Objective To evaluate the feasibility and safety of peripheral CD34+ cell mobilization in patients with severe autoimmune disease. Methods Forty-two patients underwent a total of 46 mobilizations by the regimen of cyclophosphamide 2-3 g/m2 +recombinant human granulocyte colony stimulating factor (rhG-CSF) 5 μg·kg-1·d-1. The positive selection of CD34+ cell was performed through the CliniMACS. Results In 8.1±2.3 days after administration of cyclophosphamide, the peripheral white blood cell and mononuclear cell (MNC) decreased to the lowest level. In 3.7±1.6 days after injection of rhG-CSF, the peripheral absolute MNC and CD34+ cell counts were 0.95×109/L and 0.035×109/L, respectively. After 2.4±0.6 times of leukapheresis, there gained 4.46×108/kg of MNC and 5.26×106/kg of CD34+, respectively. After mobilization, the underlying diseases were ameliorated more or less. In systemic lupus erythematosus (SLE) patients, SLE Disease Activity Index (SLEDAI) decreased from a median of 17 to 3 (P<0.01). In rheumatic arthritis patients, an American College of Rheumatology criteria for 20%(ACR20) response was achieved in all five patients. Totally, 17.4% of patients whose absolute neutrophil count <0.5×109/L suffered infection, and 31.0% of patients had bone pain after the injection of rhG-CSF. Two patients suffered severe complications, one with acute renal failure and recovered by hemodialysis, the other died of thrombotic thrombocytopenic purpura. Failed mobilization occurred in three patients. Conclusions Sufficient CD34+ cells can be mobilized by low dose of cyclophosphamide and rhG-CSF. CD34+ cell mobilization for treatment of severe autoimmune disease not only is appropriate in both effectiveness and safety but ameliorates disease also.

To the Editor: We report a 27-year-old woman with a 2-month history of irregular uterine bleeding between periods. Blistering on the head was noted in the patient at the age of 2 months, and she gradually developed red maculopapular lesions over her neck and back during puberty, which were itchy after exposure to physical friction. On admission, physical examination had revealed diffuse faint maculopapular rash mainly on the neck and back [Figure 1]. Pelvic examination showed a smooth red lump (5 cm) on the cervix. No abnormality was detected in the complete blood count. Afterwards, the cervical lump was resected and confirmed to be the infiltration of mature mast cells (MCs) after careful pathological investigation. Additionally, a skin nodule biopsy also indicated mass aggregation of MCs, and a bone marrow (BM) biopsy showed a mass of round MCs with abundant cytoplasmic granules. The MCs accounted for about 22.5% of all cells in the aspirate smears. Flow cytometry immunophenotypic analysis on BM suggested that an aberrant MC population had represented 10% of the total analyzed cells, which was positive for CD117, CD33, and CD9, partially positive for CD2 and CD68, and negative for CD25. Genetic test through exome-wide sequencing for the receptor tyrosine kinase (KIT) gene had revealed K509I mutation.

Living/controlled cationic ring-opening polymerization （ROP） of L-lactide is a promising approach to isotactic-rich and crystalline poly（L-lactide）.In contrast with the unsubstituted lactones,L-LA can not be polymerized by organic Lewis acids or carbenium ions,and the state of the art in this field is the cationic ROP of L-LA catalyzed by Br?nsted acid/alcohol system via activated monomer ***,we reported the first example of controlled cationic ROP of L-LA by using Meerweintype ion pair[Me3O]+[B（C6F5）4]-as the catalyst.[Me3O]+[B（C6F5）4]-promoted rapid L-LA cationic polymerization in the absence of alcohol,producing isotactic-rich and crystalline PLLA without transesterification and epimerization side *** activated chain end mechanism,involving twice SN2 substitution and configuration-inversion （S→R→S） with the assistance of released Me2O,was proposed and further verified by density functional theory and control *** work expands the catalytic toolbox of isotactic-rich and crystalline polylactide *** represents a unique example of cationic-controlled polymerization of L-LA catalyzed by an organic ion pair.

