Rapid detection of fluoroquinolone residues in aquatic products based on a gold-labeled microwell immunochromatographic assay

Objectives:Fluoroquinolones(FQs)are widely used in aquaculture,and their residues have caused many problems threatening human health.Here,this study aims to develop a colloidal gold immunochromatographic strip based on gold-labeled microwells to screen the residues of FQs on site.Materials and methods:The Protein A Magarose Beads affinity chromatography method was adopted to purify the ascites against FQs.By using a strategy of heterologous coating antigen,different coating antigens are applied to detect FQs.The gold-labeled microwell immunochromatographic assay was used to improve the sensitivity of the test strip by the advanced reaction of antigen and antibody.Results:The antibodies were verified to be of high purity up to 99%,and the titer reached 1:1024000.The combination(enoxacin-OVA and the antibody)detected the 4 banned FQs(pefloxacin,PEF;norfloxacin,NOR;lomefloxacin,LOM;ofloxacin,OFL)with the 50%inhibiting concentration(IC50)values ranging from 1.3 to 2.1 ng/mL and cross-reactions ranging from 67.3%to 106.1%.The analysis of spiked crucian carp,silver carp,grass carp,and shrimp samples showed that the limit of detection for PEF,NOR,LOM,and OFL was 4μg/kg.A comparative study with liquid chromatography tandem mass spectrometry(LC-MS/MS)demonstrated that the assay provided an effective screening tool for the rapid detection of FQs residues.Conclusions:The results indicated that the test strip can realize full coverage recognition of the 4 banned FQs and has good accuracy,specifi-city,reproducibility,and stability;therefore,they are more suitable for rapid detection of FQs in aquatic products.

In this paper,we develop a novel global-attentionbased neural network(GANN)for vision language intelligence,specifically,image captioning(language description of a given image).As many previous works,the encoder-decoder framework is adopted in our proposed model,in which the encoder is responsible for encoding the region proposal features and extracting global caption feature based on a specially designed module of predicting the caption objects,and the decoder generates captions by taking the obtained global caption feature along with the encoded visual features as inputs for each attention head of the decoder layer.The global caption feature is introduced for the purpose of exploring the latent contributions of region proposals for image captioning,and further helping the decoder better focus on the most relevant proposals so as to extract more accurate visual feature in each time step of caption generation.Our GANN is implemented by incorporating the global caption feature into the attention weight calculation phase in the word predication process in each head of the decoder layer.In our experiments,we qualitatively analyzed the proposed model,and quantitatively evaluated several state-of-the-art schemes with GANN on the MS-COCO dataset.Experimental results demonstrate the effectiveness of the proposed global attention mechanism for image captioning.

Doxycycline hydrochloride and florfenicol combination(DoxHcl&FF)is an effective treatment for respiratory diseases.In the study,our objective Was to evaluate the activity of DoxHcl&FF against Actinobacillus pleuropneumoniae(APP)in porcine pulmonary epithelial lining fluid(PELF)and the optimal dosage scheme to avoid the development of resistance.The DoxHcl&FF Was administered intramuscularly(IM)at 20mg/kg,and the PELF was collected at differ-ent time points.The minimum inhibitory concentration(MIC)and time-mortality curves were also included in the study.Based on the sigmoid Emax equation and dose equations,the study integrated the in vivo pharmacokinetic data of infected pigs and ex vivo pharmacodynamic data to obtain the area under concentration time curve(AUCo-24h)MIC values in PELF and achieve bacteriostatic activity,bactericidal activity and the virtual eradication of bacteria.The study showed that the combination of DoxHcl and FF caused no significant changes in PK parameters.The peak concentration(Cmax)of FF in healthy and diseased pigs was 8.87±0.08 and 8.67±0.07μg/mL,the_AUCo-24h were.172.75±2.52 and 18022±3.13 h-μg/mL,the Cmax of DoxHcl was 7.91±0.09 and 7.99±0.05μg/mL,and the AUCo-24h was 129.96±3.70 h-μg/mL and 169.82±4.38 h-μg/mL.DoxHcl&FF showed strong concentra-tion-dependent tendencies.The bacteriostatic,bactericidal,and elimination activity were calculated as 5.61,18.83 and 32.68 h,and the doses were 1.37(bacteriostatic),4.59(bactericidal)and 7.99(elimination)mg/kg.These findings indicated that the calculated recommended dose could assist in achieving more precise administration,increasing the effectiveness of DoxHcl&FF treatment for APP infections.

Background The superiority of balloon angioplasty plus aggressive medical management(AMM)to AMM alone for symptomatic intracranial artery stenosis(sICAS)on efficacy and safety profiles still lacks evidence from randomised controlled trials(RCTs).Aim To demonstrate the design of an RCT on balloon angioplasty plus AMM for sICAS.Design Balloon Angioplasty for Symptomatic Intracranial Artery Stenosis(BASIS)trial is a multicentre,prospective,randomised,open-label,blinded end-point trial to investigate whether balloon angioplasty plus AMM could improve clinical outcome compared with AMM alone in patients with sICAS.Patients eligible in BASIS were 35–80 years old,with a recent transient ischaemic attack within the past 90 days or ischaemic stroke between 14 days and 90 days prior to enrolment due to severe atherosclerotic stenosis(70%–99%)of a major intracranial artery.The eligible patients were randomly assigned to receive balloon angioplasty plus AMM or AMM alone at a 1:1 ratio.Both groups will receive identical AMM,including standard dual antiplatelet therapy for 90 days followed by long-term single antiplatelet therapy,intensive risk factor management and life-style modification.All participants will be followed up for 3years.Study outcomes Stroke or death in the next 30 days after enrolment or after balloon angioplasty procedure of the qualifying lesion during follow-up,or any ischaemic stroke or revascularisation from the qualifying artery after 30 days but before 12 months of enrolment,is the primary outcome.Discussion BASIS trail is the first RCT to compare the efficacy and safety of balloon angioplasty plus AMM to AMM alone in sICAS patients,which may provide an alternative perspective for treating sICAS.Trial registration number NCT03703635;https://www.***.gov.Table 1 Continued Inclusion criteria BASIS,Balloon Angioplasty for Symptomatic Intracranial Artery Stenosis;Hb,haemoglobin;INR,international normalized ratio;sICAS,symptomatic intracranial artery stenosis.

