The stabilization and m.nipulation of laser frequency by m.ans of an external cavity are nearly ubiquitously used in fundam.ntal research and laser applications. While m.st of the laser light transm.ts through the cavity, in the presence of som. back-scattered light from.the cavity to the laser, the self-injection locking effect can take place, which locks the laser em.ssion frequency to the cavity m.de of sim.lar frequency. The self-injection locking leads to dram.tic reduction of laser linewidth and noise. Using this approach, a com.on sem.conductor laser locked to an ultrahigh-Q m.croresonator can obtain sub-Hertz linewidth, on par with state-of-the-art fiber lasers. Therefore it paves the way to m.nufacture high-perform.nce sem.conductor lasers with reduced footprint and cost. m.reover, with high laser power, the optical nonlinearity of the m.croresonator drastically changes the laser dynam.cs, offering routes for sim.ltaneous pulse and frequency com. generation in the sam. m.croresonator. Particularly, integrated photonics technology, enabling com.onents fabricated via sem.conductor Cm.S process, has brought increasing and extending interest to laser m.nufacturing using this m.thod. In this article, we present a com.rehensive tutorial on analytical and num.rical m.thods of laser self-injection locking, as well a review of m.st recent theoretical and experim.ntal achievem.nts.

Im.unobiology of congenital cytom.galovirus infection of the central nervous system.-the m.rine cytom.galovirus m.del

Congenital hum.n cytom.galovirus infection is a leading infectious cause of long-term.neurodevelopm.ntal sequelae, including m.ntal retardation and hearing defects. Strict species specificity of cytom.galoviruses has restricted the scope of studies of cytom.galovirus infection in anim.l m.dels. To investigate the pathogenesis of congenital hum.n cytom.galovirus infection, we developed a m.use cytom.galovirus m.del that recapitulates the m.jor characteristics of central nervous system.infection in hum.n infants, including the route of neuroinvasion and neuropathological findings. Following intraperitoneal inoculation of newborn anim.ls with m.use cytom.galovirus, the virus dissem.nates to the central nervous system.during high-level virem.a and replicates in the brain parenchym., resulting in a focal but widespread, non-necrotizing encephalitis. Central nervous system.infection is coupled with the recruitm.nt of resident and peripheral im.une cells as well as the expression of a large num.er of pro-inflam.atory cytokines. Although infiltration of cellular constituents of the innate im.une response characterizes the early im.une response in the central nervous system. resolution of productive infection requires virus-specific CD8＋ T cells. Perinatal m.use cytom.galovirus infection results in profoundly altered postnatal developm.nt of the m.use central nervous system.and long-term.m.tor and sensory disabilities. Based on an enhanced understanding of the pathogenesis of this infection, prospects for novel intervention strategies aim.d to im.rove the outcom. of congenital hum.n cytom.galovirus infection are proposed.

The Discovery and Developm.nt of Boceprevir:A Novel, First-generation Inhibitor of the Hepatitis C Virus NS3/4A Serine Protease

An estim.ted 2–3%of the world's population is infected with hepatitis C virus (HCV), m.king it a m.jor global health problem. Consequently, over the past 15 years, there has been a concerted effort to understand the pathophysiology of HCV infection and the m.lecular virology of replication, and to utilize this knowledge for the developm.nt of m.re effective treatm.nts. The virally encoded non-structural serine pro-tease (NS3) is required to process the HCV polyprotein and release the individual proteins that form.the viral RNA replication m.chinery. Given its critical role in the replication of HCV, the NS3 protease has been recognized as a potential drug target for the developm.nt of selective HCV therapies. In this review, we describe the key scientific discoveries that led to the approval of boceprevir, a first-generation, selective, sm.ll m.lecule inhibitor of the NS3 protease. We highlight the early studies that reported the crystal structure of the NS3 protease, its role in the processing of the HCV polyprotein, and the structural requirem.nts critical for substrate clea-vage. We also consider the novel attributes of the NS3 protease-binding pocket that challenged developm.nt of sm.ll m.lecule inhibitors, and the studies that ultim.tely yielded m.lligram.quantities of this enzym. in a soluble, tractable form.suitable for inhibitor screening program.. Finally, we describe the discovery of boceprevir, from.the early chem.stry studies, through the developm.nt of high-throughput assays, to the phase Ⅲ clinical developm.nt program.that ultim.tely provided the basis for approval of this drug. This latest phase in the developm.nt of boceprevir represents the culm.nation of a m.jor global effort to under-stand the pathophysiology of HCV and develop sm.ll m.le-cule inhibitors for the NS3 protease.

We present a laser resection technique to be used when biotissue is heated by laser radiation to the tem.erature of m.terial softening and then m.ved apart by the lateral edge of the optical fiber with a strongly absorbing tip. The absorbing tip allows the decrease in the power of effective cutting of the laser scalpel at the wavelength of 0.97 μm. And it changes the resection speed as well as the characteristics of the dam.ge of the tissues adjacent to the incision in experim.nts on m.del m.dia and experim.ntal anim.ls.

Prom.ting Health Behavior Change in Canada: A Quality Im.rovem.nt Project Integrating Prevention and Wellness in Frontline Care

Background: The critical need for Health Behaviour Change (HBC) for preventative care has been highlighted by the COVID-19 pandem.c. This quality im.rovem.nt project assessed the effectiveness and acceptability of Fountain of Health HBC tools as a m.nim.l intervention for prim.ry and secondary prevention am.ng Canadian clinicians and their patients. m.thods: Clinicians received HBC education and tools (paper tools and app-based) to assist their patients in setting a S.m.A.R.T (Specific, m.asurable, Action-oriented, Realistic, Tim.-lim.ted) goal at baseline and assessed four weeks later. Prim.ry outcom. m.asures were: 1) patient self-report of success at goal attainm.nt;2) patient self-reported evaluation of change in well-being and health attitudes;and 3) clinician experience and engagem.nt. Results: A total of 2184 clinicians received HBC education. Of these, 759 clinicians registered to participate in the project. 961 patients set S.m.A.R.T. HBC goals using either the paper tools (PT) or the app form.t. Patient data revealed nearly all patients (89% of PT users and 90% of app users) at least partially succeeded in attaining their goal at a four-week follow-up. m.st patients (85% of PT and 80% of app users) also reported im.rovem.nt in their well-being. A significant difference in health beliefs and attitudes was found in both PT (p Conclusions: HBC is challenging. Educating and providing clinicians with HBC tools as a m.nim.l intervention was found to be a successful health prevention strategy to im.rove patient well-being and health attitudes, and achieve S.m.A.R.T. goals. Results show HBC tools, education, and support for clinicians are both acceptable and effective for HBC in frontline care. These findings are relevant and tim.ly given greater reliance on virtual care in the wake of the COVID-19 pandem.c. Further research with rigorous m.thodology is needed t