To the Editor： Castleman＇s disease （CD） describes a group of lymphoproliferative disorders with heterogeneous clinical manifestations.[1] CD comprises at least two distinct diseases （unicentric CD [UCD] and multicentric CD [MCD]） with respective clinical features and prognoses： UCD often involves one lymph node area with favorable outcome： MCD is a systemic disease with relatively poor prognosis which involves multiple lymph node areas with constitutional symptoms （e.g. fever, night sweats, and weakness） and organ dysfunction. According to pathogenesis, MCD can be further divided into different subtypes based on the status of human herpes virus-8 （HHV-8） infection. Of the HHV-8 negative MCD （also named as idiopathic MCD [iMCD]）, there is a peculiar subtype named TAFRO syndrome which constitutes a constellation of unique clinical manifestations （T： Thrombocytopenia, A： Anasarca, F： myelofibrosis, R： Renal dysfunction, and O： Organomegaly）. This syndrome, first reported in Japan[2] has been reported mostly in the Japanese population and occasionally in Caucasians. There is no such case reported in China. We herein report the first patient with TAFRO syndrome in China with confirmed myelofibrosis.

Protein arginine methyltransferase-6 participates in a range of biological functions,particularly RNA processing,transcription,chromatin remodeling,and endosomal ***,it remains unclear whether protein arginine methyl transferase-6 modifies neuropathic pain and,if so,what the mechanisms of this *** this study,protein arginine methyltransferase-6 expression levels and its effect on neuropathic pain were investigated in the spared nerve injury model,chronic constriction injury model and bone cancer pain model,using immunohistochemistry,western blotting,immunoprecipitation,and label-free proteomic *** results showed that protein arginine methyltransferase-6 mostly co-localized withβ-tubulinⅢin the dorsal root ganglion,and that its expression decreased following spared nerve injury,chronic constriction injury and bone cancer *** addition,PRMT6 knockout(Prmt6~(-/-))mice exhibited pain ***,the development of spared nerve injury-induced hypersensitivity to mechanical pain was attenuated by blocking the decrease in protein arginine methyltransferase-6 ***,when protein arginine methyltransferase-6 expression was downregulated in the dorsal root ganglion in mice without spared nerve injury,increased levels of phosphorylated extracellular signal-regulated kinases were observed in the ipsilateral dorsal horn,and the response to mechanical stimuli was ***,protein arginine methyltransferase-6 appeared to contribute to spared nerve injury-induced neuropathic pain by regulating the expression of heterogeneous nuclear ***,protein arginine methyltransfe rase-6-mediated modulation of hete rogeneous nuclear ribonucleoprotein-F expression required amino atids 319 to 388,but not classical H3R2 *** findings indicated that protein arginine methyltransferase-6 is a potential therapeutic target fo r the treatment of peripheral neuro pathic pain.

This study presents a novel method to fabricate metal-decorated,sulfur-doped layered double hydroxides （M/SLDH） through spontaneous redox and sulfurization *** developed Ag/SLDH and Pt/SLDH catalysts with abundant heterogeneous interfaces and hierarchical nanostructures demonstrated outstanding oxygen evolution reaction （OER） and hydrogen evolution reaction （HER） performance,achieving low overpotentials of 212 and 35 mV at 10 mA cm-2in 1 M KOH,*** both anode and cathode in water splitting,they required only 1.47 V to reach 10 mA cm-2and exhibited high structural robustness,maintaining stability at 1000 mA cm-2for 300 ***-situ Raman analysis revealed that the synergistic effects of metal nanoparticles and S doping significantly promote the transformation into the S-Co1-xFexOOH layer,which serves as the active phase for water ***,ultraviolet photoelectron spectroscopy （UPS） and density functional theory （DFT） analyses indicated that incorporating metal nanoparticles and S doping increase electron density near the Fermi level and reduce reaction energy barriers,thus enhancing intrinsic OER and HER *** study provides a scalable strategy for synthesizing high-performance electrocatalysts for water splitting,with promising potential for broader applications.