In this paper,a cost-effective and miniaturized instrument is proposed,which is based on a tunable modulated grating Y-branch(MG-Y)laser for rapid temperature measurement using a Fabry-Perot interferometer(FPI)sensor.The FPI sensor with a 1463-μm cavity length is a short segment of a capillary tube sandwiched by two sections of single-mode fibers(SMFs).This system has a broad tunable range(1527 nm-1567 nm)with a wavelength interval of 8 pm and a tuning rate of 100 Hz.Temperature sensing experiments are carried out to investigate the performance of the system by demodulating the absolute cavity length of the FPI sensor using a cross-correlation algorithm.Experimental results show that the sensor can reach the response time as short as 94 ms with the sensitivity of 802 pm/C.Benefiting from the homemade and integrated essential electrical circuits,the entire system has the small size,low cost,and practical application potential to be used in the harsh environment for rapid temperature measurement.

Purpose As a fourth-generation light source,High Energy Photon Source(HEPS)has much more stringent requirements to the beam orbit stability in both horizontal and vertical directions than the previous sources due to the much smaller beam sizes.Methods A Fast Orbit FeedBack(FOFB)system,with the closed-loop bandwidth around 500 Hz,is needed to meet the critical requirements.The latency of the FOFB system is the key to achieve these requirements.Results Proper design and implementation of the feedback logic to have low latency is illustrated in the paper.Calculation accuracy is kept in the whole operation except for the last-minute data translation into 32-bit floating point number.

In the Qaidam basin,Northwest China,karstic fissures and caves are developed in many places of the Lower Carboniferous Chengqianggou Fm.and Huaitoutala Fm.and the Upper Carboniferous Keluke Fm.,in which are found typical karstic marks.Several suites of transgressive carbonate rocks formed during the Late Paleozoic epoch are preserved as reservoir properties in the Qaidam basin,with paleokarst-related cave-fractured diagenetic structures,which provided the basis for cavefractured development.Carbonate and clastic samples covering or infilling caves and fractures in Carboniferous strata were collected in the eastern Qaidam basin to focus on petrological and geochemical analyses of karst and infilling materials.Based on this study of the Caledonian diagenetic sequence,and bitumen-infilled inclusion temperature and burial history,there existed a continuous atmospheric freshwater leaching process,which played a critical role in the construction of the Carboniferous supergene paleokarsts.Investigation and survey of the tectonic setting,plate paleolatitude and river development intensity also proves that there was abundant rainfall during the Indosinian period,after which the strata undergo a shallow burial process.Because Indosinian period is earlier than the key timing of hydrocarbon-generation of the Carboniferous source rocks,this kind of karst reservoir has potential significance for oil-gas resources in this area.

Human noroviruses(HuNoVs)are major foodborne pathogens that cause nonbacterial acute gastroenteritis worldwide.As the tissue-culture system for HuNoVs is not mature enough for routine detection of the virus,detection is mainly dependent on molecular approaches such as reverse transcription polymerase chain reaction(RT-PCR)and reverse transcription quantitative real-time polymerase chain reaction(RTqPCR).The widely used primers and probes for RT-qPCR were established in the early 2000s.As HuNoVs are highly variant viruses,viral genome mutations result in previously designed primers and/or probes that were perfectly matched working less efficiently over time.In this study,a new duplex RT-qPCR(ND-RT-qPCR)was designed for the detection of genogroup Ⅰ(GⅠ)and genogroup Ⅱ(GⅡ)HuNoVs based on an analysis of viral sequences added in the database after 2010.Using long transcribed viral RNAs,the results demonstrate that the sensitivity of ND-RT-qPCR is as low as one genomic copy for both GⅠ and GⅡ HuNoVs.The performance of ND-RT-qPCR was further evaluated by a comparison with the commonly used Kageyama primer/probe sets for RT-qPCR(Kageyama RT-qPCR)for 23 HuNoV-positive clinical samples.All five GⅠ samples were registered as positive by ND-RT-qPCR,whereas only two samples were registered as positive by Kageyama RT-qPCR.All 18 GⅡ samples were registered as positive by ND-RT-qPCR,while 17 samples were registered as positive by Kageyama RT-qPCR.The sensitivity reflected by the quantification cycle(Cq)value was lower in ND-RT-qPCR than in Kageyama RT-qPCR.Our data suggest that ND-RT-qPCR could be a good fit for the detection of current strains of HuNoVs.

研究目的:分析“非粮化”现象的形成逻辑,确立法律规制理念,提出完善相应法律制度的路径。研究方法:法律文本分析、法律规范解释。研究结果:(1)我国当前“非粮化”现象体现出国家耕地保护政策目标与种植主体经营权之间的矛盾;(2)当前耕地保护立法与中央政策中防止“非粮化”的要求不适配,缺乏明确可操作的法律保障制度;(3)“非粮化”现象本质上是市场失灵的表现,可采用经济法中的国家干预理论予以治理,以实现市场系统与政府系统资源的有效配置。研究结论:防止“非粮化”的法律制度构建应当以国家干预理论为核心,以赋权、限权、激励的设计思路为导向,最终实现兼顾保障农民收益与国家粮食安全目标的效果。
