoplasmic reticulum pro- teins are transiently up-regulated. Furthermore, several NAc-domain-containing proteins are among the transcription factors regulated. candidate cis-acting elements which may coordinate the transcriptional co-regulation of genes sets include both G-box variants and sequence motifs with no similarity to known plant c/s-elements.