Si-containing Mg alloys solidified at conventional rates often contain coarse and sharp Mg2Si phases,which can result in inferior material *** this study,Mg-6wt.%Si （Mg-6Si） alloy was prepared by wire arc additive manufacturing （WAAM）,employing the gas tungsten arc welding technique with rapid *** microstructures and mechanical properties of the WAAM alloy were investigated and compared with those of the as-cast samples produced using a metal *** results indicate that the WAAM Mg-6Si is harder and stronger than the as-cast *** microhardness of the WAAM Mg-6Si increases by 36.6%in comparison to that of as-cast Mg-6Si ***,the average tensile strengths at room temperature and 150°C increases by 63.4%and 21.3%,*** refines both the Mg2Si phase and the overall grains,resulting in a homogeneous morphology and improved mechanical *** granular Mg2Si phase,characterized by fine particles with a diffused distribution,shows a significant increase in *** acicular Mg2Si phase is distributed along the grain boundaries,and its concentration significantly *** average grain size of the Mg2Si phase is about 9.20μm,about 5 times *** refinement and distribution of the granular Mg2Si phase,as well as the reduction in the amount of needle-like Mg2Si particles,are the key factors for improving the mechanical properties of WAAM Mg-6Si alloy.

Background:Bendamustine was approved in China on May 26th,2019 by the National Medical Product Administration for the treatment of indolent B-cell non-Hodgkin lymphoma(NHL).The current study was the registration trial and the first reported evaluation of the efficacy,safety,and pharmacokinetics of bendamustine in Chinese adult patients with indolent B-cell NHL following relapse after chemotherapy and rituximab ***:This was a prospective,multicenter,open-label,single-arm,phase 3 study(NCT01596621;C18083/3076)with a 2-year follow-up *** patients received bendamustine hydrochloride 120 mg/m2 infused intravenously on days 1 and 2 of each 21-day treatment cycle for at least six planned cycles(and up to eight cycles).The primary endpoint was the overall response rate(ORR);and secondary endpoints were duration of response(DoR),progression-free survival(PFS),safety,and *** were classified according to their best overall response after initiation of *** of patients in each response category(complete response[CR],partial response[PR],stable disease,or progressive disease)were summarized along with a twosided binomial exact 95%confidence intervals(CIs)for the ***:A total of 102 patients were enrolled from 20 centers between August 6th,2012,and June 18th,*** the time of the primary analysis,the ORR was 73%(95%CI:63%–81%)per Independent Review Committee(IRC)including 19%CR and 54%*** the follow-up period,the median DoR was 16.2 months by IRC and 13.4 months by investigator assessment;the median PFS was 18.6 months and 15.3 months,*** most common non-hematologic adverse events(AEs)were gastrointestinal toxicity,pyrexia,and *** 3/4 neutropenia was reported in 76%of *** AEs were reported in 29 patients and five patients died during the *** analysis indicated that the characteristics of bendamustine and its metabolites M3 and M4 were generally consistent

An ever-increasing market demand has stimulated the soaring enthusiasm of researchers to develop wide-temperature supercapacitors(SCs). The active electrode is one of the most important parts of SC, which is directly related to the energy density,power transmission and long-term cyclability of the device in the wide-temperature environment. Compared with the SC electrodes aimed for room-temperature application, the SC electrodes for operating at wide-temperature scene often face greater challenges. In this review, the main challenges of SC electrodes under various temperature conditions, including low, high and cross-fade temperatures, are summarized. The relevant performance decay and failure mechanisms of wide-temperature SC electrodes are analyzed. In addition, this review deals with the recent studies and developments in robust wide-temperature SC electrodes with respect to the rational design of electrode structures and the exploitation of advanced active materials. Finally, the future directions for exploring reliable wide-temperature SCs are also proposed.
